近年来,肝炎发病率高,青叶胆(Swertia mileensis)因此被大量采挖,野生青叶胆资源逐年减少。为发掘新药源,我们对云南产狭叶獐牙菜(S.angustifolia)和显脉獐牙菜(S.nervasa)的化学成分进行了研究。本文报道从中分离鉴定的4个单萜环烯醚甙Ⅰ—Ⅳ。Ⅰ、Ⅲ、Ⅳ分别为维哥罗甙(vegeloside)、獐牙菜甙(sweroside)和獐牙菜苦甙(swertiamarin),Ⅱ是Ⅰ的同分异构体,命名为显脉獐牙菜甙(nervoside)。显脉獐牙菜甙(Ⅱ)白色无定形粉末,味极苦。UVλ-_(max)~(EtOH):243nm(log ε-3.89)。IR v_(max)~(KBr) cm~(-1):3400、1700和1610,显示单萜环烯醚的特征吸收峰显脉獐牙菜;显脉獐牙菜甙;单萜环烯醚甙 相似文献

Patch-clamping and cell imageanalysis techniques were used to study the expression of thevolume-activated Cl current,ICl(vol), and regulatory volume decrease (RVD)capacity in the cell cycle in nasopharyngeal carcinoma cells (CNE-2Z). Hypotonic challenge caused CNE-2Z cells to swell and activated aCl current with a linear conductance, negligibletime-dependent inactivation, and a reversal potential close to theCl equilibrium potential. The sequence of anionpermeability was I > Br > Cl > gluconate. The Cl channelblockers tamoxifen, 5-nitro-2-(3-phenylpropylamino)benzoic acid (NPPB),and ATP inhibited ICl(vol). Synchronous cultures of cells were obtained by the mitotic shake-off technique and by adouble chemical-block (thymidine and hydroxyurea) technique. Theexpression of ICl(vol) was cell cycle dependent,being high in G1 phase, downregulated in S phase, butincreasing again in M phase. Hypotonic solution activated RVD, whichwas cell cycle dependent and inhibited by the Cl channelblockers NPPB, tamoxifen, and ATP. The expression of ICl(vol) was closely correlated with the RVDcapacity in the cell cycle, suggesting a functional relationship.Inhibition of ICl(vol) by NPPB (100 µM)arrested cells in G0/G1. The data also suggest that expression of ICl(vol) and RVD capacity areactively modulated during the cell cycle. The volume-activatedCl current associated with RVD may therefore play animportant role during the cell cycle progress. 相似文献

AIMS: To optimize the production of linolenic acid by disrupted mycelia of Mortierella isabellina. METHODS AND RESULTS: Effects of incubation conditions such as incubation time, pH of reaction mixture, concentration of Mg2+ or malate and incubation temperature on production of linolenic acid were studied. The production of gamma-linolenic acid reached 224 mg g-1 dry cells when the reaction mixture was composed of 1.0 g (dry mycelial mass) of disrupted mycelia of M. isabellina, 50 ml (50 mmol l(-1)) potassium phosphate buffer supplemented with 0.312 mmol l(-1) of Mg2+ and 10 mmol l(-1) of malate, pH 7.0 and incubated at 5 degrees C for 1 day. CONCLUSIONS: Incubation temperature, concentration of Mg2+ and malate showed major effects on the increased linolenic acid production. SIGNIFICANCE AND IMPACT OF THE STUDY: This study highlights conditions for increasing gamma-linolenic acid production by cell-free mycelia of M. isabellina and an insight into rapidly gaining high production of polyunsaturated fatty acids. 相似文献

Disruption of Smad4 in neural crest cells leads to mid-gestation death with pharyngeal arch, craniofacial and cardiac defects 总被引:1,自引:0,他引:1
TGFβ/BMP signaling pathways are essential for normal development of neural crest cells (NCCs). Smad4 encodes the only common Smad protein in mammals, which is a critical nuclear mediator of TGFβ/BMP signaling. In this work, we sought to investigate the roles of Smad4 for development of NCCs. To overcome the early embryonic lethality of Smad4 null mice, we specifically disrupted Smad4 in NCCs using a Cre/loxP system. The mutant mice died at mid-gestation with defects in facial primordia, pharyngeal arches, outflow tract and cardiac ventricles. Further examination revealed that mutant embryos displayed severe molecular defects starting from E9.5. Expression of multiple genes, including Msx1, 2, Ap-2α, Pax3, and Sox9, which play critical roles for NCC development, was downregulated by NCC disruption of Smad4. Moreover, increased cell death was observed in pharyngeal arches from E10.5. However, the cell proliferation rate in these areas was not substantially altered. Taken together, these findings provide compelling genetic evidence that Smad4-mediated activities of TGFβ/BMP signals are essential for appropriate NCC development. 相似文献

Semi-interpenetrating polymer network (semi-IPN) hydrogels were prepared by UV irradiation of water-soluble N-carboxylethyl chitosan (CECS) and 2-hydroxyethyl methacrylate (HEMA) aqueous solutions in the presence of D-2959 as photoinitiator. Hydrogels were characterized by using scanning electron microscopy (SEM), thermal gravimetric analysis (TGA) and X-ray diffractometry (XRD). SEM showed that semi-IPN hydrogels displayed porous surface and therefore had high surface area. XRD indicated that CECS/poly (HEMA) semi-IPN hydrogels had amorphous structure. The thermal stability and equilibrium degree of swelling improved obviously with increase of CECS content. Differential scanning calorimetry (DSC) indicated that free water content increased with increase of CECS content while bonded water content decreased. Cytotoxicity results suggested that semi-IPN hydrogels had good biocompatibility. From these preliminary evaluations, it is possible to conclude that these materials have potential applications in the biomedical field. 相似文献
