Plant seeds contain a large number of protease inhibitors of animal, fungal, and bacterial origin. One of the well-studied
families of these inhibitors is the Bowman-Birk family(BBI). The BBIs from dicotyledonous seeds are 8K, double-headed proteins.
In contrast, the 8K inhibitors from monocotyledonous seeds are single headed. Monocots also have a 16K, double-headed inhibitor.
We have determined the primary structure of a Bowman-Birk inhibitor from a dicot, horsegram, by sequential edman analysis
of the intact protein and peptides derived from enzymatic and chemical cleavage. The 76-residue-long inhibitor is very similar
to that ofMacrotyloma axillare. An analysis of this inhibitor along with 26 other Bowman-Birk inhibitor domains (MW 8K) available in the SWISSPROT databank
revealed that the proteins from monocots and dicots belong to related but distinct families. Inhibitors from monocots show
larger variation in sequence. Sequence comparison shows that a crucial disulphide which connects the amino and carboxy termini
of the active site loop is lost in monocots. The loss of a reactive site in monocots seems to be correlated to this. However,
it appears that this disulphide is not absolutely essential for retention of inhibitory function. Our analysis suggests that
gene duplication leading to a 16K inhibitor in monocots has occurred, probably after the divergence of monocots and dicots,
and also after the loss of second reactive site in monocots.

Efficient Agrobacterium tumefaciens-mediated transformation and a higher recovery of transformed plants of cucumber cv. Poinsett76 were achieved via direct organogenesis from cotyledon explants. Stable transformants were obtained by inoculating explants with A. tumefaciens strains EHA105 or LBA4404, both harboring the binary vector pME508, which contains the neomycin phosphotransferase II (nptII) and phosphinothricin resistance genes (bar) conferring resistance to kanamycin and PPT, respectively, as selectable markers and the sgfp-tyg gene for the green fluorescent protein (GFP) as a visual marker driven by the constitutive CaMV35S promoter in the presence of acetosyringone (50 μM). Transformed shoots were obtained on MS Murashige and Skoog (Plant Physiol. 15: 473–497, 1962) medium supplemented with 1 mg L−1 benzyladenine (BA), 20 mg L−1 l-glutamine and 2 mg L−1 phosphinothricin (PPT) or 100 mg L−1 kanamycin. The regenerated shoots were examined in vivo using a hand-held long wave UV lamp for GFP expression. The GFP screening helped identify escapes and chimeric shoots at regular intervals to increase the growth of transformed shoots on cotyledon explants. Elongation and rooting of putative transformants were achieved on PPT (2 mg L−1) containing MS media with 0.5 mg L−1 gibberellic acid (GA3) and 0.6 mg L−1 indole butyric acid (IBA), respectively. PCR and Southern analyses confirmed the integration of the sgfp gene into the genome of T0 and the progenies. T1 segregation of transgenic progeny exhibited Mendelian inheritance of the transgene. The use of EHA105 resulted in 21% transformation efficiency compared to 8.5% when LBA4404 was used. This higher rate was greatly facilitated by PPT selection coupled with effective screening of transformants for GFP expression, thus making the protocol highly useful for the recovery of a higher number of transgenic cucumber plants. 相似文献

It is of interest to analyse and compare the antioxidant and anti-diabetic activity of ethanolic extracts of Andrographis echioides and Andrographis paniculata. Andrographis echioides and Andrographis paniculata were collected from a local farm. In vitro antioxidant activity was assessed by the potential of Piperine, Lupeol, beta sitosterol; DPPH free radical scavenging assay was performed by Liyana Pathirana and Shahidi method. In vitro anti-diabetic activity was assessed by alpha amylase inhibitory activity and alpha glucosidase inhibitory activity. The data were analysed by one-way-ANOVA to check the statistical significance among the groups and considered at the levels of p<0.05. Both the ethanolic extracts of Andrographis echioides and Andrographis paniculata showed significant antioxidant and anti-diabetic potential in a dose-dependent manner (100-500µg) and can be used as potential antidiabetic agents. Similar to antioxidant potential, Andrographis paniculata exhibited an increased anti-diabetic potential compared to Andrographis echioides. Data shows that the ethanolic extracts of Andrographis echioides and Andrographis paniculata possessed antioxidant and anti-diabetic activity and hence our present findings conclude that both plants can be considered for the development of natural drugs for the management of diabetes. 相似文献

For the past few decades, intensive studies have been carried out in an attempt to understand how the amino acid sequences of proteins encode their three dimensional structures to perform their specific functions. In order to understand the sequence-structure relationship of proteins, several sub-sequence search studies in non-redundant sequence-structure databases have been undertaken which have given some fruitful clues. In our earlier work, we analyzed a set of 3124 non-redundant protein sequences from the Protein Data Bank (PDB) and retrieved 30 identical octapeptides having different secondary structures. These octapeptides were characterized by using different computational procedures. This prompted us to explore the presence of octapeptides with reverse sequences and to analyze whether these octapeptides would adopt similar structures as that of their parent octapeptides. Our identical reverse octapeptide search resulted in the finding of eight octapeptide pairs (octapeptide and reverse octapeptide) with similar secondary structure and 23 octapeptide pairs with different secondary structures. In the present work, the geometrical and biophysical characteristics of identical reverse octapeptides were explored and compared with unrelated octapeptide pairs by using various computational tools. We thus conclude that proteins containing identical reverse octapeptides are not very abundant and residues in the octapeptide pairs do not contribute to the stability of the protein. Furthermore, compared to unrelated octapeptides, identical reverse octapeptides do not show certain biophysical and geometrical properties. 相似文献

Standard hematology parameters were determined for 122 sloth bears (Melursus ursinus ursinus) at the Sur Sarovar Bird Sanctuary, Uttar Pradesh, India (27 degrees 0'N; 77 degrees 45'E), and the Bannerghatta Biological Park, Karnataka, India (12 degrees 48'N; 77 degrees 34'E) from March 2003 to July 2006. These two native sloth bear habitats have different climatic conditions and provided an opportunity to examine the effect of climate on the physiologic hematology values of these bears. We primarily analyzed the influence of age, sex, season, and body weight on the different hematology parameters. Several values were significantly different in sloth bear cubs (= 1 yr) when compared to adult and sub-adult bears (>1 yr). The cubs had a lower erythrocyte count, hemoglobin concentration, packed cell volume (PCV), and mean cell hemoglobin (MCV) values when compared to adult and subadult bears. The cubs also had higher leukocyte counts, due to higher circulating neutrophils, as compared to adult and subadult bears. Within subadult and adult bears, we also identified a sexual dimorphic difference in leukocyte count in adult and subadult bears, wherein female bears had higher counts than males. This difference was the result of a significantly higher number of circulating neutrophils in female bears. Platelet counts were also higher in females as compared to males. On comparing different seasons, leukocyte counts were higher in winter as compared to the summer and monsoon seasons. When compared based on location, erythrocyte counts were higher in subadult and adult bears at Bannerghatta, which was at a higher altitude than Sur Sarovar. Within subadult and adult bears, we did not find any significant influence of age or body weight on the different hematologic parameters. In this study we have obtained mean hematologic values for sloth bears in their native habitat to serve as a reference for this species. Retinol-binding protein (RBP) is secreted out of the cell in its ligand-bound holo-form. The apo-form of RBP is selectively retained within the endoplasmic reticulum (ER) by a mechanism that remains unknown. Using isolated microsomal system, we have recapitulated the biogenesis of RBP involving its oxidative folding and assembly with transthyretin in the ER. In addition to dissecting its pathway of disulfide oxidation, we have analyzed association of its early folding intermediates with ER-chaperones. Our results show that of the three intramolecular disulfides present in RBP (4–160, 70–174, and 120–129) the smallest loop (120–129) was most critical for RBP to fold. Its absence caused RBP to aggregate into an intermolecular disulfide-linked structure. After acquisition of the small loop, formation of one of the two big disulfides (4–160 or 70–174) was sufficient for RBP to acquire a folded state. Using cross-linking in intact microsomes and sedimentation on sucrose gradients, we show that newly synthesized RBP is associated with a complex of chaperones consisting of Grp94, BiP, PDI, and calnexin. The complex was constitutively present in the ER, independent of the presence of folding substrates. RBP dissociated from this complex coincident with the formation of one of the two big disulfide loops, whereas RBP mutant lacking both the large disulfides showed persistent association. While highlighting the matrix-like characteristics of ER in isolated microsomal system our results provide insight into RBP folding and assembly mechanisms that will aid our understanding of its complex secretion properties. 相似文献
