Equilibrium, thermochemical, and time-resolved fluorescence measurements have been carried out in order to compare pig heart lactate dehydrogenase (LDH) and cytoplasmic malate dehydrogenase (MDH). The differences in the thermodynamic parameters for binding of NADH and NAD+ show the same pattern for both enzymes. The stronger binding of NADH is entropy-based, which can be understood as reflecting electrostatic interactions. The tryptophan fluorescence of MDH and LDH differ for the free enzymes and in quenching by NADH. The differences can be accounted for in terms of a single long-lived tryptophan residue present in LDH and not in MDH.  相似文献

Bovine muscle pyruvate kinase was inactivated by treatment with trinitrobenzenesulfonic acid; approximately one trinitrophenyl group was incorporated per subunit. ADP or Mg-ADP decreased the rate of inactivation but Mg++ alone or phosphoenolpyruvate had no effect. The inactivated protein was treated with trypsin and the trinitrophenylated peptide isolated by gel filtration. Homogeneity of the isolated peptide was shown by high voltage electrophoresis and high pressure liquid chromatography. Amino acid analysis and sequence determination revealed the presence of an acidic peptide 34 amino acids long and containing ?-trinitrophenylated lysine.  相似文献

The micellar properties of mixed, bovine gangliosides and purified galactosyl-N-acetylacetylgalactosaminyl (N-acetylneuraminyl) galactosylglucosylceramide were studied by gel filtration, equilibrium dialysis, and band and boundary centrifugation in sucrose gradients. The dissociation of micelles is very slow (days) in water and required us to approach equilibrium by association of monomers rather than by the dissociation of micelles. The gangliosides were therefore first converted into very low molecular weight aggregates (1-3 molecules) by dissolving them in Me2SO. Galactosyl-N-acetylgalactosaminyl(N-acetylneuraminyl)galactosylglucosylceramide was then diluted into aqueous sucrose gradients and sedimented by the boundary centrifugation technique. This gave a sedimenting micelle and a nonsedimenting monomer concentration of (1-2) x 10-10 M (or less) which corresponds to the critical micelle concentration value. The mixed gangliosides revealed two micellar sizes (i.e., 10 and 4.5 S), the slower sedimenting species being formed from the larger one with time (days). The critical micelle concentration of the mixed gangliosides was found to be approximately 10-8 M by a gel filtration, equilibrium dialysis, and band centrifugation.  相似文献

The lymphocyte surface membranes from normal and leukaemic or lymphomatous cells from man and mouse were isolated, characterized, and analyzed both biochemically and by diphenyl hexatriene fluorescence polarization. The cholesterol/phospholipid molar ratio for all the pure lymphocyte plasma membranes was 0.45--0.50, and the fluorescence polarization results showed that values much higher than this were not credible. The lipid composition of all the plasma membranes was remarkably similar, except for the concentration of free fatty acids and glycerides. The latter two were particularily high in the mouse lymphoma membrane and these, rather than a low cholesterol concentration, were responsible for the increased fluidity of the cells. The most prominent protein in most of the plasma membrane preparations was actin. This is found only by some authors, and its presence probably depends on the method of lymphocyte disruption.  相似文献

The role of four forms of cytochrome P-450 from rabbit liver in the metabolic activation of two suspected carcinogens, 2-aminoanthracene and benz[α]pyrene, was investigated with a S. typhimurium tester strain, TA 98. Each of the forms, 2,3,4 and 6 was reconstituted with NADPH-cytochrome P-450 reductase and lipid, and assay conditions were established such that the cytochrome P-450 concentration was rate-limiting. Under these conditions, cytochrome P-450 form 4, but not the other forms, converted 2-aminoanthracene into a potent mutagen. In contrast, form 6 was the only form which metabolized benz[α]pyrene to a mutagen. These results indicate that specific cytochrome P-450 forms preferentially activate particular mutagens.  相似文献

Vanadate produces a positive inotropic effect on ventricular muscle from rat, rabbit, guinea pig and cat; a positive inotropic effect on the atria of rat and rabbit, but a negative inotropic effect on the atria of guinea pig and cat. The effects of vanadate are completely reversible and occur in a concentration range of 10?5M to 10?3M. In this same concentration range, vanadate also causes a marked activation of cardiac adenylate cyclase suggesting that the positive inotropic action might be due in part to an elevation of cyclic AMP levels. The effects of vanadate are not influenced by alprenolol, cimetidine, or mepyramine, indicating a lack of involvement of β-adrenergic or histamine H2 and H1 receptors.  相似文献

Ultraviolet light-induced unscheduled DNA synthesis in primary cultures of human placentae examined as a function of radiation-dose and repair-incubation period was found to be dependent upon cell type and independent of gestational age. Primary cultures obtained by continuous harvesting of enzymatically released cells from fragments of 11-week and term specimens contained cytotrophoblasts and fibroblasts. Fibroblasts exhibited 3-fold more repair than did cytotrophoblasts from the same organ at both 11 weeks and term.  相似文献
