Pre‐replication complex (pre‐RC) is critical for DNA replication initiation. CDT1 and MCM2 are the subunits of pre‐RC, and proper regulation of CDT1 and MCM2 are necessary for DNA replication and cell proliferation. The present study aimed to explore the role of CDT1 and MCM2 in oocyte meiotic maturation and early embryonic development. The depletion and overexpression of Cdt1 and Mcm2 in oocyte and zygote were achieved by microinjecting specific siRNA and mRNA to explored their functions in oocyte meiotic maturation and embryonic development. Then, we examined the effect of CDT1 and MCM2 on other signal pathways by immunostaining the expression of related maker genes. We showed that neither depletion nor overexpression of Cdt1 affected oocyte meiotic progressions. The CDT1 was degraded in S phase and remained at a low level in G2 phase of zygote. Exogenous expression of Cdt1 in G2 phase led to embryo attest at zygote stage. Mechanistically, CDT1 overexpression induced DNA re‐replication and thus DNA damage check‐point activation. Protein abundance of MCM2 was stable throughout the cell cycle, and embryos with overexpressed MCM2 could develop to blastocysts normally. Overexpression or depletion of Mcm2 also had no effect on oocyte meiotic maturation. Our results indicate that pre‐RC subunits CDT1 and MCM2 are not involved in oocyte meiotic maturation. In zygote, CDT1 but not MCM2 is the major regulator of DNA replication in a cell cycle dependent manner. Furthermore, its'' degradation is essential for zygotes to prevent from DNA re‐replication in G2 stage.Pre‐replication complex (pre‐RC) is formed in G1 phase during which CDT1 is localized in nucleus. When zygote enters S phase, CDT1 is degraded and stays at a low level in G2 phase. However, the expression of exogenous Cdt1 mRNA in G2 phase of fertilized egg, mimicking events in which CDT1 degradation is disrupted, pre‐RC reassembles and leads to DNA re‐replication, and thus DNA damage check point activation, which results in embryo arrest at G2/M phase. 相似文献

The dibenzothiophene (DBT) desulfurization pathway of a facultative thermophilic bacterium Mycobacterium sp. X7B was investigated. Metabolites were identified by gas chromatography-mass spectrometry, and the results showed that 2-hydroxybiphenyl, the end product of the previously reported sulfur-specific pathway (also called 4S pathway), was further converted to 2-methoxybiphenyl. This is the first strain to possess this ability and therefore, an extended 4S pathway was determined. In addition, the DBT-desulfurizing bacterium Mycobacterium sp. X7B was able to grow on DBT derivatives such as 4-methylDBT and 4,6-dimethylDBT. Resting cells could desulfurize diesel oil (total sulfur, 535 ppm) after hydrodesulfurization. GC flame ionization detection and GC atomic emission detection analyses were used to qualitatively evaluate the effect of Mycobacterium sp. X7B treatment on the content of the diesel oil. The total sulfur content of the diesel oil was reduced 86% using resting cell biocatalysts for 24 h at 45 degrees C. 相似文献

Noxious stimuli applied at doses close to but below the threshold of cell injury induce adaptive responses that provide a defense against additional stress. Epileptic preconditioning protects neurons against status epilepticus and ischemia; however, it is not known if the converse is true. During hypoxia/ischemia (H/I), lactate released from astrocytes is taken up by neurons and is stored for energy, a process mediated by monocarboxylate transporter 4 (MCT4) in astroglia. The present study investigated whether H/I preconditioning can provide protection to neurons against epilepsy through upregulation of MCT4 expression in astrocytes in vitro and in vivo. An oxygen/glucose deprivation protocol was used in primary astrocyte cultures, while rats were subjected to an intermittent hypoxia preconditioning (IHP) paradigm followed by lithium-pilocarpine-induced epilepsy as well as lactate transportation inhibitor injection, with a subsequent evaluation of protein expression as well as behavior. H/I induced an upregulation of MCT4 expression, while an IHP time course of 5 days provided the greatest protection against epileptic seizures, which was most apparent by 3 days after IHP. However, lactate transport function disturbances can block the protective effect induced by IHP. These findings provide a potential basis for the clinical treatment of epilepsy. 相似文献

A series of physostigmine analogues were prepared and evaluated for cholinesterase inhibition activities, including acetylcholinesterase (AChE) and butyrylcholinesterase (BChE). 脱落酸(abscisic acid,ABA)对植物的生长发育具有独特的调控功能,并在植物适应逆境环境中发挥重要作用。9-顺式环氧类胡萝卜素双加氧酶(NCED)是高等植物中ABA生物合成途径的一个关键酶。根据GenBank中的植物NCED基因的同源序列设计简并引物,通过RT-PCR及RACE技术从枸杞叶片中克隆到1个编码NCED的基因,命名为LbNCED。其cDNA全长为2316 bp,含有1个1824 bp的开放阅读框,编码1个含607氨基酸残基,分子量为67.38 kDa、等电点(pI)为6.43的假定蛋白,其氨基酸序列与番茄(Lycopersicon esculentum)和马铃薯(Solanum tuberosum)的同源性达90%,在N-末端具有1个含15个氨基酸的叶绿体转运肽。Southern杂交结果表明,该基因在枸杞基因组中以低拷贝形式存在。盐处理和脱水处理的枸杞叶片中LbNCED基因的表达与内源ABA的积累同步变化。 相似文献

Revegetation is a traditional practice widely used for soil and water conservation on the Loess Plateau in China. However, there has been a lack of reports on soil microbial–biochemical indices required for a comprehensive evaluation of the success of revegetation systems. In this study, we examined the effects of revegetation on major soil nutrients and microbial–biochemical properties in an artificial alfalfa grassland, an enclosed natural grassland, and an artificial shrubland (Caragana korshinskii), with an abandoned cropland as control. Results showed that at 0–5, 5–20, and 20–40 cm depths, soil organic carbon, alkaline extractable nitrogen and available potassium were higher in natural grassland and artificial shrubland compared with artificial grassland and abandoned cropland. Soil microbial biomass C (Cmic) and phosphorous (Pmic) substantially decreased with depth at all sites, and in abandoned cropland was significantly lower than those of natural grassland, artificial grassland, and artificial shrubland at the depth of 0–5 cm. Soil microbial biomass N (Nmic) was higher in artificial shrubland and abandoned cropland compared with that in natural and artificial grasslands. Both Cmic and Pmic were significantly different between the 23‐year‐old and the 13‐year‐old artificial shrublands at the 0–5 cm depth. The activities of soil invertase, urease, and alkaline phosphatase in natural grassland and artificial shrubland were higher than those in artificial grassland and abandoned cropland. This study demonstrated that the regeneration of both natural grassland and artificial shrubland effectively preserved and enhanced soil microbial biomass and major nutrient cycling, thus is an ecologically beneficial practice for recovery of degraded soils on the Loess Plateau. 相似文献
