A total of 108 samples of pickles, which were produced in districts with high and low incidences of stomach cancer in Japan, were extracted with methanol-chloroform. The extracts were bioassayed with Salmonella tester strains. The pickles produced in the high-cancer-incidence district were more mutagenic than those produced in the low-incidence district. The most mutagenic sample among 24 pickle specimens collected in the high-incidence district induced 130 revertants/mg of the crude extract for strain TA98. The mutagenic compounds were purified, and 2 flavonols, quercetin and rhamnetin, were identified as the major mutagens in the pickles by gas chromatography-mass spectrometry. The quantities of the 2 compounds were determined as 6.60 mg for quercetin, and 1.96 mg for rhamnetin per gram of the crude extract. The mutagenic activities of the pickles produced in the 2 districts were closely related to the amounts of quercetin in them.  相似文献

Environmental virus surveillance was conducted at two independent sewage plants from urban and rural areas in the northern prefecture of the Kyushu district, Japan, to trace polioviruses (PVs) within communities. Consequently, 83 PVs were isolated over a 34-month period from April 2010 to January 2013. The frequency of PV isolation at the urban plant was 1.5 times higher than that at the rural plant. Molecular sequence analysis of the viral VP1 gene identified all three serotypes among the PV isolates, with the most prevalent serotype being type 2 (46%). Nearly all poliovirus isolates exhibited more than one nucleotide mutation from the Sabin vaccine strains. During this study, inactivated poliovirus vaccine (IPV) was introduced for routine immunization on 1 September 2012, replacing the live oral poliovirus vaccine (OPV). Interestingly, the frequency of PV isolation from sewage waters declined before OPV cessation at both sites. Our study highlights the importance of environmental surveillance for the detection of the excretion of PVs from an OPV-immunized population in a highly sensitive manner, during the OPV-to-IPV transition period.  相似文献

The possible mechanism of wheeze generation in tracheostenosis was identified by measuring inspiratory and expiratory flow in a "morphological and distensible" realistic tracheostenosis model. The shape of the model was based on CT (Computed Tomography) images of a patient that had tracheostenosis. A trachea consists of tracheal cartilage rings and smooth muscle. Spatial variation of wall distensibility was achieved in the model by varying the wall thickness based on the elastic modulus measured in pig airways. The spatial variation influenced the flow in the airway and the turbulence production rate decreased faster at smooth muscles. Using the model, we investigated the mechanism of wheeze generation by focusing on the turbulence intensity. The turbulence intensity in expiratory flow was about twice that in inspiratory flow, and larger vortices existed in post-stenosis in expiratory flow, and thus might contribute to wheeze generation.  相似文献

Vanadate increased lipoprotein lipase (LPL) activity in the isolated fat pads in a time- and dose-dependent manner. The increasing effect of vanadate was inhibited by amiloride, similar to that of insulin, and it also was not additive to that of insulin. Although the increasing effects of vanadate and insulin were preserved in K(+)-free medium, appreciable decreases in both effects were observed by replacement of Na+ with choline ion or omission of Ca2+ in the medium. Vanadate showed the full effect in the presence of cycloheximide at concentrations that inhibited protein synthesis of the fat pads, suggesting that the action of vanadate is not due to the increase in protein synthesis. Tetrakis (acetoxymethyl) ester of quin 2 at 50 microM concentration never inhibited the action of vanadate though it showed a little inhibition at a concentration of 300 microM. No inhibition of the action of vanadate was observed with ruthenium red. These results suggest that vanadate increases the LPL activity via a process less sensitive to the intracellular Ca2+ concentration. Adrenaline, dibutyryl cyclic AMP, and 3-isobutyl-1-methylxanthine all inhibited the action of vanadate, suggesting that the action is inhibited with increase in the intracellular concentration of cyclic AMP. Monensin and carbonyl cyanide m-chlorophenylhydrazone inhibited the action of vanadate. In contrast, the action of insulin was never inhibited by monensin. Tunicamycin and 2-deoxyglucose, at rather high concentrations, inhibited both actions. These findings suggest that vanadate increases the LPL activity through mechanisms of action involving amiloride- and monensin-sensitive pathways dependent on energy.  相似文献

Rhizomes of Curcuma phaeocaulis Valeton (Zingiberaceae) have traditionally been used for controlling inflammatory conditions. Numerous studies have aimed to isolate and characterize the bioactive constituents of C. phaeocaulis. It has been reported that its anti‐inflammatory properties are a result of cyclooxygenase‐2 inhibition; however, its effect on the T‐cell function remains to be elucidated. In this study, four known sesquiterpenoids, viz., ar‐turmerone (TM), germacrone (GM), (+)‐(4S,5S)‐germacrone‐4,5‐epoxide (GE), and curzerenone (CZ), were isolated from C. phaeocaulis rhizomes and evaluated for their effects on the CD4+ T‐cell function. While GM, GE, and CZ had no effect on the activation of splenic T cells or CD4+ T cells, TM suppressed the interferon (IFN)‐γ production, without affecting the interleukin (IL)‐4 expression. TM also decreased the expression of IL‐2 in CD4+ T cells, but did not change their cell‐division rates upon stimulation. These results suggest that TM, a major constituent of C. phaeocaulis rhizomes selectively exerts potent anti‐inflammatory effects via suppression of the inflammatory cytokines IFN‐γ and IL‐2.  相似文献

To enhance DNA cleavage by zinc-finger nucleases (ZFNs), we sandwiched a DNA cleavage enzyme with two artificial zinc-finger proteins (AZPs). Because the DNA between the two AZP-binding sites is cleaved, the AZP-sandwiched nuclease is expected to bind preferentially to a DNA substrate rather than to cleavage products and thereby cleave it with multiple turnovers. To demonstrate the concept, we sandwiched a staphylococcal nuclease (SNase), which cleaves DNA as a monomer, between two three-finger AZPs. The AZP-sandwiched SNase cleaved large amounts of dsDNA site-specifically. Such multiple-turnover cleavage was not observed with nucleases that possess a single AZP. Thus, AZP-sandwiched nucleases will further refine ZFN technology.  相似文献
