Endothelin-1 (ET-1) is a potent vasoconstrictorpeptide, which also potentiates contractions to norepinephrine in humaninternal mammary and coronary vessels. Exercise causes a redistribution of blood flow, i.e., the increase in working muscles that is partly attributable to a decrease in visceral blood flow. We hypothesized thatexercise causes a tissue-specific increase in ET-1 expression ininternal organs. We studied whether exercise affects expression ofpreproET-1 mRNA in the kidneys and lungs. The rats performed treadmillrunning (0% grade) for 45 min at a speed of 25 m/min. The plasmaconcentrations of ET-1, epinephrine, and norepinephrine were greater inthe exercise rats than in the sedentary control rats. The expression ofpreproET-1 mRNA in the kidneys was markedly higher in the exercise ratsthan in the sedentary control rats, whereas that in the lungs did notdiffer between the two groups. Therefore, the present study provides apossibility that the exercise-induced increase in production of ET-1 inthe kidneys causes vasoconstriction and hence decreases blood flow inthe kidneys through its direct vasoconstrictive action and/orits indirect effect of enhancing vasoconstrictions to norepinephrine.

The effects of sugars on the glucosylation of exogenous hydroquinone(HQ) was investigated by supplying individual sugars simultaneouslywith HQ to a suspension culture of Catharanthus roseus cells.The production of arbutin was enhanced as much as 2- to 3-foldby sucrose or glucose at concentrations of up to 6%, with theenhancement being directly dependent on the concentration ofthe sugar. The exogenously added sugar was not metabolized andremained unchanged. Sorbitol also promoted the production ofarbutin in a similar manner. Sucrose improved the viability of cells and, in cultures suppliedwith sucrose and HQ, the activity of UDP-glucose: hydroquinoneglucosyltransferase increased over a much longer period of timethan that in control cultures supplemented with HQ only. Fourteen species of fossil wood belonging to eleven genera and seven families were identified from 38 well preserved specimens collected from the Lower Coal-bearing Formation of Janggi Group at Donghae-myeon, Pohang City, Gyeongsangbuk-do Prefecture, Korea. Seven new taxa were found and described; they are Carya koreana Jeong et Kim, Betula janggiensis Jeong et Kim, Carpinus donghaensis Jeong et Kim, Ostrya geumgwangensis Jeong et Kim, Stewartia pseudo-camellioxylon Jeong et Kim, Acer minokamoensis Jeong, Kim et Suzuki and Acer pohangensis Jeong et Kim. The most abundant taxa are Betulaceae, Aceraceae, Ulmaceae, and Wataria. Except Wataria of which the habitat preference has not been confirmed, these taxa are major elements of cool-temperate vegetation and are similar to the Aniai-type flora of Japan. Previous studies of fossil plants from the Geumgwangdong Shale (leaves and seeds), the Upper Coal-bearing Formation (fossil woods) and the Yeonil Group (leaves and seeds) and this study, show that the climate of the Pohang Basin changed from cool-temperate to warm-temperate and subtropical during the Miocene.  相似文献

Reactive oxygen species (ROS) primarily produced via NADPH oxidase play an important role for killing microorganisms in neutrophils. In this study we examined if ROS production in Human promyelocytic leukemia cells (HL60) differentiated into neutrophil-like cells (dHL60) induces ER stress and activates the unfolded protein response (UPR). To cause ROS production cells were treated with PMA or by chronic hyperglycemia. Chronic hyperglycemia failed to induce ROS production and did not cause activation of the UPR in dHL60 cells. PMA, a pharmacologic NADPH oxidase activator, induced ER stress in dHL60 cells as monitored by IRE-1 and PERK pathway activation, and this was independent of calcium signaling. The NADPH oxidase inhibitor, DPI, abolished both ROS production and UPR activation. These results show that ROS produced by NADPH oxidase induces ER stress and suggests a close association between the redox state of the cell and the activation of the UPR in neutrophil-like HL60 cells.  相似文献

Antibody-dependent cellular cytotoxicity (ADCC) is an important effector function determining the clinical efficacy of therapeutic antibodies. Core fucose removal from N-glycans on the Fc portion of immunoglobulin G (IgG) improves the binding affinity for Fcγ receptor IIIa (FcγRIIIa) and dramatically enhances ADCC. Our previous structural analyses revealed that Tyr–296 of IgG1-Fc plays a critical role in the interaction with FcγRIIIa, particularly in the enhanced FcγRIIIa binding of nonfucosylated IgG1. However, the importance of the Tyr–296 residue in the antibody in the interaction with various Fcγ receptors has not yet been elucidated. To further clarify the biological importance of this residue, we established comprehensive Tyr–296 mutants as fucosylated and nonfucosylated anti-CD20 IgG1s rituximab variants and examined their binding to recombinant soluble human Fcγ receptors: shFcγRI, shFcγRIIa, shFcγRIIIa, and shFcγRIIIb. Some of the mutations affected the binding of antibody to not only shFcγRIIIa but also shFcγRIIa and shFcγRIIIb, suggesting that the Tyr–296 residue in the antibody was also involved in interactions with FcγRIIa and FcγRIIIb. For FcγRIIIa binding, almost all Tyr–296 variants showed lower binding affinities than the wild-type antibody, irrespective of their core fucosylation, particularly in Y296K and Y296P. Notably, only the Y296W mutant showed improved binding to FcγRIIIa. The 3.00 Å-resolution crystal structure of the nonfucosylated Y296W mutant in complex with shFcγRIIIa harboring two N-glycans revealed that the Tyr-to-Trp substitution increased the number of potential contact atoms in the complex, thus improving the binding of the antibody to shFcγRIIIa. The nonfucosylated Y296W mutant retained high ADCC activity, relative to the nonfucosylated wild-type IgG1, and showed greater binding affinity for FcγRIIa. Our data may improve our understanding of the biological importance of human IgG1-Fc Tyr–296 in interactions with various Fcγ receptors, and have applications in the modulation of the IgG1-Fc function of therapeutic antibodies.  相似文献

Serum soluble interferon-α/β receptor (sIFN-α/βR) and high-sensitivity C-reactive protein (hs-CRP) levels were evaluated in the patients with gastrointestinal and hepatobiliary-pancreatic cancer. We compared the sensitivity and specificity of serum sIFN-α/βR with that of serum hs-CRP and evaluated the two diagnostic parameters in combination. Serum sIFN-α/βR levels were measured in 92 patients and 25 healthy individuals by enzyme-linked immunosorbent assay. The diagnoses were 37 cases of hepatocellular carcinoma, 17 cases of pancreatic cancer, 15 cases of colon cancer, 13 cases of biliary tract cancer, and 10 cases of gastric cancer. Serum levels of sIFN-α/βR and hs-CRP were significantly higher in the patients than in healthy individuals (p < 0.05). The optimal cut-off values of sIFN-α/βR and hs-CRP were 3600 pg/ml and 0.5 μg/ml, respectively. The sensitivity and specificity for these thresholds were 94.6% and 88.0%, whereas positive predictive and negative predictive values were 96.7% and 81.5%. These results suggest that a combination of serum sIFN-α/βR and hs-CRP thresholds may be more reliable diagnostic parameter for gastrointestinal and hepatobiliary-pancreatic cancer.  相似文献

Introduction – Moso‐bamboo (Phyllostachys pubescens) is well known as an edible shoot in Asia, and the stems of moso‐bamboo are used as tableware due to its characteristic odour. Despite the pleasant odour of bamboo stems, no detailed analysis of the volatile compounds has been reported. Objective – To clarify the potent odourants contributing to the characteristic aroma of the bamboo, the aroma extract dilution analysis (AEDA) method was performed through gas chromatography olfactometry (GC‐O) analysis. In addition, relative flavour activity (RFA) was calculated, in which both the flavor dilution (FD) factor and weight percentage of each compound are involved. Results – Eighty‐nine compound in bamboo stems oil were identified by GC and GC‐MS. The main components of the oil were palmitic acid (16.5%), (E)‐nerolidol (10.2%) and indole (8.1%). In sensory analysis, 18 aroma‐active compounds were detected by aroma extract dilution analysis (AEDA). The most intense aroma‐active compounds were eugenol (sweet, clove‐like, green) and (E)‐2‐nonenal (green). Conclusion – The results of the sniffing test, RFA and FD factor indicated that (E)‐2‐nonenal and eugenol were estimated to have a bamboo‐like aroma, and aldehyde compounds, such as a phenylacetaldehyde (floral) and C9–C10 unsaturated aldehydes, make the aroma of bamboo. A new type of dendritic molecules Gd-DTPA-XDA-D1-Glc(OH), which work as a functionalized ligand coordinating gadolinium(III) ion at the center of their frameworks with two glucose moieties on the molecular surfaces, were readily synthesized with high yield. The recently described human anion channel Anoctamin (ANO) protein family comprises at least ten members, many of which have been shown to correspond to calcium-activated chloride channels. To date, the only reported human mutations in this family of genes are dominant mutations in ANO5 (TMEM16E, GDD1) in the rare skeletal disorder gnathodiaphyseal dysplasia. We have identified recessive mutations in ANO5 that result in a proximal limb-girdle muscular dystrophy (LGMD2L) in three French Canadian families and in a distal non-dysferlin Miyoshi myopathy (MMD3) in Dutch and Finnish families. These mutations consist of a splice site, one base pair duplication shared by French Canadian and Dutch cases, and two missense mutations. The splice site and the duplication mutations introduce premature-termination codons and consequently trigger nonsense-mediated mRNA decay, suggesting an underlining loss-of-function mechanism. The LGMD2L phenotype is characterized by proximal weakness, with prominent asymmetrical quadriceps femoris and biceps brachii atrophy. The MMD3 phenotype is associated with distal weakness, of calf muscles in particular. With the use of electron microscopy, multifocal sarcolemmal lesions were observed in both phenotypes. The phenotypic heterogeneity associated with ANO5 mutations is reminiscent of that observed with Dysferlin (DYSF) mutations that can cause both LGMD2B and Miyoshi myopathy (MMD1). In one MMD3-affected individual, defective membrane repair was documented on fibroblasts by membrane-resealing ability assays, as observed in dysferlinopathies. Though the function of the ANO5 protein is still unknown, its putative calcium-activated chloride channel function may lead to important insights into the role of deficient skeletal muscle membrane repair in muscular dystrophies.  相似文献
