There is continuing international interest in exploring and developing the therapeutic potential of marine–derived small molecules. Balancing the strategies for ocean based sampling of source organisms versus the potential to endanger fragile ecosystems poses a substantial challenge. In order to mitigate such environmental impacts, we have developed a deployable artificial sponge. This report provides details on its design followed by evidence that it faithfully recapitulates traditional natural product collection protocols. Retrieving this artificial sponge from a tropical ecosystem after deployment for 320 hours afforded three actin–targeting jasplakinolide depsipeptides that had been discovered two decades earlier using traditional sponge specimen collection and isolation procedures. The successful outcome achieved here could reinvigorate marine natural products research, by producing new environmentally innocuous sources of natural products and providing a means to probe the true biosynthetic origins of complex marine–derived scaffolds.  相似文献

Diarrheagenic Escherichia coli (DEC) cause acute and persistent diarrhoea worldwide, but little is known about their epidemiology in Mexico. We determined the prevalence of bacterial enteropathogens in 831 children with acute diarrhoea over a four-year period in Yucatan, Mexico. Six DEC supplementary virulence genes (SVG), mainly associated with enteroaggregative E. coli (EAEC), were sought in 3100 E. coli isolates. DEC was the most common bacterial enteropathogen (28%), surpassing Salmonella (12%) and Shigella (9%). Predominant DEC groups were diffusely adherent E. coli (DAEC) (35%), EAEC (24%), and enteropathogenic E. coli (EPEC) (19%). Among children with DEC infections, 14% had severe illness mainly caused by EPEC (26%) and DAEC (18%); 30% had moderate diarrhoea mainly caused by DAEC (36%), mixed DEC infections (33%) and EAEC (32%). DAEC was most prevalent during spring, while ETEC, EAEC and EPEC predominated in summer. EAEC was more frequent in children 6–24 months old than in those younger than 6 months of age (P = 0.008, OR = 4.2, 95% CI, 1.3–13.9). The presence of SVG dispersin, (aatA), dispersin-translocator (aatA), enteroaggregative heat-stable toxin 1 (astA), plasmid encoded toxin (pet), cytolethal distending toxin (cdt) was higher in DEC than non-DEC strains, (36% vs 26%, P <0.0001, OR = 1.5, 95% CI, 1.3–1.8). 98% of EAEC-infected children harboured strains with SVG; 85% carried the aap-aatA gene combination, and 33% of these also carried astA. 28% of both EPEC and ETEC, and 6% of DAEC patients had strains with SVG. 54% of EPEC patients carried pet-positive strains alone or in combination with astA; only this DEC group harboured cdt-positive isolates. All ETEC patients carried astA- or astA-aap-positive strains. astA and aap were the most common SVG in DAEC (3% and 2%) and non-DEC strains (21% and 13%). DEC carrying SVG are an important cause of moderate to severe bacterial diarrhoea in Mexican children.  相似文献

The collision avoidance and resolution multiple access (CARMA) protocol is presented and analyzed. CARMA uses a collision avoidance handshake in which the sender and receiver exchange a request to send (RTS) and a clear to send (CTS) before the sender transmits any data. CARMA is based on carrier sensing, together with collision resolution based on a deterministic tree-splitting algorithm. For analytical purposes, an upper bound is derived for the average number of steps required to resolve collisions of RTSs using the tree-splitting algorithm. This bound is then applied to the computation of the average channel utilization in a fully connected network with a large number of stations. Under light-load conditions, CARMA achieves the same average throughput as multiple access protocols based on RTS/CTS exchange and carrier sensing. It is also shown that, as the arrival rate of RTSs increases, the throughput achieved by CARMA is close to the maximum throughput that any protocol based on collision avoidance (i.e., RTS/CTS exchange) can achieve if the control packets used to acquire the floor are much smaller than the data packet trains sent by the stations. Simulation results validate the simplifying approximations made in the analytical model. Our analysis results indicate that collision resolution makes floor acquisition multiple access much more effective. Animal locomotion requires highly coordinated working of the segmental neuronal networks that control the limb movements. Experiments have shown that sensory signals originating from the extremities play a pivotal role in controlling locomotion patterns by acting on central networks. Based on the results from stick insect locomotion, we constructed an inter-segmental model comprising local networks for all three legs, i.e. for the pro-, meso- and meta-thorax, their inter-connections and the main sensory inputs modifying their activities. In the model, the local networks are uniform, and each of them consists of a central pattern generator (CPG) providing the rhythmic oscillation for the protractor-retractor motor systems, the corresponding motoneurons (MNs), and local inhibitory interneurons (IINs) between the CPGs and the MNs. Between segments, the CPGs are connected cyclically by both excitatory and inhibitory pathways that are modulated by the aforementioned sensory inputs. Simulations done with our network model showed that it was capable of reproducing basic patterns of locomotion such as those occurring during tri- and tetrapod gaits. The model further revealed a number of elementary neuronal processes (e.g. synaptic inhibition, or changing the synaptic drive at specific neurons) that in the simulations were necessary, and in their entirety sufficient, to bring about a transition from one type of gait to another. The main result of this simulation study is that exactly the same mechanism underlies the transition between the two types of gait irrespective of the direction of the change. Moreover, the model suggests that the majority of these processes can be attributed to direct sensory influences, and changes are required only in centrally controlled synaptic drives to the CPGs.  相似文献

Understanding the lateral organization of biological membranes plays a key role on the road to fully appreciate the physiological functions of this fundamental barrier between the inside and outside regions of a cell. Ternary lipid bilayers composed of a high and a low melting temperature lipid and cholesterol represent a model system that mimics some of the important thermodynamical features of much more complex lipid mixtures such as those found in mammal membranes. The phase diagram of these ternary mixtures can be studied exploiting fluorescence microscopy in giant unilamellar vesicles, and it is typically expected to give rise, for specific combinations of composition and temperature, to regions of two-phase coexistence and a region with three-phase coexistence, namely, the liquid-ordered, liquid-disordered, and solid phases. Whereas the observation of two-phase coexistence is routinely possible using fluorescence microscopy, the three-phase region is more elusive to study. In this article, we show that particular lipid mixtures containing diphytanoyl-phosphatidylcholine and cholesterol plus different types of sphingomyelin (SM) are prone to produce bilayer regions with more than two levels of fluorescence intensity. We found that these intensity levels occur at low temperature and are linked to the copresence of long and asymmetric chains in SMs and diphytanoyl-phosphatidylcholine in the lipid mixtures. We discuss the possible interpretations for this observation in terms of bilayer phase organization in the presence of sphingolipids. Additionally, we also show that in some cases, liposomes in the three-phase coexistence state exhibit extreme sensitivity to lateral tension. We hypothesize that the appearance of the different phases is related to the asymmetric structure of SMs and to interdigitation effects.  相似文献
