In the present study, the biosynthesis of silver nanoparticles (AgNPs) using Neurospora intermedia, as a new non-pathogenic fungus was investigated. For determination of biomass harvesting time, the effect of fungal incubation period on nanoparticle formation was investigated using UV–visible spectroscopy. Then, AgNPs were synthesized using both culture supernatant and cell-free filtrate of the fungus. Two different volume ratios (1:100 and 1:1) of the culture supernatant to the silver nitrate were employed for AgNP synthesis. In addition, cell-free filtrate and silver nitrate were mixed in presence and absence of light. Smallest average size and highest productivity were obtained when using equal volumes of the culture supernatant and silver nitrate solution as confirmed by UV–visible spectra of colloidal AgNPs. Comparing the UV–visible spectra revealed that using cell-free filtrate for AgNP synthesis resulted in the formation of particles with higher stability and monodispersity than using culture supernatant. The absence of light in cell-free filtrate mediated synthesis led to the formation of nanoparticles with the lowest rate and the highest monodispersity. The presence of elemental silver in all prepared samples was confirmed using EDX, while the crystalline nature of synthesized particles was verified by XRD. FTIR results showed the presence of functional groups which reduce Ag+ and stabilize AgNPs. The presence of nitrate reductase was confirmed in the cell-free filtrate of the fungus suggesting the potential role of this enzyme in AgNP synthesis. Synthesized particles showed significant antibacterial activity against E. coli as confirmed by examining the growth curve of bacterial cells exposed to AgNPs. 相似文献

Nepeta pogonosperma is an important medicinal plant with anti-inflammatory effects. An efficient and reliable transformation system for this plant was developed through optimization of several factors which affected the rate of Agrobacterium rhizogenes mediated transformation. Five bacterial strains, A4, ATCC15834, LBA9402, MSU440 and A13, two explant types, leaves and stems, and several co-cultivation media were examined. The maximum rate of hairy root induction was obtained from stem explants using MSU440 and ATCC15834 bacterial strains. A drastic increase in the frequency of transformation (91 %) was observed when MS medium lacking NH4NO3, KH2PO4, KNO3 and CaCl2. Hairy root lines were confirmed by polymerase chain reaction (PCR) using primers of the rolB gene. According to Southern blot analysis, one T-DNA copy was inserted into each of the hairy root lines. In the present study, transgenic hairy roots have been obtained trough genetic transformation by A. rhizogenes harbouring two plasmids, the Ri plasmid and pBI121 binary vector harbouring gus reporter gene. Expression of the gus gene in transgenic hairy root was confirmed by histochemical GUS assay. 相似文献

Background
It has been hypothesized in the literature that exposure to extremely low frequency electromagnetic fields (50 or 60 Hz) may lead to human health effects such as childhood leukemia or brain tumors. In a previous study investigating multiple types of cells from brain and kidney of the mouse (Acta Neuropathologica 2004; 107: 257–264), we found increased unrepaired nuclear DNA single strand breaks (nDNA SSB) only in epithelial cells of the choroid plexus in the brain using autoradiographic methods after a continuous eight-week 50 Hz magnetic field (MF) exposure of adult mice with flux density of 1.5 mT.Methods
In the present study we tested the hypothesis that MF exposure with lower flux densities (0.1 mT, i.e., the actual exposure limit for the population in most European countries, and 1.0 mT) shows similar results to those in the previous study. Experiments and data analysis were carried out in a similar way as in our previous study.Results
Continuous eight-week 50 Hz MF exposure with 0.1 mT or 1.0 mT did not result in increased persisting unrepaired nDNA SSB in distinct types of cells in the brain, kidney, and liver of adult mice. MF exposure with 1.0 mT led to reduced unscheduled DNA synthesis (UDS) in epithelial cells in the choroid plexus of the fourth ventricle in the brain (EC-CP) and epithelial cells of the cortical collecting duct in the kidney, as well as to reduced mtDNA synthesis in neurons of the caudate nucleus in the brain and in EC-CP.Conclusion
No evidence was found for increased persisting unrepaired nDNA SSB in distinct types of cells in the brain, kidney, and liver of adult mice after continuous eight-week 50 Hz magnetic field exposure with flux density of 0.1 mT or 1.0 mT. 相似文献34.
Animals can exhibit complex movement patterns that may be the result of interactions with their environment or may be directly the mechanism by which their behaviour is governed. In order to understand the drivers of these patterns we examine the movement behaviour of individual desert locusts in a homogenous experimental arena with minimal external cues. Locust motion is intermittent and we reveal that as pauses become longer, the probability that a locust changes direction from its previous direction of travel increases. Long pauses (of greater than 100 s) can be considered reorientation bouts, while shorter pauses (of less than 6 s) appear to act as periods of resting between displacements. We observe power-law behaviour in the distribution of move and pause lengths of over 1.5 orders of magnitude. While Lévy features do exist, locusts' movement patterns are more fully described by considering moves, pauses and turns in combination. Further analysis reveals that these combinations give rise to two behavioural modes that are organized in time: local search behaviour (long exploratory pauses with short moves) and relocation behaviour (long displacement moves with shorter resting pauses). These findings offer a new perspective on how complex animal movement patterns emerge in nature. 相似文献

We describe a 20-point linkage analysis map of chromosome 11q22-23 that is based on genotyping 249 families (59 CEPH and 190 A-T). Monte Carlo linkage analyses of 176 ataxia-telangiectasia (A-T) families localizes the major A-T locus to the region between S1819(A4) and S1818(A2). When seven nonlinking families were excluded from subsequent analyses, a 2-lod support interval of ~500 kb was identified between S1819(A4) and S1294. No recombinants were observed between A-T and markers S384, B7, S535, or S1294. Only 17 of the international consortium families have been assigned to complementation groups. The available evidence favors either a cluster of A-T genes on chromosome 11 or intragenic defects in a single gene. 相似文献

Host stage preference, functional response and mutual interference of Aphidius matricariae (Haliday) (Hym.: Braconidae: Aphidiinae), parasitoid of the black bean aphid, Aphis fabae Scopoli (Hom.: Aphididae) were investigated in a growth chamber at a temperature of 25°C, 65% relative humidity and a photoperiod of 16 h light : 8 h dark on the PP8 variety of sugar beet. Nicholson's model and linear regression were used to determine per capita searching efficiency and interference coefficient, respectively. The highest parasitism percentage was observed on the third instar nymphs of A. fabae in both choice and no‐choice preference tests. There was significant difference between rates of parasitism on different stages of A. fabae (P < 0.01). Using logistic regression, a type II functional response was determined for A. matricariae. The Holling and Rogers models were used for estimating searching efficiency (a) and handling time (Th). The data was fitted by the Rogers equation better than by the Holling disc equation based on the R2 values. The estimated values of searching efficiency and handling time were 0.040 ± 0.013/h and 3.439 ± 0.383 h, respectively. The per capita searching efficiency decreased significantly from 0.272 to 0.139 as parasitoid densities increased from one to five. Therefore, different host‐parasitoid ratios could affect the efficacy of A. matricariae. 相似文献
