Isolation and characterization of circulating low molecular weight peptides in steer, sheep and rat portal and peripheral blood.

1. Low mol. wt peptides in plasma were isolated by reverse-phase HPLC from steer and sheep carotid arterial and rat heart blood and portal blood from all three species. 2. Elution profiles for peptide fractions were similar but the concentration of peptide-bound amino acids (PBAA) in fractions corresponding to different mol. wt peptides was not constant across species. 3. PBAA contributed between 65 and 78% to the plasma amino acid pool in steer and sheep but only 52% in the rat (P less than 0.05). 4. The percentage of many individual amino acids present in either free amino acid (FAA) or PBAA pools was different for ruminant compared with rat plasma but it was similar for steer and sheep apart from branch-chain amino acids (P less than 0.05).     

Ability of thawed tiger (Panthera tigris) spermatozoa to fertilize conspecific eggs and bind and penetrate domestic cat eggs in vitro.

Electroejaculates from tigers were collected and half was used fresh to inseminate tiger eggs in vitro and domestic cat eggs stored in a hypertonic salt solution. The remainder was pelleted, frozen in a solution of 20% egg yolk, 11% lactose and 4% glycerol, thawed and cultured with tiger and domestic cat eggs. The motility index ((sperm % motility)+(status rating x 20))/2 for thawed spermatozoa was about 86% of that in fresh aliquots. Of the 49 tiger oocytes inseminated in vitro with fresh spermatozoa, 34 (69.4%) cleaved, compared with 33 of 47 oocytes (70.2%) cultured with thawed spermatozoa (P > 0.05). Embryos generated by either sperm treatment could develop in vitro to the 16-cell or morula stage. Fresh and thawed tiger spermatozoa were equally capable (P > 0.05) of binding and penetrating the outer and inner zona pellucida of domestic cat eggs. These results demonstrate the ability of frozen-thawed tiger spermatozoa to (i) penetrate homologous and heterologous eggs and (ii) result in conspecific, advanced development of preimplantation embryos in vitro.     

Purine metabolite concentrations in portal and peripheral blood of steers, sheep and rats.

1. The concentration of purine derivatives in portal and peripheral blood of steers, sheep and rats was measured by reverse-phase high performance liquid chromatography. 2. Nucleotides, nucleosides (apart from inosine), adenine and guanine were not found in the plasma samples. Allantoin, uric acid, hypoxanthine and xanthine accounted for virtually all purine metabolites in plasma samples. 3. Non-oxidized derivatives (hypoxanthine and xanthine) were consistently detected in sheep but not in steer or rat plasma samples showing a differential availability of reutilizable purine derivatives between species.     

Isolation of a Pseudomonas solanacearum-specific DNA probe by subtraction hybridization and construction of species-specific oligonucleotide primers for sensitive detection by the polymerase chain reaction.

A subtraction hybridization technique was employed to make a library enriched for Pseudomonas solanacearum-specific sequences. One cloned fragment, PS2096, hybridized under stringent conditions to DNA of 82 P. solanacearum strains representing all subgroups of the species. Other plant-associated bacteria, including closely related species such as Pseudomonas capacia, Pseudomonas picketti, or Pseudomonas syzygii, did not hybridize to PS2096. A minimum number of between 4 x 10(5) and 4 x 10(6) P. solanacearum cells could routinely be detected with PS2096 labelled either with [32P]dCTP or with digoxigenin-11-dUTP. To improve the sensitivity of detection, PS2096 was sequenced to allow the construction of specific oligonucleotide primers to be used for polymerase chain reaction (PCR) amplification. After 50 cycles of amplification, 5 to 116 cells, depending on the strain, could reproducibly be detected by visualization of a 148-bp PCR product on an agarose gel. A preliminary field trial in Burundi with the probe and PCR primers has confirmed that they are sensitive tools for specifically detecting low-level infections of P. solanacearum in potato tubers.     

Physiological responses of red foxes (Vulpes vulpes) to surgery

Radio transmitters were surgically implanted into the abdomens of red foxes (Vulpes vulpes). Blood samples were taken before, immediately after, and 8 hr after surgery and analyzed for hormonal, biochemical, electrolyte and hematologic changes. Samples were taken at the same times from control foxes. Adrenocorticotropin increased after surgery (P less than 0.05), but returned to pre-surgery values after 8 hr. Cortisol increased and remained elevated in the surgery group relative to pre-surgery values or to control values (P less than 0.05); Triiodothyronine and thyroxine both decreased from post-surgery values 8 hr later (P less than 0.05). Creatine kinase, total bilirubin and aspartate aminotransferase increased after 8 hr in both surgery and control groups (P less than 0.05). Carbon dioxide increased under anesthesia in both groups, but returned to initial values after 8 hr (P less than 0.05). The white blood cell count increased after 8 hr only in the surgery group (P less than 0.05). There were no differences between the groups for any value obtained from the initial blood sample. These data indicate that abdominal surgery results in prolonged adrenocortical activity and decreased thyroid hormone levels, but otherwise has minimal systemic effects in red foxes.     

The influence of social and endocrine factors on urine-marking by captive wolves (Canis lupus)

Although serum hormones varied seasonally in all adult animals, only dominant male and female wolves urine-marked. Serum testosterone and urine-marking rates, which increased during the fall/winter breeding season, were positively correlated in both male and female dominant wolves. Estradiol, which increased in conjunction with proestrus and estrus, was not correlated with female urinemarking. These findings suggest that hormonal influence on urine-marking in the wolf is modulated by social factors and contrast with those for both domestic dogs and coyotes, two other members of the genus Canis.

蓖麻蚕Philosamia cynthia ricni血淋巴含两种凝集素,一种凝集兔新鲜红血球,凝血活力被L-鼠李糖和D-半乳糖抑制;另一种凝集戊二醛固定的人和鸡的红血球,凝血活力被岩藻糖抑制.它们在蚕的不同生长阶段及在蚕体各组织中的分布和凝血活力显著不同.血淋巴中这两种凝集素的凝血活力明显比其他组织中高.卵中测不到这两种凝集素活力.本文对这两种凝集素在蚕体中可能的生理功能进行了讨论.     

Modulation of glial glycogen metabolism by 5-hydroxytryptamine in leech segmental ganglia

Leech segmental ganglia (16 out of 23 per animal) were divided into experimental and control groups (4 ganglia per group). The amounts of glycogen in the ganglia were assayed by a specific extraction procedure and fluorimetry, or by liquid scintillation counting following labelling of the glycogen by [³H]glucose. Within any individual animal the amounts of glycogen in the ganglia were relatively constant (max. variation 16%). 5-HT (10⁻⁶–10⁻⁴ M) reduced in a dose-dependent manner the endogenous glycogen (max. 20% reduction), and the [³H]glycogen (max. 60% reduction). The glycogenolytic effect was studied by light-microscope autoradiography in serial sections of segmental ganglia previously exposed to [³H]glucose. The 5-HT-mediated glycogenolysis was localized principally in the glial cells surrounding the neuron perikarya. 5-HT, in addition to its conventional transmitter role, may regulate the supply of energy substrate from glial cells to neurons within domains defined by the projections of the neurons from which it is released.     

Penetration of zona-free hamster ova by Siberian tiger sperm

Sperm from the electroejaculate of captive Siberian tigers (Panthera tigris altaica) penetrated zona pellucida-free hamster ova in vitro, evidenced by a decondensation reaction. This assay, when used in conjunction with semen analysis, may be useful in assessing the fertility potential of males of this and other related felids. Such information is an important step in developing successful long-term management strategies for captive and wild populations of this severely endangered species.
