Soybean seeds contain three lipoxygenase (Lox) enzymes that are controlled by separate genes, Lox1, Lox2 and Lox3. Lipoxygenases play a role in the development of unpleasant flavors in foods containing soybean by oxidation of polyunsaturated fatty acids. Null alleles for all three enzymes have been identified, lox1, lox2 and lox3, and are known to be inherited as simple recessive alleles. Previous studies determined that a missense mutation rendered Lox2 inactive; however, the genetic cause of either lox1 or lox3 mutation was not known. The objectives of this study were the molecular characterization of both lox1 and lox3 mutant alleles and the development of molecular markers to accelerate breeding for Lox-free soybean varieties. We identified two independent mutant alleles as the genetic causes of the lack of Lox1 in seeds of two lox1 mutant soybean lines. Similarly, a mutant allele that truncates Lox3 in a lox3 mutant soybean line was identified. Molecular markers were designed and confirmed to distinguish mutant, wild type, and heterozygous individuals for Lox1, Lox2 and Lox3 genes. Genotype and Lox phenotype analysis showed a perfect association between the inheritance of homozygous lox mutant alleles and the lack of Lox activity. Molecular characterization of a seed-lipoxygenase-free soybean line led to the discovery that an induced recombination event within the Lox1 gene was responsible for breaking the tight linkage in repulsion phase between mutant alleles at the Lox1 and Lox2 loci. The molecular resources developed in this work should accelerate the inclusion of the lipoxygenase-free trait in soybean varieties.  相似文献

The relative decomposability of corn (Zea mays L.) residues from insect (Bt)-protected hybrids and conventional hybrids cultivated under insect pressure was investigated in two studies. Above-ground biomass, residue macromolecular composition, and stalk physical strength were also measured. In the first decomposition study, chopped residues (stalks and leaves) were used from a corn rootworm-protected (Cry3Bb1) hybrid and its non-Bt near isoline that were grown in replicated plots infested with corn rootworms (Diabrotica spp.). In the second study, residue (intact stalk sections) was used from three European corn borer (ECB, Ostrinia nubilalis Hübner)-resistant (Cry1Ab) hybrids representing different seed manufacturer/maturity date series, their non-Bt near isolines, two Cry3Bb1-protected isolines, and three additional conventional hybrids, all cultivated in replicated plots under conditions of elevated ECB pressure. In both studies, insect-resistant residues decomposed at rates similar to their non-protected near isolines. No evidence was found that insect-protected hybrids produced more above-ground biomass or had distinct residue composition. While some measures of mechanical stalk strength indicated that ECB-damaged stalks were not as stiff as protected stalks, these physical differences did not translate into differences in residue decomposition. We conclude that while individual hybrids may vary in their production of biomass, residue composition or residue decomposability, these characteristics do not systematically vary with the presence of the Bt gene conferring insect resistance, even under conditions of insect pressure.  相似文献

Predators exert strong top-down pressure on herbivorous insects, but research on how predators affect herbivore fitness often focuses on the more active juvenile and adult life stages while ignoring the pupal or cocoon life stage. Few studies have investigated predation of lepidopteran pupae or cocoons and even fewer have investigated species that are not forest pests. Here we present a study on overwinter survival for two moth species in the family Limacodidae, a group of polyphagous species found in deciduous forests. We placed cocoons of the saddleback caterpillar, Acharia stimulea (Clemens), and the spiny oak-slug caterpillar, Euclea delphinii (Boisduval), in the field under saplings of six different tree species and monitored predation and survival. This is the first study to examine predation rate among different host plants within a site. We found that cocoon predation was fairly high and differed significantly between limacodid species (29% for A. stimulea vs. 22% for E. delphinii). Predation rate did not differ among the six host plant species that we tested and also did not vary annually. Through phenotypic selection analyses, we found that cocoon mass affected both the likelihood of predation and overwinter survival; larger cocoons were less likely to be depredated and more likely to successfully emerge the following year. Overall our results indicate that cocoon predation is an important source of mortality for these two limacodid species and that there may be positive selection for greater cocoon mass for both limacodid species.  相似文献

Background  

The alteration of fatty acid profiles in soybean [Glycine max (L.) Merr.] to improve soybean oil quality is an important and evolving theme in soybean research to meet nutritional needs and industrial criteria in the modern market. Soybean oil with elevated oleic acid is desirable because this monounsaturated fatty acid improves the nutrition and oxidative stability of the oil. Commodity soybean oil typically contains 20% oleic acid and the target for high oleic acid soybean oil is approximately 80% of the oil; previous conventional plant breeding research to raise the oleic acid level to just 50-60% of the oil was hindered by the genetic complexity and environmental instability of the trait. The objective of this work was to create the high oleic acid trait in soybeans by identifying and combining mutations in two delta-twelve fatty acid desaturase genes, FAD2-1A and FAD2-1B.  相似文献

Background

Mutations in the dihydrofolate reductase (dhfr) and dihydropteroate synthase (dhps) genes of Plasmodium falciparum are associated with resistance to anti-folate drugs, most notably sulphadoxine-pyrimethamine (SP). Molecular studies document the prevalence of these mutations in parasite populations across the African continent. However, there is no systematic review examining the collective epidemiological significance of these studies. This meta-analysis attempts to: 1) summarize genotype frequency data that are critical for molecular surveillance of anti-folate resistance and 2) identify the specific challenges facing the development of future molecular databases.

Methods

This review consists of 220 studies published prior to 2009 that report the frequency of select dhfr and dhps mutations in 31 African countries. Maps were created to summarize the location and prevalence of the highly resistant dhfr triple mutant (N51I, C59R, S108N) genotype and dhps double mutant (A437G and K540E) genotype in Africa. A hierarchical mixed effects logistic regression was used to examine the influence of various factors on reported mutant genotype frequency. These factors include: year and location of study, age and clinical status of sampled population, and reporting conventions for mixed genotype data.

Results

A database consisting of dhfr and dhps mutant genotype frequencies from all African studies that met selection criteria was created for this analysis. The map illustrates particularly high prevalence of both the dhfr triple and dhps double mutant genotypes along the Kenya-Tanzania border and Malawi. The regression model shows a statistically significant increase in the prevalence of both the dhfr triple and dhps double mutant genotypes in Africa.

Conclusion

Increasing prevalence of the dhfr triple mutant and dhps double mutant genotypes in Africa are consistent with the loss of efficacy of SP for treatment of clinical malaria in most parts of this continent. Continued assessment of the effectiveness of SP for the treatment of clinical malaria and intermittent preventive treatment in pregnancy is needed. The creation of a centralized resistance data network, such as the one proposed by the WorldWide Antimalarial Resistance Network (WWARN), will become a valuable resource for planning timely actions to combat drug resistant malaria.  相似文献

Many endocarditis pathogens activate human platelets and this has been proposed to contribute to virulence. Here we report for the first time that many clinical isolates of Enterococcus faecalis, a common pathogen in infective endocarditis, aggregate human platelets. 84 isolates from human blood and urine were screened for their ability to aggregate platelets from four different donors. Platelet aggregation occurred for between 11 and 65% of isolates depending on the donor. In one donor, a significantly larger proportion of isolates from blood than from urine caused platelet aggregation. Median time to aggregation was 11 min and had a tendency to be shorter for blood isolates as compared to urine isolates. Immunoglobulin G (IgG) was shown to be essential in mediating activation and aggregation. Platelet aggregation could be abolished by an IgG-specific proteinase (IdeS), by an antibody blocking FcRγIIa on platelets, or by preabsorption of plasma with an E. faecalis isolate. Fibrinogen binding to bacteria or platelets does not contribute to platelet activation or aggregation under our experimental conditions. These results indicate that platelet activation and aggregation by E. faecalis is dependent on both host and bacterial factors and that it may be involved in the pathogenesis of invasive disease with this organism.  相似文献

A Synechococcus sp. strain PCC 7002 Fe bioreporter was constructed containing the isiAB promoter fused to the Vibrio harveyi luxAB genes. Bioreporter luminescence was characterized with respect to the free ferric ion concentration in trace metal-buffered synthetic medium. The applicability of the Fe bioreporter to assess Fe availability in the natural environment was tested by using samples collected from the Baltic Sea and from the high-nutrient, low-chlorophyll subarctic Pacific Ocean. Parallel assessment of dissolved Fe and bioreporter response confirmed that direct chemical measurements of dissolved Fe should not be considered alone when assessing Fe availability to phytoplankton.  相似文献

Lithic photoautotrophic communities function as principal primary producers in the world's driest deserts, yet many aspects of their ecology remain unknown. This is particularly true for Asia, where some of the Earth's oldest and driest deserts occur. Using methods derived from plant landscape ecology, we measured the abundance and spatial distribution of cyanobacterial colonization on quartz stony pavement across environmental gradients of rainfall and temperature in the isolated Taklimakan and Qaidam Basin deserts of western China. Colonization within available habitat ranged from 0.37+/-0.16% to 12.6+/-1.8%, with cold dry desert sites exhibiting the lowest abundance. Variation between sites was most strongly correlated with moisture-related variables and was independent of substrate availability. Cyanobacterial communities were spatially aggregated at multiple scales in patterns distinct from the underlying rock pattern. Site-level differences in cyanobacterial spatial pattern (e.g. mean inter-patch distance) were linked with rainfall, whereas patchiness within sites was correlated with local geology (greater colonization frequency of large rocks) and biology (dispersal during rainfall). We suggest that cyanobacterial patchiness may also in part be self-organized - that is, an outcome of soil water-biological feedbacks. We propose that landscape ecology concepts and models linking desert vegetation, biological feedbacks and ecohydrological processes are applicable to microbial communities.  相似文献
