ObjectivePhosphorous is an essential micronutrient of plants and involved in critical biological functions. In nature, phosphorous is mostly present in immobilized inorganic mineral and in the fixed organic form including phytic acid and phosphoesteric compounds. However, the bioavailability of bound phosphorous could be enhanced by the use of phosphate solubilizing microorganisms such as bacteria and fungi. The phytases are widespread in an environment and have been isolated from different sources comprising bacteria and fungi.MethodologyIn current studies, we show the successful use of gamma rays and EMS (Ethyl Methane Sulphonate) mutagenesis for enhanced activity of phytases in a fungal strain Sporotrichum thermophile.ResultsWe report an improved strain ST2 that could produce a clear halo zone around the colony, up to 24 mm. The maximum enzymatic activity was found of 382 U/mL on pH 5.5. However, the phytase activity was improved to 387 U/ml at 45 °C. We also report that the mutants produced through EMS showed the greater potential for phytase production.ConclusionThe current study highlights the potential of EMS mutagenesis for strain improvement over physical mutagens. 相似文献
Grapevine moth, Lobesia botrana (Lep. Tortricidae) is a key pest of grape in Iran and other vineyards of the world. In this study, eight constant rearing temperatures (5, 10, 15, 20, 25, 30, 32 and 35 ± 1 °C) along with 60 ± 10% RH and a 16:8 (L:D) h photoperiod were chosen for demographic studies of the grapevine moth. Immature stages were unable to develop when reared at 5 and 35 °C, and the progeny moths were unable to successfully mate at 10, 15 and 32 °C. The overall developmental time of juveniles decreased at 30 °C (from 320.7 ± 3.4 d at 10 °C to 34.2 ± 0.2 d) followed by an increase to 42.5 ± 0.6 d at 32 °C. Based on values of the stable population growth parameters, the temperature of 25 °C was found to be optimal for propagation of grapevine moth. The highest values of the intrinsic rate of increase, gross and net reproductive rates were 0.0719 d??1, 55.5 and 23 females per generation, respectively, at 25 °C. Since jackknife and bootstrap estimates of mean and standard error were mainly similar, both methods may equally be used for uncertainty estimates. Our data suggest that cold storage of grapes will help to control grapevine moth infestations and damage. In many grape growing regions of Iran, the first generation is expected to cause damage. It is expected since our reproductive life table analysis suggests that the hot summer temperatures may restrict pest development during subsequent generations. 相似文献
Lepidoptera is the second most diverse insect order outnumbered only by the Coeleptera. Acetylcholinesterase (AChE) is the major target site for insecticides. Extensive use of insecticides, to inhibit the function of this enzyme, have resulted in the development of insecticide resistance. Complete knowledge of the target proteins is very important to know the cause of resistance. Computational annotation of insect acetylcholinesterase can be helpful for the characterization of this important protein. Acetylcholinesterase of fourteen lepidopteran insect pest species was annotated by using different bioinformatics tools. AChE in all the species was hydrophilic and thermostable. All the species showed lower values for instability index except L. orbonalis, S. exigua and T. absoluta. Highest percentage of Arg, Asp, Asn, Gln and Cys were recorded in P. rapae. High percentage of Cys and Gln might be reason for insecticide resistance development in P. rapae. Phylogenetic analysis revealed the AChE in T. absoluta, L. orbonalis and S. exigua are closely related and emerged from same primary branch. Three functional motifs were predicted in eleven species while only two were found in L. orbonalis, S. exigua and T. absoluta. AChE in eleven species followed secretory pathway and have signal peptides. No signal peptides were predicted for S. exigua, L. orbonalis and T. absoluta and follow non secretory pathway. Arginine methylation and cysteine palmotylation was found in all species except S. exigua, L. orbonalis and T. absoluta. Glycosylphosphatidylinositol (GPI) anchor was predicted in only nine species. 相似文献
The penicillin derivative amoxicillin (AMX) plays an important role in treating various types of infections caused by bacteria. However, excessive use of AMX may have negative health effects. Therefore, it is of utmost importance to detect and quantify the AMX in pharmaceutical drugs, biological fluids, and environmental samples with high sensitivity. Therefore, this review article provides valuable and up-to-date information on nanostructured material-based optical and electrochemical sensors to detect AMX in various biological and chemical samples. The role of using different nanostructured materials on the performance of important optical sensors such as colorimetric sensors, fluorescence sensors, surface-enhanced Raman scattering sensors, chemiluminescence/electroluminescence sensors, optical immunosensors, optical fibre-based sensors, and several important electrochemical sensors based on different electrode types have been discussed. Moreover, nanocomposites, polymer, and MXenes-based electrochemical sensors have also been discussed, in which such materials are being used to further enhance the sensitivity of these sensors. Furthermore, nanocomposite-based photo-electrochemical sensors and the market availability of biosensors including AMX have also been discussed briefly. Finally, the conclusion, challenges, and future perspectives of the above-mentioned sensing techniques for AMX detection are presented. 相似文献
Myzus persicae is a devastating pest affecting potato production. Intron-containing hairpin RNA (ihpRNA) silenced the odorant-binding protein 8 (OBP8) for enhanced protection against Myzus persicae in potatoes. OBP8 was isolated from M. persicae, sequenced, with the allotment of GenBank ID. ERNAi was used to design siRNA targets from OBP8 with no off-targets. Multiple Sequence Alignment show M. persicae OBP8 resemblance with Acyrthosiphon pisum, Rhopalosiphum maidis, Aphis fabae, and Sitobion avenae. dsRNA-OBP8 (7 µg/µL) oral feeding resulted in a 69% mortality, and 58% OBP8 reduced expression 8D post-feeding compared to control. Golden Gate cloning is used for RNA interference taking advantage of type IIs restriction enzyme Eco31I. ihpRNA-OBP8 introduced by agroinfiltration in Solanum tuberosum. Transgenic S. tuberosum fed Myzus show 57.6% mortality and 49% reduction in OBP8 expression 8D post-feeding, compared to control. This work proves OBP8 as promising ihpRNA targets in potato and related crops for whom Myzus is a destructive pest.
Present study provides an easy and efficient protocol for large scale clonal propagation of Coleus forskohlii, a threatened medicinal plant of commercial importance. Basal leaf lamina excised from upper three nodes of shoot was used as explant and its size, position, orientation and season of collection were initially optimized to select the most responsive explant condition. Enhanced shoot production and proliferation has been achieved on medium containing 2 μM BA + 0.1 μM NAA wherein, a highest number of 35 shoots/explant were produced. The regenerated shoots of varied length (3–5 cm) were transferred to root induction medium comprising of IBA, NAA and IAA (1–5 μM) in half-strength MS medium to determine the most suitable shoot length for proper root induction. Rooted plantlets were acclimatized in field conditions after proper hardening. Histological analysis was also carried out to confirm the nature of origin of shoot buds from leaf explants. 相似文献

This research work was carried out to compare the color stripping efficiency of optimized biological method with the chemical stripping, commonly employed in the textile industries. Knitted fabric dyed with Reactive black B dye in 2, 4 and 6% shades strengths was subjected to chemical and biological stripping processes individually. Biological stripping process was found many fold superior to chemical one. It was noted that shade strength does not showed any pronounced effect on the bursting strength of fabric but biological and chemical treatment affect the quality of fabrics in terms of bursting strength/durability of fabric. White rot fungus Ganoderma lucidum IBL-05 showed good potential for decolorization/color stripping of cotton fabric dyed with Reactive black B under optimized set of conditions. The chemical stripping technology is inferior to biological stripping process regarding the quality of fabric and percent color removal from cotton fabric dyed with Reactive black B dye. 相似文献
