The tubulin-colchicine complex instead of tubulin was used in an imidazole buffer throughout experiments. The interaction with calcium was examined, especially in the GDP state. The high affinity sites of calcium took part in the polymerization of the complex in the GTP state, while the low ones participated in the depolymerization. The complex had 2 high affinity sites with the dissociation constant of 11.5 x 10(-6) M, and 16 low affinity sites with the dissociation constant of 2.27 x 10(-4) M in the GTP state. In the case of GDP state, the dissociation constant of the high affinity site was 7.2 x 10(-6) M, and the low affinity site was not observed. The ultracentrifugal experiment indicated a little compact structure in the GTP state compared with the GDP state. This agreed with the results of calcium binding. 相似文献

A key feature of signal processing in the mammalian retina is parallel processing, where the segregation of visual information, e.g., brightness, darkness, and color, starts at the first synapse in the retina, the photoreceptor synapse. These various aspects are transmitted in parallel from the input neurons of the retina, the photoreceptor cells, through the interconnecting bipolar cells, to the output neurons, the ganglion cells. The photoreceptors and bipolar cells release a single excitatory neurotransmitter, glutamate, at their synapses. This parsimony is contrasted by the expression of a plethora of glutamate receptors, receptor subunits, and isoforms. The detailed knowledge of the synaptic distribution of glutamate receptors thus is of major importance in understanding the mechanisms of retinal signal processing. Survival ability of Maia squinado to emersion and subsequent reimmersion was determined in winter and summer conditions. Male spider crabs were less tolerant of emersion than females. Emersion (up to 24 h in summer and to 48 h in winter) induced a marked reduction of nitrogen excretion, especially ammonia excretion. Increase in blood ammonia content was rapid and very high in summer (1750 micromol l(-1)), but non-lethal levels. Estimation of the body ammonia overload showed that only 30% of unexcreted ammonia accumulated in blood. The ammonia release at reimmersion indicated that ammonia also accumulated in other body compartments. Increase in blood urate content, which indirectly reduces ammonia production, was similar at both seasons. Emersed M. squinado was rapidly resorting to anaerobic metabolism, especially in summer when its blood haemocyanin content is low. A strong hyperglycemia was developed in the first 12 h of emersion at both seasons. Mortality occurring beyond 24 h of reimmersion, when the body ammonia overload is cancelled and the recovery of most of blood components is achieved, remains unexplained. 相似文献

1. From 1993 to 1995, we studied the summer denning ecology of 40 radio-collared striped skunks Mephitis mephitis (32 females, eight males) during the waterfowl nesting season in Saskatchewan, Canada.
2. Forty-seven natal dens were used by 30 females. Natal den sites were either underground burrows (55%) or holes underneath buildings (45%), and were preferably located in farmsteads.
3. Male and female striped skunks used similar resting sites which consisted of above-ground retreats (57%), underground burrows (23%), buildings (17%) and culverts (3%).
4. Habitat preferences for resting sites did not differ between males and females: both sexes preferred farmsteads and wetlands, whereas managed waterfowl nesting areas, woodland and miscellanous habitats were avoided. Cropland was avoided for both natal dens and resting sites.
5. Females did not simultaneously share natal dens or resting sites, but both types of retreats were occupied by different females over time. Natal dens were occupied for 47 ± 3 days for females using a single den vs. 60 ± 3 days for females using >1 natal dens. Den switching may have been induced by a build-up of faeces.

The Anion Cl−/HCO3− Exchangers AE1, AE2, and AE3 are membrane pH regulatory ion transporters ubiquitously expressed in vertebrate tissues. Besides relieving intracellular alkaline and CO2 loads, the AEs have an important function during development and cell death and play a central role in such cellular properties as cell shape, metabolism, and contractility. The activity of AE(s) are regulated by neurohormones. However, little is known as to the intracellular signal transduction pathways that underlie this modulation. We show here that, in cardiomyocytes that express both AE1 and AE3, the purinergic agonist, ATP, triggers activation of anion exchange. The AE activation is observed in cells in which AE3 expression was blocked but not in cells microinjected with neutralizing anti-AE1 antibodies. ATP induces tyrosine phosphorylation of AE1, activation of the tyrosine kinase Fyn, and association of both Fyn and FAK with AE1. Inhibition of Src family kinases in vivo by genistein, herbimycin A, or ST638 prevents purinergic activation of AE1. Microinjection of either anti-Cst.1 antibody or recombinant CSK, both of which prevent activation of Src family kinase, significantly decreases ATP-induced activation of AE. Microinjection of an anti-FAK antibody as well as expression in cardiomyocytes of Phe397 FAK dominant negative mutant, also prevents purinergic activation of AE. Therefore, tyrosine kinases play a key role in acute regulation of intracellular pH and thus in cell function including excitation–contraction coupling of the myocardium. 相似文献

Microcosm tests simulating bioslurry reactors with 40% soilcontent, containing high concentrations of TNT and/or RDX,and spiked with either [14C]-TNT or[14C]-RDX were conducted to investigate the fate ofexplosives and their metabolites in bioslurry treatment processes.RDX is recalcitrant to indigenous microorganisms in soil andactivated sludge under aerobic conditions. However, soilindigenous microorganisms alonewere able to mineralize 15% of RDX to CO2 underanaerobic condition, and supplementation of municipal anaerobicsludge as an exogenous source of microorganismssignificantly enhanced the RDX mineralization to 60%. RDXmineralizing activity of microorganisms in soil and sludge wassignificantly inhibited by the presence of TNT. TNTmineralization was poor (< 2%) and was not markedlyimproved by the supplement ofaerobic or anaerobic sludge. Partitioning studies of[14C]-TNT in the microcosmsrevealed that the removal of TNTduring the bioslurry process was due mainly to thetransformation of TNT and irreversiblebinding of TNT metabolites onto soil matrix. Abstract. The subalpine oligotrophic heath-grasslands, assigned to the Pulsatillo-Vaccinietum and Violo-Nardetum, occurring at the upper ridges in the Vosges at 1100 - 1300 m a.s.l. are of conservational interest because they form natural, mature and stable ecosystems. The Pulsatillo-Vaccinietum is particularly rich in endemic plant species and subspecies due to a local oceanic-subarctic climate and the continuous occurrence of the community throughout the Holocene period, when the association was not affected by changes in the prevailing environmental conditions. After a period of abandonment during and after World War II, agriculture was intensified through mineral fertilization and liming, sometimes mowing, and even ploughing and sowing. Seven pastures were selected to estimate the present plant biodiversity. Losses of biodiversity were assessed on the Kastelberg ridge by diachronic analysis of the vegetation composition and structure over the last 30 years. Species diversity changed in both associations through a decrease in the abundance of oligotrophic, acidophilic species, typical of the mountains of western Europe, and an extension of grassland types of the Violo-Nardetum trifolietosum. Strategies for a return to more natural conditions include the abandonment of fertilization and liming, except locally around farms. Regular mowing is needed in order to decrease the nutrient level of the sites. 相似文献

Most treeline populations in northeastern Canada are monospecific stands of black spruce (Picea mariana [Mill.] B.S.P.), a hardy, cold-tolerant species able to withstand harsh climatic conditions under different growth forms. In the forest tundra, black spruce thrives in protected areas and exhibits a normal arborescent growth form, but in exposed sites, upright stems are damaged above the snowpack by snow abrasion and wind. In this study, the development of damaged growth forms was examined in a moderately exposed habitat. Five developmental stages were identified and described using detailed stem analysis of 13 spruce trees. Four different types of damaged growth forms were identified according to variations in supra-nival (above snow) stem height and number. At the site scale, the age structure of supra-nival shoots, based on a larger sample of 256 stems, was unimodal, suggesting a synchronous development of the spruce stand in which 46% of the shoots were initiated during the 1960s and 1970s. Subfossil trunks on the ground were all depressed trees, indicating that the former vegetation was a krummholz, not a forest. This indicates the recent development of the small-tree stand above the snowpack, probably triggered by recent milder conditions associated with snowier winters in the last decades. 相似文献
