Dyslipidemia is one of the major risk factors for cardiovascular diseases (CVDs). Current strategies are not effective in the management of dyslipidemia. Thus, there is a necessity to find new preventative and therapeutic approaches. In recent years, herbal medicine has drawn great attention regarding the prevention and management of dyslipidemia. Rosmarinus officinalis, commonly known as rosemary, is an evergreen shrub containing several polyphenols. The plant grows in the Mediterranean and South American regions. Rosemary and its main components have antioxidant, anti-inflammatory, and lipid-lowering properties. The present review has focused on in vivo and in vitro studies on the hypolipidemic effects of rosemary and its main constituents as well as their functional mechanisms. Studies have described lipid-scavenging activities of rosemary through its flavonoid contents. Modulating inflammation and oxidative stress have been described as possible mechanisms by which rosemary ameliorates dyslipidemia. However, the exact mechanisms are not fully understood yet. Conducting experimental and clinical trial studies are recommended to confirm the safety and efficacy of rosemary in the prevention and management of dyslipidemia and other cardio-metabolic diseases.  相似文献

In this study, hard shell of apricot stones was selected from agricultural solid wastes to prepare effective and low cost adsorbent for the gold separation from gold-plating wastewater. Different adsorption parameters like adsorbent dose, particle size of activated carbon, pH and agitation speed of mixing on the gold adsorption were studied. The results showed that under the optimum operating conditions, more than 98% of gold was adsorbed onto activated carbon after only 3 h. The equilibrium adsorption data were well described by the Freundlich and Langmuir isotherms. Isotherms have been used to obtain thermodynamic parameters. Gold desorption studies were performed with aqueous solution mixture of sodium hydroxide and organic solvents at ambient temperatures. Quantitative recovery of gold ions is possible by this method. As hard shell of apricot stones is a discarded as waste from agricultural and food industries, the prepared activated carbon is expected to be an economical product for gold ion recovery from wastewater.  相似文献

Alteration in the normal regulatory pathway of differentiation can lead to the induction of programmed cell death. Accordingly, some chemicals like staurosporine, nerve growth factor, pituitary adenylate cyclase activating peptide, and trimethyltin are shown to be able to induce differentiation in vitro, via different mechanisms in the PC12 cell line. Hence, understanding the details of the molecular mechanisms of differentiation induction by these small molecules are important for further application of these molecules in neurogenesis. Therefore, we sought to determine these signaling pathways, using gene regulatory networks analysis. Then, we have conducted a comparative analysis of the alterations in the gene expression pattern of the PC12 cell lines in response to these chemicals at the early stages. Based on the comparative analysis and previous knowledge, we have proposed the affected pathways during differentiation and apoptosis. Our findings could be useful in the development of protocols to reprogramming of neurons by such small molecules with high efficiency.  相似文献

The width of the DNA minor groove varies with sequence and can be a major determinant of DNA shape recognition by proteins. For example, the minor groove within the center of the Fis–DNA complex narrows to about half the mean minor groove width of canonical B-form DNA to fit onto the protein surface. G/C base pairs within this segment, which is not contacted by the Fis protein, reduce binding affinities up to 2000-fold over A/T-rich sequences. We show here through multiple X-ray structures and binding properties of Fis–DNA complexes containing base analogs that the 2-amino group on guanine is the primary molecular determinant controlling minor groove widths. Molecular dynamics simulations of free-DNA targets with canonical and modified bases further demonstrate that sequence-dependent narrowing of minor groove widths is modulated almost entirely by the presence of purine 2-amino groups. We also provide evidence that protein-mediated phosphate neutralization facilitates minor groove compression and is particularly important for binding to non-optimally shaped DNA duplexes.  相似文献

Eleven animal taxa were identified as prey of the Deepwater Goby, Chasar bathybius (Kessler, 1877), by analysing stomach contents: these were some species of Gobiidae, Clupeidae, Atherina boyeri, Carassius gibelio, Liza sp., Cardium sp., and Rhithropanopeus sp. With a frequency of 66%, Neogobius fluviatilis was the most prevalent food item, and was the dominant prey taxon in all seasons and age groups. C. bathybius exhibited a specialist feeding strategy on this fish and the other prey types. The dietary breadth is highest in spring according to the Shannon index.  相似文献

Background:

DNA vaccination with plasmid encoding bacterial, viral, and parasitic immunogens has been shown to be an attractive method to induce efficient immune responses. Bacteria of the genus Brucella are facultative intracellular pathogens for which new and efficient vaccines are needed.

Methods:

To evaluate the use of a DNA immunization strategy for protection against brucellosis, a plasmid containing the DNA encoding the Brucella melitensis (B. melitensis) 31 kDa outer membrane protein, as a potent immunogenic target, was constructed.

Results:

The constructed plasmid, pcDNA3.1+omp31, was injected intramuscularly into mice and the expression of omp31 RNA was assessed by RT-PCR. The integrity of the pcDNA3.1+omp31 construct was confirmed with restriction analysis and sequencing. Omp31 mRNA expression was verified by RT-PCR.

Conclusion:

Our results indicate that the pcDNA3.1+omp31 eukaryotic expression vector expresses omp31 mRNA and could be useful as a vaccine candidate.Key Words: Brucella melitensis, omp31, DNA Vaccine, pcDNA3.1  相似文献

Background:

Candida albicans (C. albicans) is a major cause of candidaemia in people with impaired immunity. Blood culture is a “gold standard” for candidaemia detection but is time-consuming and relatively insensitive. We established a real-time PCR assay for C. albicans detection in blood by LightCycler PCR and melting curve analysis.

Methods:

Five milliliter blood samples from healthy volunteers were spiked with 100-106 C. albicans cells to determine the detection limit of our method. DNA was extracted from whole blood using glass beads and the QIAamp DNA Blood Mini Kit (Qiagen, Hilden Germany). DNA from C. albicans isolates were amplified with primers and inserted into Escherichia coli (E. coli) DH5α.1 cells with the TA cloning vector (Invitrogen). The plasmid was used for standardization and optimization. A quantitative PCR assay with the LightCycler amplification and detection system based on fluorescence resonance energy transfer (FRET) with two different specific probes was established. To assess the precision and reproducibility of real-time PCR the intra-assay precision was determined in six consecutive assays.

Results:

No cross-reactivity of the hybridization probes with the DNA of non-C. albicans species or human genomic DNA was observed, which confirmed its 100% specificity. The minimum limit detected was one C. albicans cell or 100 CFU/ml (10 fg) per PCR reaction. The real-time PCR efficiency rate for Candida was high (E = 1.95). Melting curve analysis of C. albicans showed a specific melting peak temperature of 65.76 °C.

Conclusion:

The real-time PCR assay we developed is highly specific and sufficiently sensitive to detect the fungal load for early diagnosis of invasive candidiasis. Key Words: Invasive candidiasis, Real-time PCR, Candida albicans  相似文献

Plasmonic circular dichroism (CD) responses of hybrid nanostructures containing noble metal nanoparticles and chiral molecules have received increasing interest with various applications in nanophotonics. Chiral biomolecules show strong CD signals typically found in the ultraviolet region, whereas, in the visible range, they produce a weak signal. Strengthening the CD signal in the visible region is of high importance, which could be achieved through fabrication of novel hybrid nanostructures. Herein, gold nanoparticles (GNPs) have been assembled via DNA linker to investigate the possibility of enhancing plasmonic CD signal in the visible range. DNA-linked assemblies with pre- and postannealed conditions were characterized by ultraviolet–visible spectroscopy, dynamic light scattering (DLS), and CD spectropolarimetry. In the presence of DNA linker with sticky ends, the aggregation phenomenon was traced by red shifts of surface plasmon resonance of nanoparticles. Time-dependent hybridization of single-stranded “sticky ends” with DNA-conjugated GNPs and increased probability of hydrogen bond formation lead to enhancement of CD signals in the ultraviolet region. Complexation of biomolecule and nanoparticle assemblies induced enhanced CD signals in the visible range, which was noticed both before and after purification. DLS characterization of the assemblies also confirmed the difference in the size of aggregates, which could be controlled by the linker molecules. This investigation encourages possibility of utilizing plasmonic CD technique as a tool for tracing fabricated nanostructure assemblies with enhanced characterization possibility.  相似文献

Earthworms have significant effect on vegetation and soil physical and chemical properties. The relationship between earthworms and biodiversity is important in ecological researches. Therefore, the effects of earthworm on plant diversity and various indices of diversity and richness, as well as physical and chemical properties of soil in shrub, grass and bare vegetation types on exclosure and grazing sites of Miankaleh Mazandaran biosphere reserve is investigated in this work. Accordingly, sampling was carried out randomly-systematically in three different vegetative types (shrubs, grass and bare soil) at 0 to 20 cm depth. Each vegetative types was placed on three transects of 100 m, 10 plots at a distance of 30 m. Earthworm and soil were sampled from the center of each plot of 25 cm × 25 cm. The Simpson and Shannon-Wiener indexes and Margalef and Menhinick richness indices were used to evaluate diversity and richness of species at different grazing intensities. Stepwise regression was used to study the relationship between diversity and richness indices with different earthworm parameters. The results showed that there was significant difference of potassium and nitrogen in vegetation types of both exclosure and grazing sites at P ≤ .01. Furthermore, the organic carbon, organic matter and potassium in the exclosure site and nitrogen and soil moisture in the grazing site are increased by earthworm abundance.  相似文献
