The in vitro virulence properties of 197 temporally and geographically related Campylobacter isolates from chicken broilers and humans were compared. Comparisons of the virulence properties associated with genotypes and biotypes were made. All isolates adhered to, and 63% invaded, INT-407 cells, whereas 13% were cytotoxic for CHO cells. CHO cell-cytotoxic extracts were also cytotoxic for INT-407 cells, but the sensitivity for Vero cells was variable. The proportion of isolates demonstrating a high invasiveness potential (>1,000 CFU ml−1) or Vero cell cytotoxicity was significantly higher for human than for poultry isolates. Invasiveness was associated with Campylobacter jejuni isolates of biotypes 1 and 2, whereas CHO and INT-407 cell cytotoxicity was associated with C. jejuni isolates of biotypes 3 and 4. Cytotoxic isolates were also clustered according to pulsed-field gel electrophoresis profiles.  相似文献

A laboratory-scale permeable biobarrier exhibited high removal efficiencies of benzene at inlet concentrations of 0.4 to 35.1?mg/L and with a limited supply of dissolved oxygen. The supplied oxygen was less than the demand for a complete aerobic oxidation of benzene. Stainless steel pieces or granulated peat moss were used as packing material for microbial support in the biobarrier. Removal efficiencies ranged from 63.9% to 99.9% in the stainless steel-packed biobarrier and from 70.4% to 97.2% in the peat moss-packed biobarrier, while benzene elimination rate changed from 0.2 to 10.4?mg/L-d and from 0.1 to 3.7?mg/L-d in the two biobarriers, respectively. The consumption of sulfate and the presence of sulfate-reducing bacteria suggested the contribution of anaerobic metabolism in the biodegradation of benzene. The biodegradation of benzene under microaerophilic conditions (defined as dissolved oxygen concentrations <2?mg/L) was demonstrated during independent batch experiments. The maximum specific rate of benzene biodegradation with concentrations of 22.0 to 65.9?mg/L under microaero-philic conditions was 2.6 mg/mg biomass-d.  相似文献

Aim The predictions from biogeographical models of poleward expansion of biomes under a warmer 2 × CO2 scenario might not be warranted, given the non‐climatic influences on vegetation dynamics. Milder climatic conditions have occurred in northern Québec, Canada, in the 20th century. The purpose of this study was to document the early signs of a northward expansion of the boreal forest into the subarctic forest‐tundra, a vast heterogeneous ecotone. Colonization of upland tundra sites by black spruce (Picea mariana (Mill.) BSP.) forming local subarctic tree lines was quantified at the biome scale. Because it was previously shown that the regenerative potential of spruce is reduced with increasing latitude, we predicted that tree line advances and recent establishment of seedlings above tree lines will also decrease northwards. Location Black spruce regeneration patterns were surveyed across a > 300‐km latitudinal transect spanning the forest‐tundra of northern Québec, Canada (55°29′–58°27′ N). Methods Elevational transects were positioned at forest–tundra interfaces in two regions from the southern forest‐tundra and two regions from the northern forest‐tundra, including the arctic tree line. The surroundings of stunted black spruce, forming the species limit in the shrub tundra, were also examined. Position, total height and origin (seed or layer) of all black spruce stems established in the elevational transects were determined. Dendrochronological and topographical data allowed recent subarctic tree line advances to be estimated. Age structures of spruce recently established from seed (< 2.5 m high) were constructed and compared between forest‐tundra regions. Five to 20‐year heat sum (growing degree‐days, > 5 °C) and precipitation fluctuations were computed from regional climatic data, and compared with seedling recruitment patterns. Results During the 20th century, all tree lines from the southern forest‐tundra rose slightly through establishment of seed‐origin spruce, while some tree lines in the northern forest‐tundra rose through height growth of stunted spruce already established on the tundra hilltops. However, the rate of rise in tree lines did not slow down with latitude. The density of < 2.5‐m spruce established by seed declined exponentially with latitude. While the majority of < 2.5‐m spruce has established since the late 1970s on the southernmost tundra hilltops, the regeneration pool was mainly composed of old, suppressed individuals in the northern forest‐tundra. Spruce age generally decreased with increasing elevation in the southern forest‐tundra stands, therefore indicating current colonization of tundra hilltops. Although spruce reproductive success has improved over the twentieth century in the southern forest‐tundra, there was hardly any evidence that recruitment of seed‐origin spruce was controlled by 5‐ to 20‐year regional climatic fluctuations, except for winter precipitation. Main conclusions Besides the milder 20th century climate, local topographic factors appear to have influenced the rise in tree lines and recent establishment by seed. The effect of black spruce's semi‐serotinous cones in trapping seeds and the difficulty of establishment on exposed, drought‐prone tundra vegetation are some factors likely to explain the scarcity of significant correlations between tree establishment and climatic variables in the short term. The age data suggest impending reforestation of the southernmost tundra sites, although the development of spruce seedlings into forest might be slowed down by the harsh wind‐exposure conditions.  相似文献

Animals adapted to high latitude environments typically live at relatively low densities and require large areas to acquire the requisites for successful reproduction. Also, the unpredictable distribution of energy across time and space, such as environmental seasonality, constrains the timing of mating and parturition and imposes limitations to reproductive strategies. For example, limited mobility due to snow accumulation may reduce mate encounters, and this selective pressure may have resulted in morphological adaptations to assess mate quality during serial encounters. We tested whether mammalian carnivores living in high latitude environments displayed greater use of copulatory competition via length of the penis bone or baculum, and whether females in such settings enhance competition among males via multi-male matings. We then examined how baculum length may correlate with other reproductive adaptations in high latitude environments such as delayed implantation and induced ovulation. Statistical methods that account for phylogeny revealed that longer bacula occur in carnivores living in high latitude environments with greater snow accumulation, and that larger bacula is also associated with delayed implantation and multi-male mating systems. In contrast, the smallest bacula were observed in polygynous species. Our results provide an example of the effects of ecology on the evolution of a secondary sexual character due to sexual selection. Selection pressures imposed by seasonality of resources and environmental conditions affecting locomotion may have led to sexual selection via mating systems for females, and counter morphological adaptations of long penis bones in males.  相似文献

FMOC-L-Leucine (F-L-Leu) is a chemically distinct PPARgamma ligand. Two molecules of F-L-Leu bind to the ligand binding domain of a single PPARgamma molecule, making its mode of receptor interaction distinct from that of other nuclear receptor ligands. F-L-Leu induces a particular allosteric configuration of PPARgamma, resulting in differential cofactor recruitment and translating in distinct pharmacological properties. F-L-Leu activates PPARgamma with a lower potency, but a similar maximal efficacy, than rosiglitazone. The particular PPARgamma configuration induced by F-L-Leu leads to a modified pattern of target gene activation. F-L-Leu improves insulin sensitivity in normal, diet-induced glucose-intolerant, and in diabetic db/db mice, yet it has a lower adipogenic activity. These biological effects suggest that F-L-Leu is a selective PPARgamma modulator that activates some (insulin sensitization), but not all (adipogenesis), PPARgamma-signaling pathways.  相似文献

The aim of the present study was to precise the origin of the particular muscle characteristics of double-muscled cattle by comparing muscle properties of Holstein and double-muscled Belgian Blue (BB) foetuses. Ten 100-day-old foetuses of each genotype were studied. The weight and length of foetuses and the length, weight and area of the Semitendinosus (ST) muscle were analysed. Contractile differentiation of the different fibre types was studied by immunohistochemistry using several monoclonal antibodies raised against different myosin heavy chain isoforms (MHC slow, fast, foetal) and by electrophoresis. Proliferation phase of myoblasts from each genotype was analysed in primary culture. On 100 days of foetal life, the foetuses of both genotypes did not show any significant differences in their weight and length. However, BB cattle already present muscle hypertrophy, which seems to originate from a higher myoblast proliferation observed in primary culture. The use of anti-MHC antibodies shows that ST muscle of BB contained a smaller proportion of primary fibres and a higher proportion of secondary fibres which will give principally fast fibres in adult muscle. Electrophoresis analysis confirms a lower proportion of slow MHC in ST of BB.  相似文献

We studied the seed dynamics of Abies balsamea and Acer saccharum in a sugar maple-yellow birch forest of Québec, Canada. Seed rain was censused every week from spring to autumn in 1988 and 1989. The soil seed bank was sampled four times during the 1988 growing season. Abies seed rain varied significantly between 1988 and 1989. A total of 92.5 ± 7.0 seeds nr-2 (mean ± 1 SE) were produced in 1988; 1989 seed production was null. Seed viability in 1988 averaged 31.5%. Dispersal during the winter period accounted for ca. 22% of the annual seed crop. The spatial dispersion pattern of the seed rain was contagious for the autumn period, but was not significantly different from random for the winter period. There was only a weak relationship in the spatial distribution of the seed rain between the autumn and winter periods. Abies did not maintain a persistent seed bank on the site; viable seeds were present on the soil only after seed dispersal had started. Postdispersal seed mortality was relatively high, ca. 70%. Acer seed rain started soon after the flowering period, but consisted at that time only of aborted, underdeveloped samaras. Viable seeds were disseminated from September through early winter. Dispersal during the autumn accounted for ca. 98% of the annual seed crop. There were significant differences between seed rain abundance of 1988 and that of 1989 (118.1 ± 9.5 samaras m-2 and 158.9 ± 16.3 samaras m-2 in 1988 and in 1989, respectively). Seed viability also varied significantly between the 2 years, i.e., 19% in 1988 and 5% in 1989. For both 1988 and 1989, samaras were contagiously dispersed over the site. There was a significant positive relationship between the spatial distribution of the seed rain in 1988 and that in 1989. Seed rain abundance was high mainly in the proximity of mature Acer trees. As for Abies, Acer did not maintain a persistent seed bank in the soil; viable seeds were present only in the autumn seed bank sampling, after seed fall had started. Postdispersal seed mortality was relatively low, i.e., 20%. Abies and Acer are quite similar in their regenerative traits, and these contrast sharply with those of Betula alleghaniensis, a regular member of this forest community. Differences in regenerative traits may contribute to the coexistence of these tree species considering the gap regime of the system studied.  相似文献

Aim Aspects of connectivity and continuity operating in the River Meuse were analysed for their contribution to the biodiversity of the floodplain. From this analysis of the diversity and composition of the meadow communities, we aimed to derive effective biodiversity conservation strategies. Location The River Meuse is one of the larger rivers in the European Western Plains ecoregion. The alluvial plains of the river have a long history of cultivation, and for these plains the floodplain meadow vegetation is a highly appreciated and valuable nature conservation asset. Method We sampled floodplain meadows from 400 km of the six geomorphic reaches of the middle to lower course of the River Meuse. For each, 50 vascular plant relevés were recorded, representing the spectrum of floodplain meadow communities of that reach. Beta diversity was calculated to quantify similarity in species pools between the reaches. A dissimilarity formula was used to determine the turnover between the reaches, and these dissimilarities were compared with a Mantel test to detect whether species composition of the floodplain meadows exhibited connectivity and continuity between the reaches. Species richness for the floodplain vegetation data of the reaches was compared with data for riparian invertebrate communities. The vegetation data sets were ordinated using detrended correspondence analysis (DCA) to reveal patterns in the floodplain meadow species composition, and the DCA axes were related to plant functional groups and population strategies. The axis scores of the species and plots were linked to river and plant species traits. Results We did not observe an overall continuity trend in similarity, nor one in diversity in a downstream direction. Lateral connectivity was highlighted by the dissimilarity between the reaches and in the influxes of species from adjacent ecoregions. The DCA ordination showed statistically significant separations between reaches and between the plant functional groups. The second DCA axis related to the longitudinal gradient of the river, whereas the first axis showed stronger correlations with river and plant species traits. We termed this axis the ‘disorder axis’. Plant invaders and avoiders are located at the left side of the disorder axis, whereas the true river‐adapted categories of resisters and endurers are at the right extremity. Contributions to the disorder were identified in terms of connectivity with adjacent ecoregions and the physical disturbance regime of natural and anthropogenic perturbations, resulting in community changes between the reaches. Main conclusions We concluded that a single overall strategy for biodiversity conservation of the river or its floodplains is not feasible. Strategies can, however, be derived for separate river reaches based on functional groups in the communities, the disorder characteristics of the reach, and the influence of surrounding ecoregions.  相似文献

Premature death has been defined as a growth stoppage linked to the accumulation of specific deletions of the mitochondrial genome (mtDNA) in Podospora anserina. This occurs only in strains carrying the AS1-4 mutation which lies in a gene encoding a cytosolic ribosomal protein. Here we describe the isolation and genetic characterization of 10 nuclear mutations which either delay the appearance of this syndrome (respite from premature death) or cause a switch to the classical senescence process (repeal of premature death). These mutations lie in at least six genes. Some cause defects at the levels of ascospore germination, growth rates, and/or sensitivity toward inhibitors of protein syntheses. All modify the onset of senescence in wild-type (AS1+) strains. The role played by these genes is discussed with respect to the control of diseases due to mtDNA rearrangements in filamentous fungi.
