The intronless mouse gene for the tissue specific splicing protein SmN is a processed pseudogene containing a stop codon after thirty-one amino acids.

The SmN protein is a component of small ribonucleoprotein particles which is closely related to the ubiquitously expressed splicing proteins SmB and B' but is expressed in only a small number of cells and tissues. We have isolated a mouse SmN-related sequence which lacks introns and contains multiple changes from the SmN cDNA sequence including a stop codon after thirty-one amino acids which would prevent it encoding functional SmN protein. This indicates that this intronless gene is a processed pseudogene and that the functional gene has yet to be isolated. In agreement with this southern blotting of mouse DNA with SmN probes reveals bands, additional to those derived from the pseudogene, which are characteristic of an intron-containing SmN gene. The relationship of the pseudogene to the functional SmN gene and to an intronless SmN-related sequence in the rat genome is discussed.     

METHODS FOR COMPUTING LYMPHOCYTE KINETICS IN MAN BY ANALYSIS OF CHROMOSOMAL ABERRATIONS SUSTAINED DURING EXTRACORPOREAL IRRADIATION OF THE BLOOD

Data on lymphocyte kinetics may be derived in patients undergoing treatment by extracorporeal irradiation of the blood by studying chromosome aberrations induced in their lymphocytes; the aberrations are observed when the lymphocytes are stimulated by phytohaemagglutinin to divide in culture. Two methods of analysis are described, one depends on the total number of dicentrics scored, the other employs the dilution principle, regarding each lymphocyte bearing one or more aberrations as specifically labelled. Two parameters may be calculated: the mean residence time of lymphocytes in the blood and the size of the extravascular pool of lymphocytes in the body. In a patient with Hodgkin's disease, the mean residence time was found to be 4.7–7.5 min and the extravascular pool 800–1070 g.     

Separation of bovine X and Y sperm based on surface differences

Aqueous two-phase partition involving thin-layer counter current distribution (TLCCD) has been used to assess surface heterogeneity of ejaculated bovine sperm. When partitioned in charge-insensitive aqueous two-phase systems, which detect non-charge associated surface properties, the sperm fractionates into two distinct populations. Using a Y-chromosome-specific DNA marker, it has been shown that one of these populations is enriched in Y chromosome bearing sperm. However, this population is not pure—it consists of 80% Y sperm, with the other 20% being X sperm. All the sperm in the original population that had begun to undergo the acrosome reaction were separated into this same peak; the sex chromosome composition of these sperm is unknown. Since the aqueous partition of sperm is based on surface properties these results suggest that two populations of Y sperm exist that have different surface characteristics. © 1993 Wiley-Liss, Inc.     

Propagation of Bacillus popilliae in laboratory fermentors

The insect pathogen Bacillus popilliae Dutky causes a fatal milky disease of Japanese beetle larvae. Spores of the bacterium offer a biological means of controlling this insect. While satisfactory sporulation in vitro has not yet been accomplished, conditions have been developed for the proliferation of vegetative cells in shaken flasks and aerated fermentors. Vegetative cultures are maintained by frequent transfer or by lyophilization. Media based on yeast extract are used routinely, but corn steep liquor and casein hydrolyzates afford comparable yields of 5 × 10⁸ cells/ml. in 16–24 hr. Nutritional requirements have been established for growth in a synthetic medium. Oxygen availability affects the pathway of carbohydrate catabolism and is necessary for optimal growth. In rapidly growing cultures, a short period of maximum viability is characteristically followed by rapid death of the cells. When inoculum size and transfer time are suitably manipulated, viable cell yields reach 1–2 × 10⁹/ml. Alternative methods of propagation, including the addition of particulate carbon, and procedures designed either to neutralize acids or to remove metabolic products by dialysis, do not markedly enhance the yield of cells per volume of medium, although viability may be prolonged.     

Stimulation of Leydig cell function by a polypeptide present in testicular interstitial fluid

Rat testicular interstitial fluid (IF) contains a polypeptide factor(s) which when added to Percoll-purified rat Leydig cells in vitro, enhances both basal and hCG-stimulated production of testosterone in a dose-dependent manner. To assess the possible physiological significance of this factor(s), its levels in testicular IF from individual rats were assessed following the suppression of intratesticular levels of testosterone by either (a) treatment with anti-LH, (b) induction of unilateral cryptorchidism, or (c) treatment with ethane-1,2-dimethane sulphonate (EDS). These treatments suppressed testosterone levels in IF by 80 to 99% and in each instance levels of the IF-factor were increased. This increase occurred as early as 5h after anti-LH treatment, but restoration of normal intratesticular levels of testosterone at 20-40 h after anti-LH treatment by testosterone injection failed to normalize levels of the IF-factor. In contrast, injection of LH into normal rats raised IF testosterone to supranormal levels and this was associated with a reduction in levels of the IF-factor, and similar effects of hCG were observed in rats pretreated with anti-LH. Therefore, whilst these studies suggest that the IF-factor(s) may be of physiological importance, its local regulation does not appear to involve a straightforward negative feedback control by testosterone.     

Separation of Spores and Parasporal Crystals of Bacillus thuringiensis in Gradients of Certain X-Ray Contrasting Agents

Spores and parasporal crystals of Bacillus thuringiensis can be separated at moderate centrifugation speeds (10,000 to 12,000 rpm) in gradients of Renografin or sodium diatrizoate.
