When rat liver mitochondria are treated with tert-butyl hydroperoxide (TBHP) in the presence of the spin trap 5,5-dimethyl-1-pyrroline N-oxide (DMPO), electron paramagnetic resonance (EPR) signals are detected attributable to spin adducts resulting from the trapping of methyl, tert-butoxyl, and tert-butylperoxyl radicals. The addition of respiratory substrate results in a 3- to 7.5-fold increase in the signal intensity of the DMPO/methyl adduct, no change in the signal intensity of the DMPO/tert-butoxyl adduct, and complete loss of the DMPO/tert-butylperoxyl adduct signal. The magnitude of increase of methyl radical production in the presence of respiratory substrate is related to the respiratory control ratio (RCR) of the mitochondrial preparation. In the presence of antimycin A, which blocks electron flow between cytochromes b and c1, no stimulation of methyl radical production is detected with respiratory substrate. Stimulation of methyl radical production by the addition of respiratory substrate is detected in cytochrome c-depleted mitochondria. A similar increase in methyl radical production is detected when ferrous cytochrome c is treated with TBHP in the presence of DMPO (as compared to when ferricytochrome c is used). These results indicate that TBHP is reduced directly by either cytochrome c1, cytochrome c, or by both of these electron transport chain components in mitochondria undergoing state 4 respiration. 相似文献

Ovaries were obtained from 78 new-born lambs (12 singletons, 25 twins, 28 triplets, 10 quadruplets and 3 quintuplets) from flocks selected for (T902, T903 and Booroola) or against (O) multiple births. Sections of the ovaries were examined with a projection microscope and the numbers of all types of follicles were estimated. There were no differences between genetic lines in the number of primordial follicles, after adjustment for litter size and sire; however, there were significantly more of these follicles in single-born lambs than in lambs born in litters of two or more within genetic lines. The population biology of Camallanus lacustris (Zoega) and the status of the eel, Anguilla anguilla (Linnaeus), as its definitive host have been studied in a small Devon lake. No clear seasonal pattern in prevalence and abundance was observed, and recruitment of the new generation may occur in all seasons. However, reproduction exhibits some seasonality as adult nematodes were the commonest stage in the parasite population from early summer onwards and production of first-stage larvae appeared to occur primarily during this period. These larvae were viable, and shown experimentally to be infective to copepods. The occurrence of the nematode and its development to full maturity in eels, the absence of its typical definitive host ( Perca fluviatilis Linnaeus) from the lake and the absence of the nematode from other species of fish in the lake indicate that eels serve as the only, true definitive host of the nematode in this locality. It is suggested that account must be taken of this alternative host when the population of C. lacustris is investigated in the future. 相似文献

The changes in propionate metabolism that accompany cobalt deficiency in sheep are described. Two groups of sheep, fed either a cobalt sufficient or deficient diet, were given an iv propionate load at intervals during a 14 w experiment. There was a tendency towards increased propionate half-life as the animals became cobalt deficient. However, significant changes in the area under the plasma methylmalonic acid-time curve occurred very early, indicating significant impairment of propionate metabolism. Despite this, the area under the plasma glucose-time curve was unaffected by cobalt deficiency, suggesting that the impairment of propionate metabolism, although significant, is not extensive. 相似文献
