A crystalline compound, named stenophyllolide, obtained from an extract of Centaurea aspera var. stenophylla was shown to be 9β,15-dihydroxygermacra-1(10),4,11-trien-6α,12-olide by X-ray analysis. The molecular structure of stenophyllolide was solved with orthorhombic space group P212121, a = 11.719 (5), b = 13.389 (5), c = 8.646 (5) Å for Z = 4, by direct methods and refined to a final R of 0.06 for 1198 observed reflections. 相似文献

epi-Inositol was synthesized in six steps in 40% overall yield from a bacterial bromobenzene metabolite. The chemoenzymatic route involved toluene dioxygenase oxidation, substrate-directed catalytic osmylation, m-CPBA epoxidation, radical debromination, and Amberlite-catalized hydrolysis. The aim of this study was to determine the effects of leptin treatment on prepro-orexin and orexin receptor expression in the rat hypothalamus. Adult male rats, food-deprived for 48 and 72 h, were treated one time with vehicle or leptin (10 microg, icv). Prepro-orexin mRNA content was measured by semiquantitative RT-PCR, Northern blot, and in situ hybridization; orexin receptor 1 and 2 mRNA content was quantified by Northern blot and/or semiquantitative RT-PCR. Our results indicate that leptin inhibits a fasting-induced increase in prepro-orexin mRNA and orexin receptor 1 mRNA levels in the rat hypothalamus, while orexin receptor 2 mRNA levels were unchanged in all situations evaluated. These data provide direct evidence for an additional mechanism of adaptation of the hypothalamus to food deprivation and for a new effect of leptin in the regulation of food intake. 相似文献

Leptin, the product of the ob gene, is a hormone secreted by adipocytes that regulates food intake and energy expenditure. The hypothalamus-pituitary-thyroid axis is markedly influenced by the metabolic status, being suppressed during food deprivation.The aim of the present study was to assess whether leptin can act as a metabolic signal connecting the adipose tissue with the pituitary-thyroid axis. We studied the effect of leptin administration (10 microg, i.c.v.) on spontaneous TSH secretion and TSH responses to TRH in euthyroid and hypothyroid food-deprived rats. Spontaneous TSH secretion was assessed over 6 h with samples taken every 7 min. Administration of leptin to food-deprived euthyroid rats led to a reversal of the inhibitory effect exerted by fasting on spontaneous TSH secretion. This stimulatory effect of leptin on spontaneous TSH appears to be dependent on the thyroid status since it could not be observed in hypothyroid rats. This data suggests that blunted spontaneous TSH secretion in food-deprived rats is a functional and reversible state, and that the decreased leptin concentrations could be the primary event responsible for the suppression of the hypothalamic-pituitary-thyroid-axis in food-deprived rats. 相似文献

Background: Restoration of virus-specific immunity by virus specific T cells (VSTs) offers an attractive alternative to conventional drugs, and can be highly effective in immunocompromised patients, including hematopoietic stem cell transplant (HSCT) recipients. However, conventional VSTs manufacture requires preparation of specialized antigen-presenting cells (APCs), prolonged ex vivo culture in serum-containing medium and antigen re-stimulation with viruses or viral vectors to provide viral antigens for presentation on APCs. Methods: To simplify this complex process, we developed a method to generate multiple VSTs by direct stimulation of peripheral blood mononuclear cells (PBMCs) with overlapping peptide libraries in serum-free medium. Results: We generated VSTs that targeted seven viruses (cytomegalovirus [CMV], Epstein-Barr virus [EBV], adenovirus [AdV], human herpesvirus 6 [HHV-6], BK virus [BKV], JC virus [JCV] and Varicella Zoster virus [VZV]) in a single line. The phenotype, growth and specificity of multiple VSTs produced in serum-free medium were equivalent to those generated in conventional serum-containing medium. Discussion: The use of serum-free medium allows this approach to be readily introduced to clinical practice with lower cost, greater reproducibility due to the absence of batch-to-batch variability in serum and without concerns for infectious agents in the serum used. 【目的】克隆小菜蛾Plutella xylostella NanosO基因PxnosO启动子,并验证其具有生殖腺特异性活性,以期应用于基因功能研究或转基因昆虫的构建,为小菜蛾等农业害虫的综合治理提供新的研究思路。【方法】根据小菜蛾基因组序列信息,利用PCR技术克隆NanosO的启动子并进行序列分析。构建PxnosO-EGFP表达质粒,利用脂质体细胞转染技术将PxnosO-EGFP和IE1-EGFP表达质粒转入到小菜蛾胚胎细胞系(Px-6)和草地贪夜蛾Spodoptera frugiperda卵巢细胞系(Sf9)中,通过激光共聚焦荧光显微镜观察和qRT-PCR技术分别定性和定量分析EGFP基因的表达,验证小菜蛾NanosO启动子的活性。【结果】克隆获得小菜蛾PxnosO (Px004767)启动子区序列,长1 743 bp。对启动子序列进行分析,发现该序列不仅包含启动子共有核心元件TATA box以及上游启动子成分CAAT box和GC box等,还包含有数十个转录因子结合位点。利用细胞转染技术,在PxnosO启动子驱动下成功地在Px-6和Sf9细胞系中表达外源基因EGFP。【结论】克隆了小菜蛾NanosO基因PxnosO启动子,在细胞水平上验证其能驱动外源EGFP基因的表达,为分析PxnosO在小菜蛾不同发育时期的表达模式和PxnosO启动子在体内的功能验证奠定基础。 相似文献

The genus Pseudo‐nitzschia contains potentially toxic species of problematic taxonomy, making it one of the most intensively studied diatom genera. The study of 35 clonal strains isolated from the Bilbao estuary, an area that experiences recurrent blooms of Pseudo‐nitzschia, revealed the presence of two new species, P. abrensis and P. plurisecta, differing from their congeners in both morphology and gene sequence. The morphological features were analyzed by LM and EM, whereas molecular analyses were based on the internal transcribed spacer (ITS) and large subunit (LSU) regions of the rDNA. P. plurisecta appears closely related to P. cuspidata/P. pseudodelicatissima in the phylogenetic tree, whereas P. abrensis forms a moderately supported clade with P. heimii/P. subpacifica and P. caciantha/P. circumpora. Comparison of the secondary structure of ITS2 regions reveals marked differences in the most highly conserved regions among related taxa. Morphologically, the new species differ from their closest congeners in the arrangement of the poroid sectors and the density of valve striae and fibulae. The two species share similar pigment composition, and belong to the group of Pseudo‐nitzschia species containing only chlorophyll c2 and c3. 相似文献

Production of compost from marine waste: evaluation of the product for use in ecological agriculture

The sea contains large amounts of resources that are sometimes considered as waste. Such material includes the waste generated by the fish-processing industry and seaweed that is washed up on shores. In this study, these waste products were windrow composted, along with pine bark as a source of carbon and aeration. The final mix proportions were 20 % seaweed, 20 % fish waste, and 60 % pine bark (v/v). After 10 weeks, stable, well-structured, hygienic compost, which was rich in organic matter and nutrients and had a low metal content, was obtained. Tests for maturity, hygiene, and phytotoxicity, along with a detailed physical and chemical characterization, showed that this compost can be used as an organic amendment and/or growth substrate for use in ecological agriculture. The only limiting feature was the high salinity, which could easily be lowered prior to composting the material. 相似文献
