An isolated strain of Bacillus subtilis identified by 16S rDNA sequence analysis produces an enantioselective ester hydrolase. Whole cells of B. subtilis (RRL BB1) and enzyme derived from it was capable of enantioselective hydrolysis of several racemates including drug intermediates with moderate to high enantioselectivity as already reported by us. In this communication, we describe cloning of the gene encoding the enantioselective esterase designated as estBB1. The primary structure of the enzyme determined from the nucleotide sequence indicated that esterase estBB1 has Mw approximately 52kDa and pI approximately 5.2 and belongs to the family of type B carboxylesterases with 50-60% similarity at amino acid level. Alignment studies of sequences of the estBB1 and Pnb esterase 56C8 from B. subtilis showed that estBB1 has an alpha/beta hydrolase fold with catalytic triad formed by Ser190, Glu305 and His394 at active site and Ser190 is located in the conserved motif -G-X-S-X-G-.  相似文献

Summary Variation at a polymorphic Aspartate aminotransferase locus was assayed in a sample of 298 accessions from the ICARDA germplasm collection of the domesticated lentil (Lens culinaris). Two alleles Aat-1 F and Aat-1 S were detected with global frequencies of 0.51 and 0.49, respectively. Fifty-nine percent of accessions were polymorphic for both alleles. The frequency of outcrossing was estimated from the observed heterozygosity to be about 1%. This is higher than direct estimates of outcrossing and implicates selection in favour of heterozygous gene combinations. Significant variation in allele frequency and in the occurrence of polymorphic accessions was observed between countries or geographic areas. Significant associations were observed between the allozymes and agronomic characters. In particular high frequency of Aat-1 F appeared to be associated with late flowering and maturity and low yield.  相似文献

Reduced Axoplasmic Somatostatin Transport in Hypothyroid Rats   总被引:1,自引:1,他引:0  
The effect of hypothyroidism on neuronal function was studied by measuring axoplasmic transport of immunoreactive somatostatin in rat sciatic nerve by the ligation technique. Accumulation of immunoreactive somatostatin proximal to a ligature was linear up to 8 h in normal, in thyroidectomized, and in parathyroidectomized rats. The transport rate was decreased by 38% in thyroidectomized rats as compared to normal rats and was unchanged in parathyroidectomized rats. Sciatic nerve content of somatostatin in hypothyroid rats did not differ from control. Reduced accumulation of immunoreactive somatostatin in hypothyroid rats may be due to a decrease in somatostatin synthesis or in axoplasmic transport, or to an increase in the degradation rate of the peptide.  相似文献

植物种群功能性状及生态对策变异对其自身环境适应能力及群落结构具有重要影响。该文基于崇明东滩湿地芦苇(Phragmites australis)种群内3个小种群不同发育时期分株集合的多项性状指标,分析了其性状及生态对策变异,并基于CSR、LHS生态对策体系,对各小种群的生态对策进行了排序。结果表明, 3个芦苇小种群的21项性状指标及C、R对策得分均存在显著差异。在LHS体系中,以比叶面积(11.5 mm2/mg)、株高(175 cm)和花序干质量(4 g)为分界线,可对3个小种群进行区分。处于生殖生长期的芦苇分株具有更大的植株大小、更高的叶片干物质含量及S对策得分,而处于营养生长期的分株则具有较小的植株大小、较大的比叶面积及R对策得分。总体而言,崇明东滩湿地3个芦苇小种群间及各小种群内处于不同生长发育阶段的分株集合间,都存在显著的性状差异及生态对策差异,且二者的差异模式基本吻合。芦苇种内性状及生态对策变异特征,可为该区芦苇种群的生态保育和恢复实践提供重要的理论参考和技术支撑。  相似文献

A large-surface biosensor technique using surface plasmon resonance (SPR) was tested for protein purification by recovery of a monoclonal antibody against human proinsulin C-peptide. Notably, both reversible attachment/desorption and actual purification of the antibody from a multi-component protein mixture was shown. For initial chip attachment of the peptide ligand, C-peptide was biotinylated and attached to neutravidin on plastic chips with a large gold surface (effective area 26 mm(2)). Antibody binding and desorption was monitored in real-time SPR, and for elution different conditions were employed. Five percent formic acid (in contact with the chip surface for 3 min) in a 60-mul segment between air bubbles was efficient for subsequent analysis. In this manner, protein amounts up to 35 pmoles were recovered in a single capture/elution cycle. Evaluation by SDS-PAGE showed essentially no carryover between fractions in this elution process, and also not with other proteins in the mixture after purification. Compared to existing commercial instruments, this technique gives higher recovery and makes it possible to monitor monitor protein binding/desorption. Recovery of affinity partners at the multi-pmole level is demonstrated for protein purification in SPR approaches.  相似文献

The imidazoline BL11282 stimulates insulin release and alters islet proteomes. Subcellular fractions of MIN6 cells showed that the membrane fraction exhibited binding to BL11282 on a Biacore chip and to BL11282-labelled magnetic beads. Bound material extracted from the beads showed a 50 kDa differential band upon SDS–PAGE and a weaker 100 kDa band. The former was sensitive to competitive removal by preincubation of the fraction with BL11282, then highlighting the 100 kDa band. Masspectrometric analysis revealed the 50 kDa band to be EF1A and the 100 kDa band to be glucose regulated P94, both of interest in insulin synthesis and secretion.  相似文献

The chemical investigation of the ethanolic extract of the root bark of Onosma hispidum following antibacterial activity directed isolation led to the isolation of 4-hydroxy-3-methoxy cinnamic acid (ferulic acid) and 4-hydroxy-3-methoxy benzoic acid (vanillic acid) which have been reported for the first time in this species. In addition to these compounds, the crude ethanolic extract and methanol fraction exhibited substantial bioactivity against species of corynebacteria, enterococci, staphylococci and streptococci. Ferulic acid was found more bioactive (being relatively more hydrophobic) compared to vanillic acid.  相似文献

A new epimeric lupane triterpenoid, 3alpha-hydroxy-20-oxo-30-norlupane (1), and a new flavanone (nubatin; 3) have been isolated from Salvia nubicola collected from Quetta, Pakistan. The structures of these constituents were elucidated with the aid of spectroscopic techniques, including two-dimensional NMR and NOE experiments. In addition to its established role in proinsulin folding, C-peptide has a function in regulation of cellular activity. The 31-residue peptide influences renal, vascular, and metabolic functions in patients with insulin-dependent diabetes mellitus. Binding to cells has been demonstrated for C-peptide, which can be displaced by its C-terminal pentapeptide. We have now used fluorescence correlation spectroscopy to investigate structural requirements on the pentapeptide part for C-peptide binding. All pentapeptide residues, E(27)GSLQ(31), were individually replaced with Ala and the capacity of the resulting peptides to displace rhodamine-labelled full-length human C-peptide from human renal tubular cell membranes was determined. This showed that Glu27 is essential for displacement, while replacement of Gly28 with Ala has little effect, and replacement of any of the three most C-terminal residues had intermediate effects. Morevover, free Glu displaces full-length C-peptide to about 50%, while free Ala, C-peptide(1-26), and the truncated pentapeptide, corresponding to the tetrapeptide G(28)SLG(31), have no displacing capacity. The peptides EVARQ (corresponding to the rat C-terminal pentapeptide) and ELGGGPGAG (corresponding to positions 11-19 of human C-peptide) do not displace human C-peptide. These results indicate that Glu27 of C-peptide is critically involved in binding to cellular targets.  相似文献

Medium-chain dehydrogenases/reductases (MDR) alcohol dehydrogenases exhibit multiple forms through a number of gene duplications. A crucial duplication was the one leading from the glutathione-dependent formaldehyde dehydrogenase line to the liver alcohol dehydrogenase (ADH) lines of vertebrates, the first duplication of which can now be further positioned at early vertebrate times. Similarly, screening of MDR forms in recently completed eukaryotic genomes of Caenorhabditis elegans and Drosophila melanogaster suggest that the MDR family may constitute a moderately sized protein family centered around a limited number of enzyme activities of five different structural types.  相似文献
