Two hundred and thirty-three thyroid lesions were studied by fine needle aspiration (FNA) cytology using standard cytologic criteria available in the literature. These included 114 cases of nodular colloid goitre (NCG), 47 cases of Hashimoto's thyroiditis (HT), 12 follicular adenomas (FAd), five cases of subacute thyroiditis and three cases of thyrotoxicosis among the benign lesions. The malignant lesions seen were 30 cases of papillary carcinoma (PCa), 16 follicular carcinomas (FCa), three cases with double lesions, e.g. papillary carcinoma with coexisting NCG, and three of papillary carcinoma with HT. Emphasis was given to eight qualitative and quantitative (morphometric) variables in these various thyroid lesions. Cell measurements were done using a Visopan Lux projection microscope. The three qualitative variables included type of nuclear membrane (regular/irregular), type of nuclear chromatin and the presence or absence of conspicuous nucleoli. The quantitative variables studied were nuclear diameter, nuclear area, cytoplasmic diameter, cytoplasmic area & N/C ratio. Statistical analysis was performed in order to know whether the standard cytologic criteria used at FNA cytology in the literature (increased cellularity, microfollicles, increased N/C ratio, absence of significant haemosiderin-laden macrophages and scanty colloid) could differentiate a follicular adenoma from a follicular carcinoma. A statistical analysis was also performed to establish the utility of the qualitative and quantitative variables. The results showed that none of the standard cytologic criteria applied could differentiate follicular adenoma from a follicular carcinoma. With regard to qualitative variables, irregularity of nuclear membrane and presence of conspicuous nucleoli were most significant in papillary carcinoma, followed by follicular carcinoma, then by follicular adenoma; these features being hardly evident in nodular colloid goitre and Hashimoto's thyroiditis. A coarse nuclear chromatin was most significant in follicular carcinomas followed by follicular adenomas. It was less obvious in the benign conditions, but more prominent in Hashimoto's thyroiditis compared with a goitre. It was also not obvious in a papillary carcinoma. Of the quantitative variables, all measurements were greatest in PCa > FCa > FAd > NCG = HT. When differentiating follicular adenoma from follicular carcinoma the qualitative variables of significance were the presence or absence of nucleoli, the chromatin pattern and regularity/irregularity of nuclear membrane; the nuclear area was the most important feature among quantitative variables.  相似文献

A census of clean room surface-associated bacterial populations was derived from the results of both the cloning and sequencing of 16S rRNA genes and DNA microarray (PhyloChip) analyses. Samples from the Lockheed Martin Aeronautics Multiple Testing Facility (LMA-MTF), the Kennedy Space Center Payload Hazard and Servicing Facility (KSC-PHSF), and the Jet Propulsion Laboratory Spacecraft Assembly Facility (JPL-SAF) clean rooms were collected during the various assembly phases of the Phoenix and Mars Science Laboratory (MSL) spacecraft. Clone library-derived analyses detected a larger bacterial diversity prior to the arrival of spacecraft hardware in these clean room facilities. PhyloChip results were in agreement with this trend but also unveiled the presence of anywhere from 9- to 70-fold more bacterial taxa than cloning approaches. Among the facilities sampled, the JPL-SAF (MSL mission) housed a significantly less diverse bacterial population than either the LMA-MTF or KSC-PHSF (Phoenix mission). Bacterial taxa known to thrive in arid conditions were frequently detected in MSL-associated JPL-SAF samples, whereas proteobacterial lineages dominated Phoenix-associated KSC-PHSF samples. Comprehensive bacterial censuses, such as that reported here, will help space-faring nations preemptively identify contaminant biomatter that may compromise extraterrestrial life detection experiments. The robust nature and high sensitivity of DNA microarray technologies should prove beneficial to a wide range of scientific, electronic, homeland security, medical, and pharmaceutical applications and to any other ventures with a vested interest in monitoring and controlling contamination in exceptionally clean environments.Planetary protection efforts work toward protecting (i) solar system bodies from contamination by terrestrial biological material (forward contamination), thus preserving opportunities for future scientific investigation, and (ii) the Earth from harmful contamination by materials returned from outer space (back contamination) (5). These approaches apply directly to the control and eradication of microorganisms present on the surfaces of spacecraft intended to land, orbit, fly by, or be in the vicinity of extraterrestrial bodies. Consequently, current planetary protection policies require that spacecraft be assembled and readied for launch in controlled clean room environments. To achieve these conditions and maintain compliance with good manufacturing practice regulations, robotic spacecraft components are assembled in ultraclean facilities. Much like facilities in the medical, pharmaceutical, and semiconductor sectors, National Aeronautics and Space Administration (NASA) spacecraft assembly clean rooms (SAC) are kept extremely clean and are maintained to the highest of industry standards (17). Filtered air circulation, controlled temperature and humidity, routine exposure to disinfectants and surfactants, and nutrient-limiting, oligotrophic conditions make it very challenging for microorganisms to persist in such environments, but these measures by no means eradicate biological contaminants entirely (18). Several investigations, both culture based and culture independent, have demonstrated that a variety of bacterial taxa are repeatedly isolated under clean room conditions (18, 24, 26; P. Vaishampayan, S. Osman, G. Andersen, and K. Venkateswaran, submitted for publication). However, despite a growing understanding of the diverse microbial populations present in SAC, predicting the true risk of any such microbes’ compromising the findings of extraterrestrial life detection efforts remains a significant challenge (30). A better understanding of the distribution and frequency at which high-risk contaminant microbes are encountered on spacecraft surfaces would significantly aid in assessing the threat of forward contamination (33).The purification of nucleic acids, subsequent PCR amplification, and shuttling of 16S ribosomal “fingerprint” genes from noncultivable microorganisms into genetically amenable lab strains of Escherichia coli have evolved into a gold standard of molecular means to elucidate the microbial diversity in a given sample. In theory, the cloning and sequencing of 16S ribosomal genes from each and every cell present, regardless of cultivability and inclusive of novel taxa, would result in a comprehensive survey of microbial communities on the surfaces of SAC and colocated spacecraft (24, 26). Unfortunately, the full-length sequencing of all 16S rRNA genes from environmental samples would be prohibitively expensive, making the approach unfeasible for generating comprehensive phylogenetic profiles of complex microbial communities.Attempting to infer population membership from clone libraries limited to hundreds or thousands of sequences has proven to be insufficient for detecting extremely low-abundance organisms. Recent analyses of phylogenetic DNA extracted from soil, water, and air revealed that laboriously derived clone libraries severely under-represent complex bacterial communities compared to very rapid (i.e., requiring only hours) DNA microarray approaches (1, 6, 11, 23, 36). One of the reasons for this is the high sensitivity of PhyloChip methodologies, which are able to detect organisms present in amounts below 10−4 abundance of the total sample (12). Numerous validation experiments using sequence-specific PCR have confirmed that taxa identified by the microarray were indeed present in the original environmental samples, despite their absence in corresponding clone libraries (3). This highlights the utility of the method compared to classical cloning. Although the analysis of each sample by the PhyloChip provides detailed information on microbial composition, the highly parallel and reproducible nature of this array allows tracking community dynamics over time and treatment. Even without prior sequence information, PhyloChip can identify specific microbial interactions that are key to particular changing environments.A comprehensive census of the microbial communities on the surfaces in three NASA SAC supporting two distinct missions was conducted. To ensure that the maximum diversity of resident microbiota was uncovered, subsamples from each clean room surface sampling were subjected to both DNA microarray protocols and conventional cloning and sequencing of 16S rRNA genes. This study, to our knowledge the first of its kind, focused on comparing the microbial diversity profiles resulting from DNA microarray analyses and conventional cloning and sequencing of 16S rRNA genes arising from a variety of low-biomass surfaces.  相似文献

Barley straw, an agricultural waste, was chemically modified and evaluated for the removal of emulsified oils from aqueous solution. The chemical modification was performed using NaOH and a cationic surfactant, hexadecylpyridinium chloride monohydrate (CPC). The surface textural and chemical properties of the surfactant modified barley straw (BMBS) were characterized by N2 adsorption, FT-IR, SEM and water soluble mineral content. The adsorption tests were carried out in batch adsorption system for removal of standard mineral oil (SMO) and canola oil (CO) from water. For both emulsified oils in wastewater, adsorption was found to be strongly related with solution pH. The isotherm study indicated that emulsified oil adsorption on BMBS could be fitted well with the Langmuir model other than Freundlich model. The maximum adsorption capacity for CO and SMO at 25 °C determined from the Langmuir isotherm is 613.3 and 584.2 mg g−1, respectively. Desorption tests in water solution show that oil is strongly bonded with adsorbent and desorption is only about 1–2% in 24 h.  相似文献

Objective

In Malaysia, futsal is a popular sport played by individuals across all ages and genders. Despite its popularity, information on futsal related injury in Malaysia is not available. The purpose of this study is to examine the injury incidence and injury patterns among amateur men and women futsal players in Malaysia.

Methods

Players reported injury to the tournament medical team during the FELDA/FAM National Futsal League 2010 were interviewed and assessed by a Sports Medicine registrar. Player''s socio-demographic profiles and information about the injury were documented in the injury report form adapted from medical report form used by FIFA: Medical Assessment and Research Centre (F-Marc).

Results

A total of 86 injuries were reported from 141 matches, equivalent to an incidence of 91.5 injuries per 1000 player hours (95% CI 72.2 to 110.8), or 61.0 injuries per 1000 player matches (95% CI 48.1 to 73.9). Most were minor injuries resulted from contact with another player. Injuries often involved the lower extremity (44%) followed by the trunk (14%) and the upper limb (13%). Ankle (n = 7; 39%) and knee (n = 6; 33%) sprains were the most prevalent diagnoses of time-loss injuries. A significant association between time-loss and type of injury was found χ 2 (1,N = 86) = 3.99, p = 0.04. In addition, time-loss injury was significantly associated with playing surface χ 2 (1,N = 86) = 10.11, p = 0.018.

Conclusion

The injury rate during the FELDA/FAM National Amateur Futsal Men''s League in Malaysia was lower compared with previous Futsal World Cups competition. Most injuries resulted from contact with another player were minor and did not lead to time-loss from participation. Time-loss injury was significantly associated with type of injury and playing surface.  相似文献

In the state Meghalaya, northeast India, >80% of the forest lands are owned by local communities and managed by traditional institutions. These forests are under severe threats due to a number of human disturbances. The present study was conducted to assess the plant diversity and identify the community forests for priority conservation in Khasi Hills of Meghalaya. Floristic explorations carried out in the 87 forests reveals the presence of 1300 plant species of which 400 are either rare, endemic or threatened. Of the different forest categories, reserve forests had the highest number of species (1190), followed by sacred forests (987 species) and village forests (786 species). Majority of the forests (56) had high-species richness, irreplaceability level (42 forests) and vulnerability level (54). In terms of area, 13.8% (1666.8 ha) fall under low risk while 1855 ha under high risk zone. High risk zone was mostly represented by village forests. An area of 7661.56 ha of community forests falls under high priority category and hence calls for immediate conservation actions. The conservation priority map generated in the present study will help to concentrate the protection strategy to the demarcated and adjoining areas and help conservationists and planners to evolve effective strategies for conservation of the community forests.  相似文献

Excess sugar consumption has been shown to contribute directly to weight gain, thus contributing to the growing worldwide obesity epidemic. Interestingly, increased sugar consumption has been shown to repeatedly elevate dopamine levels in the nucleus accumbens (NAc), in the mesolimbic reward pathway of the brain similar to many drugs of abuse. We report that varenicline, an FDA-approved nicotinic acetylcholine receptor (nAChR) partial agonist that modulates dopamine in the mesolimbic reward pathway of the brain, significantly reduces sucrose consumption, especially in a long-term consumption paradigm. Similar results were observed with other nAChR drugs, namely mecamylamine and cytisine. Furthermore, we show that long-term sucrose consumption increases α4β2 * and decreases α6β2* nAChRs in the nucleus accumbens, a key brain region associated with reward. Taken together, our results suggest that nAChR drugs such as varenicline may represent a novel treatment strategy for reducing sugar consumption.  相似文献

The binding of [14,15-3H]14,15-dihydroforskolin ([3H]DHF) to rat liver membranes has been further characterized and was compared with the stimulatory effect of forskolin on adenylate cyclase. The binding equilibrium dissociation constant (KD) for 14,15-dihydroforskolin obtained in inhibition experiments was 0.6 microM, with a maximal binding capacity (Bmax) of 114 pmol/mg protein. A similar KD value (0.5 microM) was derived from kinetics studies that revealed very rapid association and dissociation reactions. For structure-activity relationship studies several forskolin derivatives were synthesized and tested for their ability to inhibit [3H]DHF binding and increase adenylate cyclase activity. Among the tested compounds, forskolin itself was the most potent agonist (K1 = 0.2 microM). Further modification of the molecule in position 7 and (or) 1 decreased or abolished its agonist properties in both adenylate cyclase and binding studies. [3H]DHF binding was not affected by several nucleotides, carbohydrates, lectins, and hormone receptor agonists including isoproterenol, glucagon, and adenosine, but the steroids 17-beta-estradiol, progesterone, and testosterone showed slight inhibitory effects at unphysiologically high concentrations. [3H]DHF binding and forskolin-stimulated adenylate cyclase were sensitive to heat and N-ethylmaleimide treatment. Forskolin protected adenylate cyclase against inactivation by heat but not by N-ethylmaleimide. Preincubation of the membrane with trypsin decreased [3H]DHF binding. The results presented in this study demonstrate that the binding sites identified with [3H]DHF have a high specificity for forskolin and provide evidence that these binding sites are involved in the stimulation of adenylate cyclase by forskolin.  相似文献

Diacylglycerol acyltransferase (DGAT) catalyzes the final step in triglyceride (TG) synthesis. There are two isoforms, DGAT1 and DGAT2, with distinct protein sequences and potentially different physiological functions. To date, the ability to determine clear functional differences between DGAT1 and DGAT2, especially with respect to hepatic TG synthesis, has been elusive. To dissect the roles of these two key enzymes, we pretreated HepG2 hepatoma cells with (13)C(3)-D(5)-glycerol or (13)C(18)-oleic acid, and profiled the major isotope-labeled TG species by liquid chromatography tandem mass spectrometry. Selective DGAT1 and DGAT2 inhibitors demonstrated that (13)C(3)-D(5)-glycerol-incorporated TG synthesis was mediated by DGAT2, not DGAT1. Conversely, (13)C(18)-oleoyl-incorporated TG synthesis was predominantly mediated by DGAT1. To trace hepatic TG synthesis and VLDL triglyceride (VLDL-TG) secretion in vivo, we administered D(5)-glycerol to mice and measured plasma levels of D(5)-glycerol-incorporated TG. Treatment with an antisense oligonucleotide (ASO) to DGAT2 led to a significant reduction in D(5)-glycerol incorporation into VLDL-TG. In contrast, the DGAT2 ASO had no effect on the incorporation of exogenously administered (13)C(18)-oleic acid into VLDL-TG. Thus, our results indicate that DGAT1 and DGAT2 mediate distinct hepatic functions: DGAT2 is primarily responsible for incorporating endogenously synthesized FAs into TG, whereas DGAT1 plays a greater role in esterifying exogenous FAs to glycerol.  相似文献

MAPK-activated protein kinase-2 (MAPKAPK2) regulates the synthesis of tumor necrosis factor and other cytokines and is a potential drug target for inflammatory diseases. Five protein constructs were produced in 4-10mg quantities per liter of culture media using baculovirus-infected insect cells and characterized for kinase activity, thermal stability, and ligand-binding affinity. Compared to construct 1-370, removal of the C-terminal autoinhibitory peptide in 1-338 resulted in a destabilized but partially active nonphosphorylated enzyme; phosphorylation of 1-338 by p38alpha further increased activity 12-fold. A putative constitutively active mutant, 1-370/T222E/T334E, was 6.3-fold less active than phosphorylated 1-370. ThermoFluor, an equilibrium ligand-binding assay, was used to measure nucleotide analogue affinity for various constructs. Binding of phosphorylated nucleotides was Mg(2+)-dependent. Residues 1-40 were required for high-affinity binding of ADP, ATPgammaS, staurosporine, and K252a. A mutation M138A rendered 1-370 susceptible to p38-inhibitors SB-203580 and SB-202190 with IC50 values of 17.4 and 14.1 microM, respectively. Taken together, these studies provide information on the mechanism of ligand-binding to MAPKAPK2 that can be used in the search for selective small-molecule inhibitors.  相似文献
