Leptospira immunoglobulin-like (Lig) proteins including LigA and LigB are adhesins that bind to fibronectin, collagen, laminin and elastin. In addition, Lig proteins are fibrinogen (Fg)-binding proteins, although the physiological role of the Lig-Fg interaction is unclear. In this study, a previously identified Fg-binding region, LigBCen2 (amino acids 1014-1165 of LigB), has been further localized to LigBCen2R, which consists of the partial 11th and entire 12th Ig-like domain (amino acids 1014-1119). LigBCen2R was found to bind to the C-terminal αC domain of Fg (FgαCC; amino acids 392-644 in Fg α chain; isothermal titration calorimetry, K(D) = 0.375 μM; fluorescence spectrometry, K(D) = 0.364 μM). The quenching and blue shift observed for the maximum wavelength intensities of the tryptophan fluorescence spectra for FgαCCY570W upon LigBCen2RW1073C binding suggested an RGD motif close to the sole tryptophan on FgαCCY570W was buried in LigBCen2R upon saturation with FgαCC. A conformational change in LigBCen2R when bound to the FgαCC RGD motif blocked further binding to integrin α(IIb) β3 on platelets, thus preventing their aggregation. LigBCen2R binding to FgαCC reduced clot formation but did not affect plasminogen and tissue-type plasminogen activator interactions with FgαCC. This study is the first to report that a spirochaetal protein binds to the C-terminal αC domain of Fg, which regulates thrombosis and fibrinolysis, and may help explain the pulmonary haemorrhage and thrombocytopenia seen in clinical cases of leptospirosis. 相似文献

Phase resetting curves (PRCs) provide a measure of the sensitivity of oscillators to perturbations. In a noisy environment,
these curves are themselves very noisy. Using perturbation theory, we compute the mean and the variance for PRCs for arbitrary
limit cycle oscillators when the noise is small. Phase resetting curves and phase dependent variance are fit to experimental
data and the variance is computed using an ad-hoc method. The theoretical curves of this phase dependent method match both
simulations and experimental data significantly better than an ad-hoc method. A dual cell network simulation is compared to
predictions using the analytical phase dependent variance estimation presented in this paper. We also discuss how entrainment

The Morris water maze is an experimental procedure in which animals learn to escape swimming in a pool using environmental
cues. Despite its success in neuroscience and psychology for studying spatial learning and memory, the exact mnemonic and
navigational demands of the task are not well understood. Here, we provide a mathematical model of rat swimming dynamics on
a behavioural level. The model consists of a random walk, a heading change and a feedback control component in which learning
is reflected in parameter changes of the feedback mechanism. The simplicity of the model renders it accessible and useful
for analysis of experiments in which swimming paths are recorded. Here, we used the model to analyse an experiment in which
rats were trained to find the platform with either three or one extramaze cue. Results indicate that the 3-cues group employs
stronger feedback relying only on the actual visual input, whereas the 1-cue group employs weaker feedback relying to some
extent on memory. Because the model parameters are linked to neurological processes, identifying different parameter values

The recent spread of dreissenid mussels to various bodies of water in the western US has sparked interest by many state and federal agencies to develop protocols to stop further expansion. Quagga mussels (Dreissena rostriformis bugensis) are of particular importance as they are currently the most widespread dreissenid species in the region. This project examined the susceptibility of quagga mussels to hot-water sprays at different temperatures and durations of spray contact at Lake Mead (Nevada-Arizona, USA). Emersed adult quagga mussels were exposed to hot-water sprays at 20, 40, 50, 54, 60, 70, and 80°C for 1, 2, 5, 10, 20, 40, 80, and 160?s. Sprays at ≥60°C for 5?s were shown to be 100% lethal. Sprays of 54°C for 10?s, 50°C for 20?s, and 40°C for 40?s also resulted in 100% mortality. A spray temperature of 60°C for 5?s is recommended for mitigating fouling by quagga mussels. 相似文献
