A unique hepatitis C virus (HCV) strain JFH-1 has been shown to replicate efficiently in cell culture with production of infectious HCV. We previously developed a DNA expression system containing HCV cDNA flanked by two self-cleaving ribozymes to generate HCV particles in cell culture. In this study, we produced HCV particles of various genotypes, including 1a (H77), 1b (CG1b), and 2a (J6 and JFH-1), in the HCV-ribozyme system. The constructs also contain the secreted alkaline phosphatase gene to control for transfection efficiency and the effects of culture conditions. After transfection into the Huh7-derived cell line Huh7.5.1, continuous HCV replication and secretion were confirmed by the detection of HCV RNA and core antigen in the culture medium. HCV replication levels of strains H77, CG1b, and J6 were comparable, whereas the JFH-1 strain replicates at a substantially higher level than the other strains. To evaluate the infectivity in vitro, the culture medium of JFH-1-transfected cells was inoculated into naive Huh7.5.1 cells. HCV proteins were detected by immunofluorescence 3 days after inoculation. To evaluate the infectivity in vivo, the culture medium from HCV genotype 1b-transfected cells was inoculated into a chimpanzee and caused a typical course of HCV infection. The HCV 1b propagated in vitro and in vivo had sequences identical to those of the HCV genomic cDNA used for cell culture transfection. The development of culture systems for production of various HCV genotypes provides a valuable tool not only to study the replication and pathogenesis of HCV but also to screen for antivirals.  相似文献

Alterations in temperature (T) and vapor pressure deficit (VPD) strongly influence gas exchange, but because VPD is highly influenced by T, the effects of these two factors are difficult to separate. Here, the concomitant effects of T and VPD on CO(2) uptake, stomatal conductance, and transpiration at leaf- and canopy-levels were examined for a stand of trees (Populus deltoides) enclosed within large mesocosms. T and VPD were independently altered to yield a factorial combination of treatments of low (24 degrees C) or high (30 degrees C) T and low (0.75) or high (1.75 kPa) VPD. Traditional leaf-level gas exchange measurements were compared with whole-canopy exchange to verify typical scaling methods. Elevated T resulted in an average 40% and 14% increase in midday leaf-level and canopy-level net CO(2) uptake, respectively. Other physiological responses to elevated T and VPD were similar at both scales, but the magnitude of change was usually less pronounced at the canopy-level. Surprisingly, only minimal interactions between T and VPD were found to influence responses of CO(2) uptake and stomatal conductance at either level.  相似文献

The purpose of this research was to study the effect of the lipid matrix on the entrapment of olanzapine (OL). OL-loaded solid lipid nanoparticles (SLNs) were prepared using lipids like glyceryl monostearate (GMS), Precirol ATO 5 (PRE), glyceryl tristearate (GTS), and Witepsol E85 (WE 85)--and poloxamer 407 and hydrogenated soya phosphatidylcholine as stabilizers--using a hot melt emulsification high-pressure homogenization technique, and then characterized by particle size analysis, zeta potential, differential scanning calorimetry (DSC), and powder X-ray diffraction (pXRD). Homogenization at 10,000 psi for 3 cycles resulted in the formation of SLNs with a mean particle size of approximately 190 nm for the 4 lipids investigated. The highest partition coefficient for OL between the melted lipid and pH 7.4 phosphate buffer (pH 7.4 PB) was obtained with GTS. The entrapment efficiency was in the following order: GTS SLNs > PRE SLNs > WE 85 SLNs > GMS SLNs. DSC and pXRD showed that much of the incorporated fraction of OL existed in the amorphous state after incorporation into SLNs. A sharp increase in the flocculation of the SLN dispersions was observed upon addition of 0.6 M aqueous sodium sulfate solution. Nanoparticle surface hydrophobicity was in the following order: GTS SLNs > PRE SLNs > WE 85 SLNs > GMS SLNs. A significant increase in size and zeta potential was observed for GTS SLN and WE 85 SLN dispersions stored at 40 degrees C. Release of OL from the SLNs was sustained up to 48 hours in pH 7.4 PB and obeyed Higuchi's release kinetics.  相似文献

Tropical dry deciduous forests play a significant role in regulating the biogeochemical cycles. Present study assesses the carbon stock of tropical dry deciduous forests varying in tree density, basal cover, and diversity located in Singrauli district of Madhya Pradesh in Central India. Field sampling was carried out in six forest sites viz., Chitrangi, East Sarai, Gorbi, Renukoot, West Sarai, and Waidhan, of Singrauli. A total of 29 tree species belonging to 18 families were recorded across the forest ranges where tree density, basal area and diversity values varied from 702 (Gorbhi Range) – 1671 (East Sarai range) individuals ha?1; 15.43 (Renukhund range) – 71.76 m2 ha?1 (Chitrange range) and 0.69 (West Sarai range) – 2.52 (Gorbi range), respectively. Total biomass estimated ranged from 103.32 (Renukhund range) – 453.54 Mg ha?1 (Chitrange range) while the total tree carbon density varied from 48.97 to 214.97 Mg C ha?1. The variation in carbon storage in the studied ranges was found dependent on density of trees in different diameter and age classes and tree species diversity. Diospyros melanoxylon, Butea monosperma, Shorea robusta, Senegalia catechu, Spondias pinnata, and Lagerstroemia parviflora were the dominant species at different study sites (forest ranges) and contributed towards higher carbon storage in respective forest ranges. Study endorses field-based approach for carbon estimations based on above and belowground assessments as a more realistic approach to understand sink potential of natural forests.  相似文献

Towards a quest for establishing new antitubercular agents, we have designed new quinoline–triazole hybrid analogs in a six-step reaction sequence involving versatile reactions like Vilsmeier–Haack and click reaction protocol. The design is based on the structural modification of bedaquiline moiety and involves molecular hybridization approach. The structure of the synthesized product was elucidated by single crystal X-ray diffraction study. The synthesized target compounds were screened for their antitubercular activity against Mycobacterium bovis. Interestingly, two compounds of the series (8d and 8m) showed significant inhibition with MIC of 31.5 and 34.8 μM. Compounds bearing 3-fluoro phenyl and n-octyl groups on the 1,2,3-triazole ring emerged as the most potent leads among the compounds tested. Further these hit compounds were also screened for their cytotoxic effect on human embryonic kindey 293 (HEK293) cells and other cancer cell lines such as HeLa (Cervical), PC3 (Prostate), Panc-1 (Pancreatic) and SKOV3 (Ovarian) indicating to be safer with the minimal cytotoxicity.  相似文献

Incorporation of noncanonical disulfide linkages into single‐domain antibodies (sdAbs) has been shown to enhance thermostability and other properties. Here, we evaluated the effects of introducing a novel disulfide linkage formed between Cys residues at IMGT positions 40 and 55 on the melting temperatures (T ms), reversibility of thermal unfolding, solubility, and antigen‐binding affinities of three types of sdAbs (VHH, VH, and VL domains). The Cys40‐Cys55 disulfide linkage was tolerated by 9/9 VHHs, 12/12 VHs, and 2/11 VLs tested and its formation was confirmed by mass spectrometry. Using circular dichroism, we found that the Cys40‐Cys55 disulfide linkage increased sdAb T m by an average of 10.0°C (range: 0–21.8°C). However, enhanced thermostability came at the cost of a partial loss of refolding ability upon thermal denaturation as well as, for some sdAbs, significantly decreased solubility and antigen‐binding affinity. Thus, Cys40/Cys55 can be added to the panel of known locations for introducing stabilizing noncanonical disulfide linkages into antibody variable domains, although its effects should be tested empirically for individual sdAbs.  相似文献

Connexins, the constituent proteins of gap junctions, are transmembrane proteins. A connexin (Cx) traverses the membrane four times and has one intracellular and two extracellular loops with the amino and carboxyl termini facing the cytoplasm. The transmembrane and the extracellular loop domains are highly conserved among different Cxs, whereas the carboxyl termini, often called the cytoplasmic tails, are highly divergent. We have explored the role of the cytoplasmic tail of Cx32, a Cx expressed in polarized and differentiated cells, in regulating gap junction assembly. Our results demonstrate that compared with the full-length Cx32, the cytoplasmic tail-deleted Cx32 is assembled into small gap junctions in human pancreatic and prostatic cancer cells. Our results further document that the expression of the full-length Cx32 in cells, which express the tail-deleted Cx32, increases the size of gap junctions, whereas the expression of the tail-deleted Cx32 in cells, which express the full-length Cx32, has the opposite effect. Moreover, we show that the tail is required for the clustering of cell-cell channels and that in cells expressing the tail-deleted Cx32, the expression of cell surface-targeted cytoplasmic tail alone is sufficient to enhance the size of gap junctions. Our live-cell imaging data further demonstrate that gap junctions formed of the tail-deleted Cx32 are highly mobile compared with those formed of full-length Cx32. Our results suggest that the cytoplasmic tail of Cx32 is not required to initiate the assembly of gap junctions but for their subsequent growth and stability. Our findings suggest that the cytoplasmic tail of Cx32 may be involved in regulating the permeability of gap junctions by regulating their size.  相似文献

We report synthesis of a cerium‐activated strontium pyrophosphate (Sr2P2O7) phosphor using a high‐temperature combustion method. Samples were characterized by X‐ray diffraction (XRD), Fourier transform infrared spectroscopy (FT‐IR), photoluminescence (PL) and thermoluminescence (TL). The XRD pattern reveals that Sr2P2O7 has an α‐phase with crystallization in the orthorhombic space group of Pnam. The IR spectrum of α‐Sr2P2O7 displays characteristic bands at 746 and 1190 cm‐1 corresponding to the absorption of (P2O7)‐4. PL emission spectra exhibit a broad emission band around 376 nm in the near‐UV region due to the allowed 5d–4f transition of cerium and suggest its applications in a UV light‐emitting diode (LED) source. PL also reveals that the emission originates from 5d–4f transition of Ce3+ and intensity increases with doping concentration. TL measurements made after X‐ray irradiation, manifest a single intense glow peak at around 192°C, which suggests that this is an outstanding candidate for dosimetry applications. The kinetic parameters, activation energy and frequency factor of the glow curve were calculated using different analysis methods. Copyright © 2014 John Wiley & Sons, Ltd.  相似文献
