There are increasing numbers of patients at risk for invasive fungal infections (IFI). The early diagnosis of IFI’s is limited by the lack of sensitivity and delayed turn-around time of routine cultures. Delays in diagnosis are associated with poor outcomes, especially with immunocompromised hosts and patients within the intensive care unit (ICU). Over the last decade, fungal biomarkers such as galactomannan and 1,3-β-D-glucan (BDG) have been increasingly utilized for the early and accurate diagnosis of these infections. This review will evaluate the recent clinical data on the BDG assay, focusing on its ability to assist in the diagnosis of invasive candidiasis in the ICU, and also whether BDG kinetics can predict the prognosis of the patient. Similarly, this review will evaluate BDG in invasive Aspergillus and Pneumocystis jiroveci diagnosis and prognosis, as well as the current limitations of the assay. 相似文献

G protein-coupled receptors (GPCRs) are a class of integral membrane proteins mediating physiological functions fundamental for survival, including energy homeostasis. A few years ago, an amino acid sequence of a novel GPCR gene was identified and named GPR178. In this study, we provide new insights regarding the biological significance of Gpr178 protein, investigating its evolutionary history and tissue distribution as well as examining the relationship between its expression level and feeding status. Our phylogenetic analysis indicated that GPR178 is highly conserved among all animal species investigated, and that GPR178 is not a member of a protein family. Real-time PCR and in situ hybridization revealed wide expression of Gpr178 mRNA in both the brain and periphery, with high expression density in the hypothalamus and brainstem, areas involved in the regulation of food intake. Hence, changes in receptor expression were assessed following several feeding paradigms including starvation and overfeeding. Short-term starvation (12–48h) or food restriction resulted in upregulation of Gpr178 mRNA expression in the brainstem, hypothalamus and prefrontal cortex. Conversely, short-term (48h) exposure to sucrose or Intralipid solutions downregulated Gpr178 mRNA in the brainstem; long-term exposure (10 days) to a palatable high-fat and high-sugar diet resulted in a downregulation of Gpr178 in the amygdala but not in the hypothalamus. Our results indicate that hypothalamic Gpr178 gene expression is altered during acute exposure to starvation or acute exposure to palatable food. Changes in gene expression following palatable diet consumption suggest a possible involvement of Gpr178 in the complex mechanisms of feeding reward. 相似文献

A new method for ultrathin grafting of pNIPAm on PDMS surfaces is introduced that employs plasma activation of the surface followed by thermal polymerization. This method is optimized for human primary SMC attachment and subsequent intact cell sheet detachment by lowering the temperature. The contractile gene expression of the cells showed that the contractile phenotype of the SMCs which is induced by aligning the cells through micropatterning is more preserved after thermoresponsive cell sheet detachment in contrast with enzymatic detachment. Given its simplicity and low cost, this thermoresponsive grafting method can be utilized for engineering patterned cell sheets for future bottom-up tissue engineering techniques. 相似文献

The green twigs of 1-year-old Eucalyptus microtheca F. Muell seedlings were cultured on modified MS medium, supplemented with α-naphthalene acetic acid (NAA) and kinetin (Kin)
hormones at 12 different concentrations. After 4 weeks, the combination of 1 mg l−1 NAA + 1 mg l−1 Kin induced the highest number of axillary shoots. Meanwhile, embryogenic calli were observed in media containing 4 mg l−1 NAA + 0.5 mg l−1 Kin, without any regeneration. The hormone treatments were followed by subculturing the twigs in different levels of thidiazuron
(TDZ). The combination of 1 mg l−1 NAA + 1 mg l−1 Kin together with 0.01 mg l−1 TDZ resulted in an increase of direct shoot, while higher amounts of TDZ led to adventitious shoot induction. Somatic embryogenesis
was observed in the treatment containing 0.01 mg l−1 TDZ + 4 mg l−1 NAA + 0.5 mg l−1Kin. The peroxidase (POD) band patterns in regenerated plantlets were investigated in order to determine the effect of different
levels of TDZ on loci synthesis. A dimer locus, a tetramer locus and two epigenetic bands (a new band for NAA + Kin and the
other for TDZ) were observed in the POD profiles. In case of low (0.01 mg l−1 and 0.1 mg l−1) levels of TDZ, one heterozygote allele was disappeared from dimer locus, while at higher TDZ levels, the dimer locus lost
its stability and tetramer locus showed a high activity. Thus, POD allele patterns seems to be a feasible marker for different

forms. The integrity and performance of an HPMC matrix formulation depends on rapid hydration and gel formation upon ingestion.
Due to the recent alert issued by the Food and Drug Administration regarding the potential negative influence of alcoholic
beverages on extended release (ER) formulations, several researchers have evaluated the potential influence of hydroalcoholic
media on drug release from ER dosage forms. It has been reported that HPMC matrix formulations do not show “dose dumping”
in hydroalcoholic media. The purpose of this study was a fundamental investigation on the effect of hydroalcoholic solutions
(0–40% v/v ethanol) on textural and rheological properties of different viscosity grades of neat HPMC, as the functional ingredient
within a hydrophilic matrix. In general, hydroalcoholic solutions had little effect on gel formation and mechanical properties
of hydrated compacts, while the rheological behavior of HPMC showed dependency on the ethanol content of such solutions. 相似文献

Puromycin aminonucleoside (PAN) has been known to induce proteinuria. The increased generation of reactive oxygen species (ROS) has been implicated in this toxicity of PAN. We have reported that PAN increases the synthesis of methylguanidine (MG) and creatol which are the products of the reaction of creatinine and the hydroxyl radical in isolated rat hepatocytes. However, the mechanism for the increased ROS induced by PAN is still unclear. In this paper, we investigate the role of protein kinase C (PKC) on the PAN induced reactive oxygen generation in isolated rat hepatocytes. Isolated hepatocytes were incubated in Krebs-Henseleit bicarbonate buffer containing 3% BSA, 16.6 mM creatinine and tested reagents. MG and creatol were determined by high-performance liquid chromatography using 9,10-phenanthrenequinone for the post-labeling. PAN increased MG and creatol synthesis in isolated rat hepatocytes by 60%. 1-(5-Isoquinolinesulfonyl)-2-methylpiperazine dihydrochloride (H-7), a PKC inhibitor, at 10 and 100 μM significantly inhibited MG and creatol synthesis with or without PAN. The inhibition rate is dose dependent from 10 to 100 μM. H1004, a reagent used as control for H-7, did not affect (at 10 μM) or increased little (at 100 μM) the synthesis of MG and creatol. Ro31-8425, a potent PKC inhibitor, significantly inhibited (at 10 μM) MG synthesis in the presence of PAN. PKC in the membrane fraction, a marker of PKC activation, increased over the initial concentration by a factor of 1.65-fold at 60 min incubation and 2.16-fold at 120 min with PAN, while it changed little without PAN. These results indicate that PAN activates PKC resulting in increased hydroxyl radical generation in isolated rat hepatocytes. 相似文献

Enantioselectivity of inhibition of cytochrome P450 3A4 (CYP3A4) by ketoconazole: Testosterone and methadone as substrates 总被引:4,自引:0,他引:4
Racemic ketoconazole (KTZ) was the first orally active azole antifungal agent used in clinical practice and has become widely used in the treatment of mucosal fungal infections associated with AIDS immunosuppression and cancer chemotherapy. However, the use of KTZ has been limited because of adverse drug-drug interactions. KTZ blocks ergosterol biosynthesis by inhibiting the fungal cytochrome P450 (CYP51). KTZ is also a potent inhibitor of human cytochrome P450 3A4 (CYP3A4) enzyme, the major drug-metabolizing CYP isozyme in the human liver. We examined the enantioselective differences of KTZ in the inhibition of human CYP3A4 and in antifungal action. Dextro- and levo-KTZ exhibited modest enantioselective differences with respect to CYP3A4 inhibition of testosterone and methadone metabolism. For both substrates levo-KTZ was approximately a 2-fold more potent inhibitor. We examined the enantioselective differences in the in vitro activity of KTZ against medically relevant species of Candida and Aspergillus, as well as Cryptococcus neoformans. Overall, levo-KTZ was 2-4-fold more active than dextro-KTZ. Therefore, levo-KTZ is a more potent inhibitor of CYP3A4 and has stronger in vitro antifungal activity. Chirality 16:79-85, 2004. 相似文献
