A two-phase aqueous/organic isolation system was developed for the isolation of epoxide-degrading bacteria. The potential of this system, for the isolation of cyclohexene oxide-degrading bacteria, was assessed by comparison to an analogous system lacking co-solvent. Using the biphasic isolation strategy, an epoxide-degrading Corynebacterium sp. designated C12, was isolated and was shown to grow on cyclohexene oxide as sole source of carbon and energy. Epoxide degradation appeared to proceed via a diol intermediate implicating the involvement of an epoxide hydrolase. The epoxide hydrolase of Corynebacterium sp. C12 was shown to have activity towards a range of terminal, sub-terminal and cyclic substrates. The enantioselectivity of the hydrolysis reaction was largely dependent on the nature of the substrate. In a series of biotransformations allowed to proceed to 50% substrate conversion, the remaining epoxide ranged from low (5% ee) to moderate (60% ee) optical purity.  相似文献

  1. Positive interspecific interactions such as mutualism, commensalism, and facilitation are globally ubiquitous. Although research on positive interactions in terrestrial and marine systems has progressed over the past few decades, comparatively little is known about them in freshwater ecosystems. However, recent advances have brought the study of positive interactions in freshwater systems to a point where synthesis is warranted.
  2. In this review, we catalogue the variety of direct positive interactions described to date in freshwater ecosystems, discuss factors that could influence prevalence and impact of these interactions, and provide a framework for future research.
  3. In positive interactions, organisms exchange key resources such as nutrients, protection, transportation, or habitat to a net benefit for at least one participant. A few mutualistic relationships have received research attention to date, namely seed-dispersing fishes, crayfishes and their ectosymbiotic cleaners, and communal-spawning stream fishes. Similarly, only a handful of commensalisms have been studied, primarily phoretic relationships. Facilitation via ecosystem engineering has received more attention, for example habitat modification by beavers and bioturbation by salmon.
  4. It is well known that interaction outcomes vary with abiotic and biotic context. However, only a few of studies have examined context dependency in positive interactions in freshwater systems. Likewise, positive interactions incur costs as well as benefits; conceptualising interactions in terms of net cost/benefit to participants will help to clarify complex interactions.
  5. It is likely that there are many positive interactions that have yet to be discovered in freshwater systems. To identify these interactions, we encourage inductive natural history studies combined with hypotheses deduced from general ecological models. Research on positive interactions must move beyond small-scale experiments and observational studies and adopt a cross-scale approach. Likewise, we must progress from reducing systems to oversimplified pairwise interactions, toward studying positive interactions in broader community contexts. Positive interactions have been greatly overlooked in applied freshwater ecology, but have great potential for conservation, restoration, and aquaculture.

Buruli ulcer (BU) is a disabling and stigmatising neglected tropical disease (NTD). Its distribution and burden are unknown because of underdiagnosis and underreporting. It is caused by Mycobacterium ulcerans, an environmental pathogen whose environmental niche and transmission routes are not fully understood. The main control strategy is active surveillance to promote early treatment and thus limit morbidity, but these activities are mostly restricted to well-known endemic areas. A better understanding of environmental suitability for the bacterium and disease could inform targeted surveillance, and advance understanding of the ecology and burden of BU. We used previously compiled point-level datasets of BU and M. ulcerans occurrence, evidence for BU occurrence within national and sub-national areas, and a suite of relevant environmental covariates in a distribution modelling framework. We fitted relationships between BU and M. ulcerans occurrence and environmental predictors by applying regression and machine learning based algorithms, combined in an ensemble model to characterise the optimal ecological niche for the disease and bacterium across Africa at a resolution of 5km x 5km. Proximity to waterbodies was the strongest predictor of suitability for BU, followed potential evapotranspiration. The strongest predictors of suitability for M. ulcerans were deforestation and potential evapotranspiration. We identified patchy foci of suitability throughout West and Central Africa, including areas with no previous evidence of the disease. Predicted suitability for M. ulcerans was wider but overlapping with that of BU. The estimated population living in areas predicted suitable for the bacterium and disease was 46.1 million.These maps could be used to inform burden estimations and case searches which would generate a more complete understanding of the spatial distribution of BU in Africa, and may guide control programmes to identify cases beyond the well-known endemic areas.  相似文献

Buruli ulcer is a skin disease caused by Mycobacterium ulcerans that is spreading in tropical countries, with major public health and economic implications in West Africa. Multi-analyte profiling of serum proteins in patients and endemic controls revealed that Buruli ulcer disease down-regulates the circulating levels of a large array of inflammatory mediators, without impacting on the leukocyte composition of peripheral blood. Notably, several proteins contributing to acute phase reaction, lipid metabolism, coagulation and tissue remodelling were also impacted. Their down-regulation was selective and persisted after the elimination of bacteria with antibiotic therapy. It involved proteins with various functions and origins, suggesting that M. ulcerans infection causes global and chronic defects in the host''s protein metabolism. Accordingly, patients had reduced levels of total serum proteins and blood urea, in the absence of signs of malnutrition, or functional failure of liver or kidney. Interestingly, slow healers had deeper metabolic and coagulation defects at the start of antibiotic therapy. In addition to providing novel insight into Buruli ulcer pathogenesis, our study therefore identifies a unique proteomic signature for this disease.  相似文献

ObjectiveWe aimed to determine the factors predicting hypothyroidism after radioactive iodine (RAI) treatment in patients with toxic adenoma and toxic multinodular goiter.MethodsWe retrospectively collected the data of 237 patients with toxic multinodular goiter or toxic adenoma who had consecutively received RAI treatment between 2014 and 2020 at 2 medical centers. Patients who received the second RAI treatment and whose medical records could not be accessed were excluded from the study. Finally, 133 patients were included in the study. RAI was administered at an empirical dose of 15 or 20 mCi.ResultsThe median age of the 133 participants was 69 years (interquartile range, 62-75 years), and 64.7% of the participants were women. A total of 42.1% of the patients had toxic adenoma, whereas 57.9% of patients had toxic multinodular goiter. The median follow-up was 24 months (interquartile range, 11-38 months). During the follow-up, 61.7% of patients became euthyroid, 30.8% developed hypothyroidism, and 7.5% remained hyperthyroid. The median month of hypothyroidism onset was 4 months (interquartile range, 2-9 months). Regression analysis revealed 2 factors that could predict hypothyroidism: thyroid-stimulating hormone (odds ratio, 2.548; 95% CI, 1.042-6.231; P = .04) and thyroid volume (odds ratio, 0.930; 95% CI, 0.885-0.978; P = .005).ConclusionOverall, 30.8% of the cases developed hypothyroidism after the RAI treatment. Approximately 78% of hypothyroidism developed within the first 10 months. The risk of hypothyroidism was higher in patients with higher thyroid-stimulating hormone and smaller thyroid volume.  相似文献

Mutant aminoglycoside 3'-phosphotransferase II enzymes were produced in which Tyr218 was changed to serine, aspartic acid, or phenylalanine. In each case the mutation resulted in increased bacterial susceptibility to neomycin and kanamycin, while simultaneously increasing the Km values for these substrates. For the Ser and Asp mutants, bacterial resistance to amikacin increased, with a concomitant increase in affinity for this drug. Initial velocity studies indicated that the wild-type and mutant enzymes all followed Michaelis-Menten kinetics. Although these mutagenic substitutions changed the substrate specificity of these enzymes they did not alter the enzyme affinity for Mg(2+)-ATP.  相似文献

  1. We compared the influence of biological traits (morphology, physiology, reproduction, and life history), ecological traits (geographic distribution, habitat associations, food habits), and introduction attributes (propagule pressure, human use of a species, residence time) on invasion success of native and introduced stream fishes during the colonisation and spread stages in an Appalachian (U.S.A.) river basin.
  2. Colonisation success was positively related to residence time, benthic feeding, an equilibrium life-history strategy, and nest spawning. Successful spread was associated with tolerance to increased temperature and an equilibrium life-history strategy. The spread of introduced fishes was negatively related to gamefish status. No effect of propagule pressure was detected.
  3. Traits linked to invasion success were consistent with the hypothesis that human land-use practices increase the invasibility of highland catchments by creating novel conditions better suited to lowland and equilibrium invaders.
  4. We found biological traits to be more useful than ecological traits in predicting invasion success and suggesting invasion mechanisms. Parental care and nest association can facilitate invasions over large spatial extents for both native and introduced fishes. Analyses of suites of traits can reveal mechanisms of invasions and tactics for controlling them; such mechanisms and tactics may be system-specific and scale-dependent.
