Background

The role of Vitamin D in musculoskeletal functionality among elderly people is still controversial. We investigated the association between serum 25-hydroxyvitamin D (25OHD) levels and physical performance in older adults.

Methods

2694 community-dwelling elderly women and men from the Progetto Veneto Anziani (Pro.V.A.) were included. Physical performances were assessed by: tandem test, 5 timed chair stands (TCS), gait speed, 6-minute walking (6 mW) distance, handgrip strength, and quadriceps strength. For each test, separate general linear models and loess plots were obtained in both genders, in relation to serum 25OHD concentrations, controlling for several potential confounders.

Results

Linear associations with 25OHD levels were observed for TCS, gait speed, 6 mW test and handgrip strength, but not for tandem test and quadriceps strength. After adjusting for potential confounders, linear associations with 25OHD levels were still evident for the 6 mW distance in both genders (p = .0002 in women; <.0001 in men), for TCS in women (p = .004) and for gait speed (p = .0006) and handgrip strength (p = .03) in men. In loess analyses, performance in TCS in women, in gait speed and handgrip strength in men and in 6 mW in both genders, improved with increasing levels of 25OHD, with most of the improvements occurring for 25OHD levels from 20 to 100 nmol/L.

Conclusion

lower 25OHD levels are associated with a worse coordination and weaker strength (TCS) in women, a slower walking time and a lower upper limb strength in men, and a weaker aerobic capacity (6 mW) in both genders. For optimal physical performances, 25OHD concentrations of 100 nmol/L appear to be more advantageous in elderly men and women, and Vitamin D supplementation should be encouraged to maintain their 25OHD levels as high as this threshold.  相似文献

Receptor heteromerization is a mechanism used by G protein-coupled receptors to diversify their properties and function. We previously demonstrated that these interactions occur through salt bridge formation between epitopes of the involved receptors. Recent studies claim that calmodulin (CaM) binds to an Arg-rich epitope located in the amino-terminus of the dopamine D(2) receptor third intracellular loop. This is the same epitope involved in adenosine A(2A)-D(2) receptor heteromerization, through Coulombic interaction between the Arg residues and a phosphorylated serine (pS) located in the medial segment of the C-terminus of the A(2A) receptor. Mass spectrometric analysis indicates that an electrostatic interaction involving the D(2) receptor Arg-rich epitope and several CaM acidic epitopes are mainly responsible for the D(2) receptor-CaM binding. CaM could also form multiple noncovalent complexes by means of electrostatic interactions with an epitope localized in the proximal segment of the C-terminus of the A(2A) receptor. Ca(2+) disrupted the binding of CaM to the D(2) but not to the A(2A) receptor epitope, and CaM disrupted the electrostatic interactions between the D(2) receptor epitope and the more distal A(2A) receptor epitope. A model is introduced with the possible functional implications of A(2A)-D(2)-CaM interactions. These in vitro findings imply a possible regulatory role for CaM in receptor heteromers formation.  相似文献

An Ndh-deficient mutant of tobacco ( Nicotiana tabacum cv. Petit Havana) was prepared by disrupting the ndhF gene in a transplastomic approach. The mutant (Δ ndhF ) showed 10% of the Ndh complex activity (EC 1.6.5.3) and 8% of the NDH-F polypeptide of that of non-transformed plants. However, in Δ ndhF , NDH-A, another Ndh polypeptide, was still present at 50% of the level in non-transformed plants. Δ ndhF tobacco showed higher sensitivity than non-transformed plants to photo-oxidative stress (as judged by chlorophyll bleaching) caused by increased light intensity and paraquat applications. These photo-oxidative treatments increased the amount and activity of the Ndh complex, thylakoid peroxidase, post-illumination chlorophyll fluorescence and non-photochemical quenching (NPQ) of chlorophyll fluorescence in non-transformed but not in Δ ndhF tobacco. Highly stressed non-transformed plants showed a rapid post-rise decline of chlorophyll fluorescence, probably indicating a re-oxidation of reduced plastoquinone. The results indicate that, in normal plants, the Ndh complex and thylakoid peroxidase (EC 1.11.1.7) provide and remove electrons, respectively, to balance the redox level of the intermediates of cyclic electron transport. In this way, they optimize the generation of the transmembrane H+ gradient of thylakoids and, as a consequence, increase the NPQ and the protection against photo-oxidative stress.  相似文献

The temporal and spatial dynamics of phytoplankton have beenstudied in four sites located along the last 60 km of the riverEbro, over a period of 1 year. Diatoms and green algae werethe most abundant groups; blue-green algae were frequent onlyin autumn. Asterionella formosa dominated the winter phytoplanktonassemblages. In autumn, spring and early summer centric diatomswere dominant: Aulacoseira granulata (Ehr.) Simonsen in autunm;Cyclotella sp. p1., Skeletonema potamos (Weber) Hasle and StephanodisciLssp. p1. in spring. A great abundance of green algae was observedduring the summer, mainly in the lower sites. In the sites closerto the mouth, the spring maximum of centric diatoms extendedto the summer. Mainly in the downstream sites, a remarkablegrowth of Acrinocyclus normanii f. subsalsa (Juhl.-Daunf.) Hustedtand Stephanodiscus hantzschii f tenuis (Hust.) Hak. & Stoerm.was added to green algae in the late summer. As has been investigatedthrough a principal component analysis, the phytoplankton temporalsuccession and longitudinal differences between the sites maybe affected by the variations in flow and the increase of waterconductivity downstream; both factors seem to act together.The river is rather homogeneous with respect to the phytoplanktonassemblages during the winter and spring months, and from latespring to the following autumn, differences greatly increaseboth in time and downstream.  相似文献

Genomics and proteomics discovery is leading to the identification of all proteins and to the opportunity, and challenge, to reveal the protein recognition networks that drive virtually all biological processes. Over the past decade, biosensors have emerged as a key technology for detection and analysis of biomolecular interactions. An important limitation in developing such biosensors is that the focus has been mainly on sensor platforms, the transducing hardware that converts interaction signals into recorded data, without adequately considering the role of molecular interfaces, the elements of sensors that interact with analytes to produce signals. We have investigated this alternative focus by identifying and, where necessary, designing molecular interfaces that will more effectively drive new biosensor development and utilization in biomedical and biotechnological investigations. Here we describe our recent studies of coiled coil and lipid bilayer interfaces and the potential to use these to expand sensing technologies for multiplexed target detection and analysis in increasingly biologically relevant membrane like environments.  相似文献

G-protein-coupled receptors form homomers and heteromers; agonist-induced conformational changes within interacting receptors of the oligomer modify their pharmacology, signalling and/or trafficking. When these receptors are activated, the oligomers rearrange and cluster and a novel mechanism of receptor-operation regulation by oligomer intercommunication is possible. This intercommunication would be assisted by components of the plasma membrane and by scaffolding proteins. Receptor cross-sensitization, cross-desensitization and novel, integrated receptor responses can then develop between oligomeric receptor complexes of the cluster without direct contact between them. This concept gives a new perspective to the understanding of neurotransmission and neuronal plasticity.  相似文献

A rapid method for the identification of lactic acid bacteria (LAB) from wine has been developed. This method is based on fluorescence in situ hybridisation (FISH), using fluorescent oligonucleotide probes, homologous to 16S rDNA of those species of LAB commonly found in wines. The protocol for the specific detection of these bacteria was established through the hybridisation of 36 reference strains. The specificity of the probes was evaluated by using pure cultures. Probes were used to identify species in different wines, making it evident that direct identification and quantification from natural samples without culturing is also possible. The results show that FISH is a promising technique for the rapid identification of LAB, allowing positive identification in a few hours (4-16 h).  相似文献

The diatom community growing on cobbles and sand substrata along the Cuarto River (Córdoba, Argentina) was studied during 2000 and 2001. Multivariate analyses of the data (PCA and CCA) showed distinct differences in water chemistry and substrata types between the upstream sites (sites 1–10) and downstream sites (sites 11–19). Sites 1–10 supported an epilithic diatom community associated with low water conductivity and gravel substrata. This consisted of adnate Achnanthes (A. biasolettiana, A. minutissima), as well as stalked (Gomphonema aff. angustum, Reimeria uniseriata, Fragilaria capucina var. rumpens) or prostrate (Nitzschia lacuum) taxa. Downstream sites were associated with high conductivity, fast flowing waters and finer substrata (sand, silt), and were colonised by prostrate diatoms, including several species of Navicula (N. pupula, N. mutica, N. veneta, N. insociabilis) and Nitzschia (N. umbonata, N. palea). Variations in water flow caused significant changes in the diatom communities of the river. During periods of low flow (winter and autumn), chain forming (Diatoma vulgaris, D. moniliformis) or stalked (Synedra ulna) taxa partially replaced the former community of Navicula and Nitzschia in the downstream sites. High flow (in summer) led to diatoms of large size being replaced by smaller size diatoms, such as Achnanthes lanceolata, Navicula mutica, Hantzschia amphioxys and Amphora montana. The severe effect of floods in the lower part of the Cuarto led to these taxa having a much higher proportion in the lower stretch of the river, taking advantage of the subaerial conditions created by the floods. Local episodes of water pollution were associated with a transient shift towards the dominance of Navicula pupula, Synedra ulna, Nitzschia lacuum and Reimeria uniseriata during winter, when inputs were least diluted because of the low flow.  相似文献

Although oxidative stress has been previously described in plants exposed to uranium (U), some uncertainty remains about the role of glutathione and tocopherol availability in the different responsiveness of plants to photo-oxidative damage. Moreover, in most cases, little consideration is given to the role of water transport in shoot heavy metal accumulation. Here, we investigated the effect of uranyl nitrate exposure (50 μM) on PSII and parameters involved in water transport (leaf transpiration and aquaporin gene expression) of Arabidopsis wild type (WT) and mutant plants that are deficient in tocopherol (vte1: null α/γ-tocopherol and vte4: null α-tocopherol) and glutathione biosynthesis (high content: cad1.3 and low content: cad2.1). We show how U exposure induced photosynthetic inhibition that entailed an electron sink/source imbalance that caused PSII photoinhibition in the mutants. The WT was the only line where U did not damage PSII. The increase in energy thermal dissipation observed in all the plants exposed to U did not avoid photo-oxidative damage of mutants. The maintenance of control of glutathione and malondialdehyde contents probed to be target points for the overcoming of photoinhibition in the WT. The relationship between leaf U content and leaf transpiration confirmed the relevance of water transport in heavy metals partitioning and accumulation in leaves, with the consequent implication of susceptibility to oxidative stress.  相似文献
