We have assayed absorbance changes generated by blue light in plasma membranes, endoplasmic reticulum, and mitochondrial membranes from Neurospora crassa. Light minus dark difference spectra, obtained anaerobically in the presence of ethylenediaminetetraacetate, indicated that b-type cytochromes could be photoreduced in all three membranes. In plasma membranes, a b-type cytochrome with a distinct difference spectrum was photoreducible without addition of exogenous flavin. Addition of riboflavin greatly stimulated the photoreduction of cytochromes in endoplasmic reticulum and mitochondrial membranes. In its spectral characteristics the cytochrome on the endoplasmic reticulum resembled cytochrome b5 or nitrate reductase, while the cytochrome in mitochondrial membranes had the same spectrum as cytochrome b of the mitochondrial respiratory chain.
Cytochromes in the three membrane fractions reacted differently to blue light in the presence of various inhibitors. Potassium azide inhibited reduction of plasma membrane cytochrome b, with 50% inhibition at 1.0 millimolar. The same concentration of azide stimulated photoreduction of cytochromes in both endoplasmic reticulum and mitochondria. Although photoreduction of cytochromes in all three membranes was inhibited by salicylhydroxamic acid, cytochromes in plasma membranes were more sensitive to this inhibitor than those in endoplasmic reticulum and mitochondria. Cells grown to induce nitrate reductase activity showed an elevated amount of blue light-reducible cytochrome b in the endoplasmic reticulum.
Increasing worldwide resistance to acaricides necessitates greater research on the identification of potential acaricide targets
in ticks to aid in the control of these serious pests of medical and veterinary importance. Historically, and most likely
in the future, acaricide targets are largely of neural origin, but our knowledge of tick neurobiology is surprisingly limited.
The tick central nervous system is a fused nerve mass, termed the synganglion. Tick synganglion material is relatively easily
accessible to most researchers and employing modern amplification methods of complementary-DNA construction is readily amenable
for gene cloning investigations. The various tick neurotransmitter systems are described with emphasis on our current knowledge
of both existing and potential acaricide targets at the molecular level. We describe the impact of mass gene sequencing (expressed
sequence tag and genome projects), advances in bioinformatics and RNA-interference on target identification and validation. 相似文献
This paper presents a statistical analysis of the pooled data from a number of previous experiments concerning the dependence of the Young's moduli and strength of cancellous bone tissue upon apparent density. The results show that both the Young's moduli and the strength are proportional to the square of apparent density of the tissue and are therefore proportional to one another. It is shown that the coefficient of proportionality is different for human and bovine tissue. It is concluded that the suggestion of Wolff (Das Gesetz der Transformation der Knochen, Hirschwald, Berlin, 1892) that compact bone tissue is simply more dense cancellous bone tissue is not an accurate statement when only the mechanical properties of these two tissues are considered. It is noted that estimates for the elastic modulus of the individual trabecula of human cancellous bone vary from 1 to 20 GPa and it is suggested that this question needs further study. 相似文献
In order to understand and moderate the effects of the accelerating rate of global environmental change land managers and ecologists must not only think beyond their local environment but also put their problems into a historical context. It is intuitively obvious that historians should be natural allies of ecologists and land managers as they struggle to maintain biodiversity and landscape health. Indeed, ‘environmental history’ is an emerging field where the previously disparate intellectual traditions of ecology and history intersect to create a new and fundamentally interdisciplinary field of inquiry. Environmental history is rapidly becoming an important field displacing many older environmentally focused academic disciplines as well as capturing the public imagination. By drawing on Australian experience I explore the role of ‘environmental history’ in managing biodiversity. First I consider some of the similarities and differences of the ecological and historical approaches to the history of the environment. Then I review two central questions in Australian environment history: landscape‐scale changes in woody vegetation cover since European settlement and the extinction of the marsupials in both historical and pre‐historical time. These case studies demonstrate that environmental historians can reach conflicting interpretations despite using essentially the same data. The popular success of some environmental histories hinges on the fact that they narrate a compelling story concerning human relationships and human value judgements about landscape change. Ecologists must learn to harness the power of environmental history narratives to bolster land management practices designed to conserve biological heritage. They can do this by using various currently popular environmental histories as a point of departure for future research, for instance by testing the veracity of competing interpretations of landscape‐scale change in woody vegetation cover. They also need to learn how to write parables that communicate their research findings to land managers and the general public. However, no matter how sociologically or psychologically satisfying a particular environmental historical narrative might be, it must be willing to be superseded with new stories that incorporate the latest research discoveries and that reflects changing social values of nature. It is contrary to a rational and publicly acceptable approach to land management to read a particular story as revealing the absolute truth. 相似文献
The enzyme 2-C-methyl-D-erythritol 2,4-cyclodiphosphate (MECDP) synthase catalyzes the conversion of 4-diphosphocytidyl-2-C-methyl-D-erythritol 2-phosphate (CDP-ME2P) to MECDP, a highly unusual cyclodiphosphate-containing intermediate on the mevalonate-independent pathway to isopentenyl diphosphate and dimethylallyl diphosphate. We now report two x-ray crystal structures of MECDP synthase refined to 2.8-A resolution. The first structure contains a bound Mn(2+) cation, and the second structure contains CMP, MECDP, and Mn(2+). The protein adopts a homotrimeric quaternary structure built around a central hydrophobic cavity and three externally facing active sites. Each of these active sites is located between two adjacent monomers. A tetrahedrally arranged transition metal binding site, potentially occupied by Mn(2+), sits at the base of the active site cleft. A phosphate oxygen of MECDP and the side chains of Asp(8), His(10), and His(42) occupy the metal ion coordination sphere. These structures reveal for the first time the structural determinants underlying substrate, product, and Mn(2+) recognition and the likely catalytic mechanism accompanying the biosynthesis of the cyclodiphosphate-containing isoprenoid precursor, MECDP. 相似文献
The sequence content of mouse L cell pre-rRNA was examined by RNA gel transfer and blot hybridization. Nuclear RNAs were separated by agarose gel electrophoresis, transferred to diazo-paper, and hybridized to twelve different restriction fragments that are complementary to various sections of 45S pre-rRNA. An abundant new 34S pre-rRNA and less abundant new 37S, 26S and 17S pre-rRNAs were detected. The presence of these new pre-rRNAs suggests the existence of at least two new pre-rRNA cleavage pathways. 34S and 26S pre-rRNAs were also detected in HeLa cells suggesting that these new cleavage pathways are characteristic of mammalian cells. Further, an abundant new 12S precursor to 5.8S rRNA was also detected and is common to all the proposed cleavage pathways. The previously identified 45S, 41S, 32S and 20S pre-rRNAs were readily detected and their general structure confirmed. The 20S pre-rRNA is characteristic of the known pathway used by HeLa and other cells, and its presence suggests that growing mouse L cells use this pre-rRNA cleavage pathway. The 36S pre-rRNA characteristic of the previously described mouse L cell cleavage pathway was not detected. In all these cleavage pathways pre-rRNA cleavage sites are apparently identical and occur at or near the termini of the mature 18S, 5.8S and 28S rRNA sequences. Alpine Trifolium species have high rates of symbiotic N2-fixation which may influence the abundance and growth of plant species growing near them. The potential for facilitative
effects on plant abundance and growth in dry meadow alpine tundra of Niwot Ridge, Colo., characterized by low resource availability,
was investigated by measuring soil N, aboveground biomass production, and plant species composition in patches of Trifolium dasyphyllum and surrounding tundra. Extractable inorganic N was more than twofold greater and extractable P was 27% lower in Trifolium patches than in surrounding tundra. Aboveground production was twofold greater in Trifolium patches than in surrounding tundra. However, the difference was largely due to the production of T. dasyphyllum relative to the non-Trifolium component of biomass, which was not different between the Trifolium patches and surrounding tundra. In the Trifolium patches, the proportion of graminoid biomass was lower while the proportion of forb biomass was higher relative to surrounding
tundra. Although the abundance of some species was positively associated with the presence of Trifolium, other species were less abundant, possibly due to increased competition for P and differential abilities of alpine species
to respond to increased N availability. Trifolium may exert both facilitative and inhibitive effects on dry meadow alpine species and, in the process, substantially influence
the spatial heterogeneity in community structure and primary production.

Summary Zwitterion buffers are often used to modulate the pH of cell culture medium but their effect on cultured cells is controversial.
We found that addition of 4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid (HEPES) caused superoxide dismutase (SOD) inhibitable
increases in nitroblue tetrazolium dye reduction and SOD and catalase inhibitable decreases in the growth of cultured bovine
pulmonary artery endothelial cells. The findings suggest that HEPES stimulates endothelial cells to make toxic oxygen metabolites
that contribute to decreased cell growth.
