Summary 2-methyl-oxazolinones and thio-thiazolidones representative of aromatic and aliphatic amino acids were hydrolysed by -chymotrypsin and subtilisin. The parametric ratio kcat/Km, correlated with the enantiomeric enrichment of the reaction product, indicates that thio-thiazolidones are converted to free amino acids by enzyme with the higher degree of stereospecificity.  相似文献

The tritiated 1 antagonist prazosin [3H]PRZ binds specifically and with high affinity to postsynaptic adrenoceptors in membrane preparations from cerebral cortex. Since adrenoceptors are of protein nature, it was of interest of investigate the possible role of disulfide (—SS—) and sulfhydril (—SH) groups in the binding of [3H]PRZ. Pretreatment of the membranes with the disulfide and sulfhydryl reactivesdl0Dithiothreitol,l-Dithiothreitol, Dithioerythritol or 5,5-Dithiobis-(2-nitrobenzoic acid) (DTNB), alone or in combination with the alkylating agent N-Methylmaleimide (NMM), decreased specific [3HPRZ binding, with minor changes in the non-specific counts. Saturation experiments revealed that all these reagents reduced the affinity of the binding site for [3H]PRZ, as judged by theK d 25°C, but only the alkylating agent NMM and the oxydizing reagent DTNB produced in addition to the increase inK d, a decrease of the maximum binding capacity (B max). The present results provide evidence for a participation of—SS—and/or—SH groups in the recognition site of the 1-adrenoceptor of cerebral cortex.  相似文献

5-methoxypsoralen (5-MOP) binds to human serum low density lipoproteins (LDL) according to a two-step process. Scatchard analysis of the first step yields K = 1.4 × 105 M?1 and 4 binding sites. It involves the LDL apoprotein. The second step corresponds to a solubilization, in the lipidic core, of ? 45 molecules of 5MOP per LDL molecule. It is accompanied by a large blue shift of the 5MOP fluorescence. The ability of LDL to bind 5MOP and to carry it into various cells may explain some biological effects sometimes encountered during PUVA therapy.  相似文献

The daily rhythm of the adrenocortical cyclic nucleotides (cyclic AMP and cyclic GIMP) was studied in infant male and female Wistar rats before and after the establishment of an adult-like daily rhythm of plasma corticosterone. As in this strain the rhythm of corticosterone is known to be present on postnatal day 18, pups of 2 and 3 weeks of age were studied. The dams and the pups as well as the young adult animals were kept on a controlled 12L-12D photoperiod. Groups of 8-10 pups were killed at 4-hr intervals throughout the day. Plasma corticosterone levels and adrenal cyclic AMP and cyclic GMP concentrations were simultaneously measured and the daily patterns established. Pups of 2 weeks of age showed neither plasma corticosterone nor adrenal cyclic AMP rhythms whereas pups of 3 weeks of age exhibited a typical adult-like circadian rhythm for both variables. The patterns for adrenal cyclic GMP differed according to sex: In female pups no cyclic GMP circadian rhythm could be detected at either 2 or 3 wk. In male pups of 3 wk a typical mature rhythm for adrenal cyclic GMP was evident whereas in younger male pups (2 wk) a circadian rhythm was detected. This circadian rhythm, however, differed from mature circadian rhythm in that its peak was located at 1300 hr instead of 0700 hr. These results demonstrate that, unlike that of cyclic AMP the adrenal cyclic GMP circadian rhythm does not appear at the same time as the plasma corticosterone circadian rhythm. Moreover, a circadian rhythmicity for adrenal cyclic GMP can be found in the absence of any corticosterone circadian rhythm. These facts argue against the view of cyclic GMP being a mediator of ACTH-stimulated steroidogenesis.  相似文献

Probes carrying the Anabaena sp. strain PCC 7120 nitrogenase reductase (nifH) and nitrogenase (nifK and nifD) genes were hybridized to Southern blots of DNA from the unicellular, aerobic nitrogen-fixing cyanobacterium Gloeothece sp. strain PCC 6909 and from the filamentous cyanobacterium Calothrix sp. strain PCC 7601. These data suggest that the Gloeothece sp. nif structural proteins must be similar to those of other diazotrophs and that the ability for aerobic nitrogen fixation does not reside in the nif protein complex. We also found that the nif structural genes of Gloeothece sp. are clustered, whereas those of Calothrix sp. are arranged more like those of Anabaena sp.  相似文献

The growth requirements of inorganic and organic selenium (Se) for the dinoflagellate Peridinium cinctum fa. westii and the diatom Stephanodiscus hantzschii var. pusillus and eight species of green algae are demonstrated. A new mineral culture medium for P. cinctum is presented. Using P. cinctum as a test alga, bioassays were carried out on waters from a calciferous lake and from some acidified, fertilized, lime-enriched or humic lakes for determination of their contents of bioavailable forms of Se (bioactive Se). Some of the results were related to Tot-Se and selenite-Se, measured chemically, to pH and the occurrence of ambient phytoplankton. In calcium-rich Lake Erken (pH ~ 8) blooms and decline of algae coincided with decreases (from >70 to ~20 ng Se · l?1) and increases (up to >80 ng · l?1) of bioactive Se, indicating uptake and release of inorganic or organic forms of Se. Acidified lakes (pH < 5) generally demonstrated much lower concentrations of bioactive Se (<20 ng · l?1) than neutral, fertilized, lime-enriched, slightly acidic or humic lakes (>20 ng · l?1). The correlation between pH and bioactive Se was positive (r ~ 0.92; n = 7). The ecological importance of Se is suggested for some common species of phytoplankton.  相似文献

In the fungus Podospora anserina, the interaction between the nonallelic incompatible R and V genes has two consequences: a lytic reaction due to the synthesis of specific proteolytic enzymes, and a quenching in protein and ribonucleic acid synthesis. The incompatibility reaction when vegetative or sexual R and V cells fuse is asymmetric: it is induced only in the R protoplasm. The cessation in ribonucleic acid and protein synthesis was investigated in heterokaryotic strains carrying the antagonistic R and V genes and their "neutral" r and v alleles. Asymmetry between R and V genes lies in the fact that the strains homozygous for the R genes are the only strains that cannot grow. From these results it is postulated that the V-gene product is a diffusible cytoplasmic factor and that the R-gene product, which is nonautonomous, is a ribosomal component.  相似文献
