The ERG28-encoded protein, Erg28p, interacts with both the sterol C-4 demethylation enzyme complex as well as the late biosynthetic protein, the C-24 sterol methyltransferase (Erg6p)

In Saccharomyces cerevisiae, the C-24 sterol methyltransferase (Erg6p) converts zymosterol to fecosterol, an enzymatic step following C-4 demethylation of 4,4-dimethylzymosterol. Our previous study showed that an endoplasmic reticulum (ER) transmembrane protein, Erg28p, functions as a scaffold to tether the C-4 demethylation enzymatic complex (Erg25p-Erg26p-Erg27p) to the ER. To determine whether Erg28p also interacts with other ergosterol biosynthetic proteins, we compared protein levels of Erg3p, Erg6p, Erg7p, Erg11p and Erg25p in three pairs of erg28 and ERG28 strains. In erg28 strains, the Erg6p level in the ER fraction was decreased by about 50% relative to the wild-type strain, while ER protein levels of the four other ergosterol proteins showed no significant differences. Co-immunoprecipitation experiments, using an erg28 strain transformed with the epitope-tagged plasmid pERG28-HA and proteins detected with anti-HA and anti-Erg6p antibodies, indicated that Erg6p and Erg28p reciprocally co-immunoprecipitate. Further, the split ubiquitin yeast membrane two-hybrid system designed to detect protein interactions between membrane bound proteins also indicated an Erg28p-Erg6p interaction when pERG6-Cub was used as the bait and pERG28-NubG was used as the prey. We conclude that Erg28p may not only anchor the C-4 demethylation enzyme complex to the ER but also acts as a protein bridge to the Erg6p enzyme required for the next ergosterol biosynthetic step.     

Increased glyoxalase I levels inhibit accumulation of oxidative stress and an advanced glycation end product in mouse mesangial cells cultured in high glucose

Chronic high glucose levels lead to the formation of advanced glycation end-products (AGEs) as well as AGE precursors, such as methylglyoxal (MG) and glyoxal, via non-enzymatic glycation reactions in patients with diabetic mellitus. Glyoxalase 1 (GLO-1) detoxifies reactive dicarbonyls that form AGEs. To investigate the interaction between AGEs and GLO-1 in mesangial cells (MCs) under diabetic conditions, AGE levels and markers of oxidative stress were measured in GLO-1-overexpressing MCs (GLO-1-MCs) cultured in high glucose. Furthermore, we also examined levels of high glucose-induced apoptosis in GLO-1-MCs. In glomerular MCs, high glucose levels increased the formation of both MG and argpyrimidine (an MG-derived adduct) as well as GLO-1 expression. GLO-1-MCs had lower intracellular levels of MG accumulation, 8-hydroxy-deoxyguanosine (an oxidative DNA damage marker), 4-hydroxyl-2-nonenal (a lipid peroxidation product), and nitrosylated protein (a marker of oxidative-nitrosative stress) compared to control cells. Expression of mitochondrial oxidative phosphorylation complexes I, II, and III was also decreased in GLO-1-MCs. Furthermore, fewer GLO-1-MCs showed evidence of apoptosis as determined by terminal deoxynucleotidyl transferase-mediated dUTP nick labeling assay, and activation of both poly (ADP-ribose) polymerase 1 cleavage and caspase-3 was lower in GLO-1-MCs than in control cells cultured in high glucose. These results suggest that GLO-1 plays a role in high glucose-mediated signaling by reducing MG accumulation and oxidative stress in diabetes mellitus.     

Separation and purification of useful proteins using hydrogel ultrafiltration

The hydrogel process is a different form of ultrafiltration and has been used to separate biological molecules. In this study, the gel pore size was predicted by pulse NMR technique and neural network using a data base obtained from gel filtration chromatography and diffusion experiment. Recombinant alkaline phosphatase expressed in insect cells was concentrated 1.5 times by hydrogel ultrafiltration by swelling at 20°C and collapsing at 35°C at 53–65% separation efficiency and 78–83% enzyme recovery. Wild and recombinantAutographa californica nuclear polyhedrosis viruses (AcNPV) were also concentrated 1.4 and 1.6 times of the feed solution at 48.5 and 60.0% separation efficiency, respectively. Hydrogel ultrafiltration appears to be an attractive alternative for the concentration of AcNPV and recombinant proteins from insect cells.     

阿德福韦酯联合拉米夫定治疗乙型肝炎DNA阳性肝硬化疗效观察

目的：探讨阿德福韦酯联合拉米夫定治疗HBV DNA阳性乙型肝炎肝硬化的疗效。方法：46例HBV DNA阳性乙肝肝硬化患者随机分为对照组及观察组。在保肝等对症治疗基础上,观察组22例患者联用阿德福韦酯与拉米夫定,对照组24例患者予阿德福韦酯,总疗程均为48周。结果：在治疗12周后,观察组与对照组HBV DNA转阴率分别为54.5%、20.8%（P〈0.05）,ALT复常率分别为63.6%、33.3%（P〈0.05）。治疗24周、48周后上述指标无统计学差异。两组患者未见明显药物不良反应。结论：阿德福韦酯联合拉米夫定治疗HBV DNA阳性乙型肝炎肝硬化起效快,降低病毒载量疗效佳,安全性好。     

Synthesis and Activity Evaluation of Nasopharyngeal Carcinoma Inhibitors Based on 6‐(Pyrimidin‐4‐yl)‐1H‐indazole

Human nasopharyngeal carcinoma is a common head and neck malignancy with high incidence in Southeast Asia and Southern China. It is necessary to develop safe, effective and inexpensive anticancer agents to improve the therapeutics of patients with nasopharyngeal carcinoma. A series of small molecular compounds based on 6‐(pyrimidin‐4‐yl)‐1H‐indazole were synthesized and evaluated for antiproliferative activities against human nasopharyngeal carcinoma cell lines SUNE1. Compounds 6b , 6c , 6e and 6l showed potent antiproliferative activities similar to positive control drug cisplatin in vitro with lower nephrotoxicity than it. N‐[4‐(1H‐Indazol‐6‐yl)pyrimidin‐2‐yl]benzene‐1,3‐diamine ( 6l ) was selected for further study. It was found that 6l induced mitochondria‐mediated apoptosis and G₂/M phase arrest in SUNE1 cells. Furthermore, compound 6l at 10 mg/kg can suppress the growth of an implanted SUNE1 xenograft with a TGI% (tumor growth inhibition) value of 50 % and did not cause serious side effects in BALB/c nude mice. This study suggests that 6‐(pyrimidin‐4‐yl)‐1H‐indazole derivatives are a series of small molecule compounds with anti‐nasopharyngeal carcinoma activities.     

具有抑菌作用的放线菌GNF1的分离鉴定和生物学活性的研究

从几种复合微生物有机肥中分离出一系列不同的菌株,与实验室保存的菌株GNW(自生固氮菌)和HP2(解磷菌)混合接种培养,检测是否因基因杂交、突变等原因而产生具有抑菌作用的菌株.结果分离出一株具有抑菌作用的放线菌菌株GNF1,根据其形态学特征、生理生化特征和基于16S rRNA基因序列的系统发育分析结果,鉴定其属于链霉菌属(Streptomyces)的一个菌株,GNF1菌株的代谢产物中存在具有抑菌作用的活性成分,与植物根际促生菌GNW、HP2以及某些原核病原微生物共培养培养时能明显抑制它们的生长.     

Temperature-dependent post-diapause development and prediction of spring emergence of the pine needle gall midge (Dipt., Cecidomyiidae)

Abstract:  We determined the influence of temperature on post-diapause development of overwintered Thecodiplosis japonensis Uchida et Inouye (Dipt., Cecidomyiidae) under various treatments (12, 15, 18, 21, 24, 27 and 30°C) in an effort to predict its spring emergence. Survival and developmental period for the overwintered larvae and pupae were significantly influenced by temperature. Linear and nonlinear regression models quantitatively described temperature-dependent development and survival of T. japonensis . The survival models exhibited right-skewed bell shape patterns for all stages, indicating a more detrimental impact on survival at high temperatures. Theoretical optimum temperatures with highest survival were 22.3, 24.0 and 24.0°C for the overwintered larvae, pupae and total post-diapause development (the larvae to adults) respectively. Pupal mortality was higher at all temperatures than larval mortality and the suitable range of temperature for pupae was narrower than that of larvae. The nonlinear Briere model estimated that optimum temperatures with the fastest development were 29.1°C for larvae, 27.6°C for pupae and 27.0°C for larvae to adults. In a linear model, the lower threshold temperatures were 5.1, 7.1 and 5.9°C for larvae, pupae, and larvae to adults respectively. A predictive degree-day model was developed using trap catches of T. japonensis adult emergence during 1991–1995. The model accounted for 84.6% of year-to-year variation in adult emergence and predicted accurately the median emergence time in 1996.     

Effect of radiomodifiers on lipid peroxidation and the structural-functional state of irradiated mitochondria

SH-Containing radioprotective agents, for instance, cysteine, cystamine, dithiothreitol, exert an antioxidant effect on gamma-radiation-induced lipid peroxidation in mitochondrial membranes and, concurrently, uncouple oxidation from phosphorylation. A radiosensitizer, N-ethylmaleimide, on the contrary, being a pro-oxidant of lipid peroxidation couples oxidation and phosphorylation. At the same time, changes in the lipid peroxidation system and in the energy production processes, observed after irradiation of a mitochondrion suspension containing modifiers, are accompanied by the increased destruction of mitochondrial membranes as compared to irradiated controls.
