Sri Lankan rice farmers rarely practise green manuring and they depend mostly on chemical fertilizers for the nutrient requirement of their crops. With the removal of government subsidy on fertilizers since 1990, they are now faced with the dilemma of meeting the cost of production to sustain previous yield targets. Therefore the present study devotes to evaluate alternative cheaper nutrient sources like green manures for rice. Results of recent work in Sri Lanka have shown that Sesbania speciosa Taub. ex Engl. is a promising green manure for lowland rice in the dry zone. It was evaluated to adopt a suitable green manuring system for rice. Its year-round biomass production, nutrient accumulation during the vegetative growth and nitrogen fixation were monitored. Field trials were conducted to investigate its ability to suppress weed growth when grown during fallow periods and its contribution to soil fertility and effect on rice yield. Highest biomass of around 100 g dry matter per plant, was achieved when S. speciosa was seeded in August and harvested at flowering in November. When this was planted during a fallow period at a spacing of 0.25×0.25 m (16 plants m-2), percentage weed reduction was as much as 40%. Its N2 fixation activity was similar to that of S. sesban (L.) Merr., Nitrogen concentration in S. speciosa increased until 70 days after establishment and then decreased towards flowering. Phosphorus and K accumulation was also effective during this period. Therefore suitable time for soil-incorporation of S. speciosa is around 70 days after establishment. S. speciosa-manured and chemically fertilized rice crops were comparable in terms of grain yield, indicating the effective nutrient supplying potential of this green manure. Straw biomass was significantly high under S. speciosa, because of the high N-allocation to vegetative growth of rice under this. This evaluation shows that S. speciosa not only supplies N, but is also an effective and complete supplier of other nutrients for rice. A scheme to introduce this into rice production system is proposed, where S. speciosa is seeded/broadcast two weeks before crop harvest (around January) in Maha (major) season as well as Yala (minor) season (around June) so that its biomass can be incorporated into soil during land preparation of subsequent seasons.  相似文献

Effect of mild drying on the mineralization of soil nitrogen   总被引:6,自引:0,他引:6  
Summary Drying soil to –100 kPa increased the subsequent mineralization of nitrogen under optimal moisture conditions. The effect was greater when the soils were dried to –1500 Pa. Mineralization was greater after four cycles of wetting and drying than after one. Depending on the drying conditions, the amount of nitrogen mineralized after drying to –1500 Pa was between 6.8 and 18.2% of that mineralized after chloroform fumigation. After drying the soils the average ratio of CO2-C respired to min N was 21.1–22.3 depending on the drying conditions, whereas after chloroform treatment and autoclaving the ratio was 6.0 and 9.9 respectively. The effect of drying on nitrogen mineralization is attributed to two causes: the death and subsequent lysis of a small proportion of the soil organisms, and to the desorption of organic substances with a wide C/N ratio.Because of the stimulation of even mild drying conditions, marked differences in mineralization rates of soil nitrogen between cropping seasons must be expected.  相似文献

It is known that cyanobacteria in cyanolichens fix nitrogen for their nutrition. However, specific uses of the fixed nitrogen have not been examined. The present study shows experimentally that a mutualistic interaction between a heterotrophic N2 fixer and lichen fungi in the presence of a carbon source can contribute to enhanced release of organic acids, leading to improved solubilization of the mineral substrate. Three lichen fungi were isolated fromXanthoparmelia mexicana, a foliose lichen, and they were cultured separately or with a heterotrophic N2 fixer in nutrient broth media in the presence of a mineral substrate. Cells of the N2-fixing bacteria attached to the mycelial mats of all fungi, forming biofilms. All biofilms showed higher solubilizations of the substrate than cultures of their fungi alone. This finding has bearing on the significance of the origin and existence of N2-fixing activity in the evolution of lichen symbiosis. Further, our results may explain why there are N2-fixing photobionts even in the presence of non-fixing photobionts (green algae) in some remarkable lichens such asPlacopsis gelida. Our study sheds doubt on the idea that the establishment of terrestrial eukaryotes was possible only through the association between a fungus and a phototroph.  相似文献

Amino groups in proteins can non-enzymatically react with reducing sugars to generate a structurally diverse group of compounds referred to as advanced glycation end products (AGEs). The in vivo formation of AGEs contributes to some of the complications of diabetes including atherosclerosis, cataract formation, and renal failure. The formation of AGEs is dependent on both sugar and protein concentrations. Increases in temperature, pH, and exposure time of sugars to the proteins also play a significant role in the rate of AGE formation. This study focuses on the use of a combination of analytical techniques to study the in vitro AGE formation of HSA with dihydroxyacetone phosphate (DHAP), a ketose generated during glycolysis, and its dephosphorylated analog, dihydroxy acetone (DHA), commonly used as a browning reagent in skin tanning preparations. The extent of AGE formation was affected by DHAP and DHA concentrations and by the duration of HSA exposure to these glycating agents. Increases in temperature and pH sped the glycation process and enhanced the formation of the AGEs of HSA. MALDI-TOF mass spectroscopic data provided a reliable result to evaluate the extent of the AGE formation.  相似文献

Background

In 2009, an outbreak of dengue caused high fatality in Sri Lanka. We conducted 5 autopsies of clinically suspected myocarditis cases at the General Hospital, Peradeniya to describe the histopathology of the heart and other organs.

Methods

The diagnosis of dengue was confirmed with specific IgM and IgG ELISA, HAI and RT-PCR techniques. The histology was done in tissue sections stained with hematoxylin and eosin.

Results

Of the 319 cases of dengue fever, 166(52%) had severe infection. Of them, 149 patients (90%) had secondary dengue infection and in 5 patients, DEN-1 was identified as the causative serotype. The clinical diagnosis of myocarditis was considered in 45(27%) patients. The autopsies were done in 5 patients who succumbed to shock (3 females and 2 males) aged 13- 31 years. All had pleural effusions, ascites, bleeding patches in tissue planes and histological evidence of myocarditis. The main histological findings of the heart were interstitial oedema with inflammatory cell infiltration and necrosis of myocardial fibers. One patient had pericarditis. The concurrent pulmonary abnormalities were septal congestion, pulmonary haemorrhage and diffuse alveolar damage; one case showed massive necrosis of liver.

Conclusions

The histology supports occurrence of myocarditis in dengue infection.

Both nicotine and alcohol addictions are common chronic brain disorders that are of great concern to individuals and society. Although genetics contributes significantly to these disorders, the susceptibility genes and variants underlying them remain largely unknown. Many years of genome-wide linkage and association studies have implicated a number of genes and pathways in the etiology of nicotine and alcohol addictions. In this communication, we focus on current evidence, primarily from human genetic studies, supporting the involvement of genes and variants in the GABAergic signaling system in the etiology of nicotine dependence and alcoholism based on linkage, association, and gene-by-gene interaction studies. Current efforts aim not only to replicate these findings in independent samples, but also to identify which variant contributes to the detected associations and through what molecular mechanisms.  相似文献

Previous studies have demonstrated that the γ-aminobutyric acid type B (GABAB) receptor plays an essential role in modulating neurotransmitter release and regulating the activity of ion channels and adenyl cyclase. However, whether the naturally occurring polymorphisms in the two GABAB receptor subunit genes interact with each other to alter susceptibility to nicotine dependence (ND) remains largely unknown. In this study, we genotyped 5 and 33 single nucleotide polymorphisms (SNPs) for GABAB receptor subunit 1 and 2 genes (GABBR1, GABBR2), respectively, in a sample of 2037 individuals from 602 nuclear families of African- American (AA) or European-American (EA) origin. We conducted association analyses to determine (1) the association of each subunit gene with ND at both the individual SNP and haplotype levels and (2) the collective effect(s) of SNPs in both GABAB subunits on the development of ND. Several individual SNPs and haplotypes in GABBR2 were significantly associated with ND in both ethnic samples. Two haplotypes in AAs and one haplotype in EAs showed a protective effect against ND, whilst two other haplotypes in AAs and three haplotypes in EAs showed a risk effect for developing ND. Interestingly, these significant haplotypes were confined to two regions of GABBR2 in the AA and EA samples. Additionally, we found two minor haplotypes in GABBR1 to be positively associated with Heaviness of Smoking Index (HSI) in the EA sample. Finally, we demonstrated the presence of epistasis between GABBR1 and GABBR2 for developing ND. The variants of GABBR1 and GABBR2 are significantly associated with ND, and the involvement of GABBR1 is most likely through its interaction with GABBR2, whereas GABBR2 polymorphisms directly alter susceptibility to ND. Future studies are needed with more dense SNP coverage of GABBR1 and GABBR2 to verify the epistatic effects of the two subunit genes.  相似文献

Candida albicans is a common, opportunistic, human fungal pathogen that causes a variety of mucosal and systemic afflictions. It exists in nature both in the biofilm or the sessile phase, as well as in the free-floating or the planktonic phase. Candida biofilms, in particular, display unique characteristics that confer survival advantages over their planktonic counterparts, such as their recalcitrance to common antifungals. The mechanisms underlying Candida biofilm formation and their attributes are poorly understood. In this study, we used a 2-DE-based approach to characterize the protein markers that are differentially expressed in Candida biofilms in comparison to their planktonic counterparts. Using tandem mass spectrometric analysis, we have identified a significant number of proteins including alkyl hydroperoxide reductase, thioredoxin peroxidase, and thioredoxin involved in oxidative stress defenses that are upregulated in the biofilm phase. These proteomic findings were further confirmed by real-time PCR and lucigenin-based chemiluminescence assays. In addition, we demonstrate that a drug target for the new antifungal agent echinocandin, is abundantly expressed and significantly upregulated in Candida biofilms. Taken together, these data imply that the biofilm mode, Candida, compared with their planktonic counterparts, exhibits traits that can sustain oxidative stress (anti-oxidants), and thereby exert resistance to commonly used antifungals.  相似文献
