The mammalian skull is asymmetrically loaded during mastication because most of these animals chew on only one side at a time. This loading regime tends to twist the braincase relative to the rostral, tooth bearing part of the skull at the zone of potential weakness between the orbits. This torsional effect is exaggerated, and a postorbital bar is present, in those animals with very large masseter and pterygoid muscles. The lines of action of these muscles are oriented at large angles to the long axis of the skull in lateral view, providing large components of force that twist the skull segments relative to one another. When the temporalis is the dominant muscle, the torsional effect is usually less important, and the bar is absent, because this muscle acts at a smaller angle to the skull axis. The postorbital bar exhibits the predicted three dimensional spatial orientation required to resist torsional forces: it is a segment of an imaginary 45° helix that is wound around the skull, if the skull is idealized as a cylinder. This orientation is significant because, in general, maximum compressive and tensile shear stresses lie along 45° helices on a cylinder loaded in torsion; to resist torsion, material should be placed far from the axis of torsion and along a helix oriented at 45° to the deforming forces. Each half of a supraorbital ridge is also a segment of a 45° helix that is perpendicular to the helix passing through the postorbital bar. This model suggests that the postorbital bar is loaded in compression on the chewing side and in tension on the non-chewing side; the supraorbital ridge is loaded in tension on the chewing side and in compression on the non-chewing side. 相似文献

Hybrid vigour (heterosis) has been used for decades in crop industries, especially in the production of maize and rice. Hybrid varieties usually exceed their parents in plant biomass and seed yield. But the molecular basis of hybrid vigour is not fully understood. In this project, we studied heterosis at early stages of seedling development in Arabidopsis hybrids derived from crossing Ler and C24 accessions. We found that early heterosis is associated with non‐additive gene expression that resulted from earlier changes in gene expression in the hybrids relative to the parents. The non‐additively expressed genes are involved in metabolic pathways, including photosynthesis, critical for plant growth. The early increased expression levels of genes involved in energy production in hybrids is associated with heterosis in the young seedlings that could be essential for biomass heterosis at later developmental stages of the plant. 相似文献

Psoriasis is a common chronic inflammatory and proliferative skin disease characterised by epidermal neutrophil infiltration which may be induced by chemotactic substances in the involved epidermis. Superficial psoriatic scale was shown to contain biologically active amounts of leukotriene B4 and monohydroxy-eicosatetraenoic acid (HETE)- like material as determined by assay for chemokinetic activity in high performance liquid chromatography (HPLC) fractions of scale extracts. Extracts of scale and chamber fluid from abraded lesional and uninvolved psoriatic skin were purified by HPLC and appropriate fractions were analysed by gas chromatography - mass spectrometry (GC-MS). The following monohydroxy metabolites of arachidonic, linoleic and 11,14-eicosadienoic acids were identified : 15-HETE, 12-HETE, 11-HETE, 9-HETE, 8-HETE, 5-HETE, 13-hydroxy-octadecadienoic acid (13-HODD), 9-HODD and 15-hydroxy-eicosadienoic acid (15-HEDE). The results suggested that 12-HETE, 13-HODD and 9-HODD are the most abundant monohydroxy fatty acids in the psoriatic skin extracts described above. Assays of 13-HODD, 9-HODD and 15-HEDE for chemokinetic activity were negative with concentrations up to 10?4M. The biological significance of these three compounds in not known, but some of the hydroxylated metabolites of arachidonic acid may, by virtue of their chemotactic properties, be relevant to the pathogenesis of the psoriatic neutrophil infiltrate. 相似文献

The presence of neutrophil chemoattractant material in aqueous extracts of lesional psoriatic scale has been investigated by use of an agarose microdroplet chemokinesis method in combination with ultrafiltration and high-performance liquid chromatography (HPLC). Fractions were also assayed for murine thymocyte co-stimulating activity. Aqueous extracts of psoriatic scale contained significantly greater neutrophil chemokinetic activity than extracts of scale from normal skin. Successive ultrafiltration of extracts showed that the chemokinetic material was 10 to 30 kd. Heat lability and gel filtration HPLC characteristics suggested that the major chemokinetically active material in aqueous extracts of psoriatic scale is different from C5a des arg. Reversed-phase HPLC of 0.1% trifluoroacetic acid/acetonitrile extracts of psoriatic scale revealed two clearly resolved peaks of chemokinetic activity, the major peak consistently containing thymocyte co-stimulating activity. No significant neutrophil chemokinetic activity was seen in fractions after reversed-phase HPLC of scale from normal skin. These findings suggest that a major portion of the neutrophil chemoattractant activity in aqueous extracts of psoriatic scale is due to interleukin 1-like material, which may play a role in the pathogenesis of this disease. 相似文献

A cDNA clone bank containing 30 000 clones was constructed from sucrose gradient-fractionated mRNA from human placenta. mRNA coding for transferrin receptor (TR) was enriched by polysome immuno-adsorbed chromatography with monospecific rabbit IgG and protein-A Sepharose. The library was screened for hybridisation to 32P-labelled cDNA synthesised from immunoselected TR mRNA and from poly(A)+ RNA of the polysome fraction that failed to bind to protein-A Sepharose. Plasmids isolated from colonies showing hybridisation only to the probe made from immunoselected mRNA were then subjected to hybrid selection. Two clones, pTR-48 and pTR-67, were able to hybridise the mRNA coding for the TR. 相似文献

The activities of NAD+-dependent 15-hydroxy prostaglandin dehydrogenase in soluble fractions of rat skin and lung were compared by using a radiochemical assay method. Tritiated prostaglandin F2 alpha was incubated with NAD+ and 120,000 g supernatant of tissue homogenate. Extracted prostaglandin substrate and reaction products were separated by t.l.c. and quantitatively determined by liquid-scintillation counting. With skin 120,000 g supernatant, 10 mM-NAD+ and an incubation time of 15 min, the mean Vmax. was 5.5 nmol of prostaglandin F2 alpha converted/s per litre of reaction mixture. With lung 120,000 g supernatant, 60 mM-NAD+ and an incubation time of 5 min, the mean Vmax. was 26.9 nmol/s per litre, demonstrating 5-fold greater dehydrogenase activity in lung per unit wet weight of tissue. However, the total wet weight of skin was about 23 times that of lung, on dissection of individual rats, indicating that the entire skin may contain 4.5 times the total 15-hydroxy prostaglandin dehydrogenase activity of the lungs. Azathioprine was given to 11 patients with pemphigoid who had been on long-term maintenance therapy with prednisone or prednisolone. In nine of these prednisone therapy was withdrawn and all were maintained symptom-free on azathioprine alone, while in two the dose of prednisone was considerably reduced. One patient who had never received corticosteroids was controlled by azathioprine alone during the initial acute phase of the illness. Since azathioprine acts slowly, it is recommended that corticosteroids should be used together with azathioprine during the acute stage. Thus azathioprine is valuable in long-term management of pemphigoid, particularly in patients showing corticosteroid toxicity or in whom the minimum maintenance dose is dangerously high. 相似文献
