Detailed information of human B cell activation via TLR may lead to a better understanding of B cell involvement in autoimmunity and malignancy. In this study we identified a fundamental difference in the regulation of TLR7- and TLR9-mediated B cell stimulation: whereas the induction of polyclonal naive B cell proliferation by the TLR7 ligands resiquimod (R848) and loxoribine required the presence of plasmacytoid dendritic cells (PDCs), activation via the TLR9 ligand CpG was independent of PDCs. We found that PDC-derived type I IFN enhanced TLR7 sensitivity of B cells by selectively up-regulating TLR7 expression. In contrast the expression levels of TLR9 and of other TLRs studied remained unchanged. In the presence of type I IFN, TLR7 ligation triggered polyclonal B cell expansion and B cell differentiation toward Ig-producing plasma cells; notably, this occurred independently of T cell help and B cell Ag. Human B cells did not respond to ligands of other TLRs including TLR2, TLR4 and TLR6 with and without type I IFN. In conclusion, our results reveal a distinct regulation of TLR7 and TLR9 function in human B cells and highlight TLR7 and TLR9 as unique targets for therapeutic intervention in B cell-mediated immunity and disease. 相似文献

The phytochromes are the best studied plant photoreceptors, controlling a wide variety of responses at both whole plant and
single cell levels. Three signal transduction pathways, dependent on cGMP and/or calcium, have been found to be utilized by
phytochrome to control the expression of genes required for chloroplast development (e.g., CAB and FNR) and anthocyanin biosynthesis (e.g., CHS). In particular, cGMP is a second messenger positively regulating CHS gene expression whilst calcium and calmodulin act as negative regulators. In addition to phytochrome regulation of CHS we have begun to examine the signal transduction pathways utilized by UV photoreceptors. In contrast to phytochrome-mediated

The biotrophic smut fungus Ustilago maydis infects all aerial organs of maize (Zea mays) and induces tumors in the plant tissues. U. maydis deploys many effector proteins to manipulate its host. Previously, deletion analysis demonstrated that several effectors have important functions in inducing tumor expansion specifically in maize leaves. Here, we present the functional characterization of the effector See1 (Seedling efficient effector1). See1 is required for the reactivation of plant DNA synthesis, which is crucial for tumor progression in leaf cells. By contrast, See1 does not affect tumor formation in immature tassel floral tissues, where maize cell proliferation occurs independent of fungal infection. See1 interacts with a maize homolog of SGT1 (Suppressor of G2 allele of skp1), a factor acting in cell cycle progression in yeast (Saccharomyces cerevisiae) and an important component of plant and human innate immunity. See1 interferes with the MAPK-triggered phosphorylation of maize SGT1 at a monocot-specific phosphorylation site. We propose that See1 interferes with SGT1 activity, resulting in both modulation of immune responses and reactivation of DNA synthesis in leaf cells. This identifies See1 as a fungal effector that directly and specifically contributes to the formation of leaf tumors in maize. 相似文献

In South Africa, a plan was launched to manage separate sub-populations of endangered African wild dogs (Lycaon pictus) in several small, geographically isolated, conservation areas as a single meta-population. This intensive management approach
involves the re-introduction of wild dogs into suitable conservation areas and periodic translocations among them. To assess
the attitudes towards re-introduced wild dogs, we conducted a questionnaire survey of multiple stakeholders—local community
members, private landowners and tourists—in and around Hluhluwe-iMfolozi Park (HiP), one of the meta-population conservation
areas. Here, we document conflicting human interests over the re-introduced wild dogs. Tourists in HiP, on the one hand, expressed
overwhelmingly positive opinions about wild dogs across personal details of the respondents, but especially after having seen
free-ranging wild dogs. On the other hand, we found misconceptions and perceptions that were more negative among the rural
population around HiP, again largely independent of personal details of the participants, although educated respondents voiced
more favourable views of wild dogs. These negative attitudes were in particular due to perceived and real threats of livestock
losses. In a follow-up questionnaire survey, we also discovered apparent shortcomings of a previous short-lived conservation
education programme among the local communities adjacent to HiP. Consequently, the mitigation of the conflict between wild
dogs and rural people requires an understanding of the conditions under which livestock predation occurs, the encouragement
of practices that prevent such predation, and increasing local tolerance of co-existence with wild dogs through both economic
and non-monetary incentive schemes as well as continued conservation education. 相似文献

Integrin-extracellular matrix interactions play important roles in the coordinated integration of external and internal cues that are essential for proper development. To study the role of beta1 integrin in the mammary gland, Itgbeta1(flox/flox) mice were crossed with WAPiCre transgenic mice, which led to specific ablation of beta1 integrin in luminal alveolar epithelial cells. In the beta1 integrin mutant mammary gland, individual alveoli were disorganized resulting from alterations in cell-basement membrane associations. Activity of focal adhesion kinase (FAK) was also decreased in mutant mammary glands. Luminal cell proliferation was strongly inhibited in beta1 integrin mutant glands, which correlated with a specific increase of p21 Cip1 expression. In a p21 Cip1 null background, there was a partial rescue of BrdU incorporation, providing in vivo evidence linking p21 Cip1 to the proliferative defect observed in beta1 integrin mutant glands. A connection between p21 Cip1 and beta1 integrin as well as FAK was also established in primary mammary cells. These results point to the essential role of beta1 integrin signaling in mammary epithelial cell proliferation. 相似文献

Improvements in the ability to hand‐rear birds in captivity have aided zoological institutions in the sustainable management of these species, and have provided opportunities to examine their physical growth in varying conditions. Monitoring the weight gain and development of chicks is an important aspect of developing a hand‐rearing protocol. In this paper we provide the institutional history for a colonial species of passerine, the Taveta golden weaver, at Disney's Animal Kingdom®, in order to demonstrate the methods of establishing a successful breeding program which largely incorporates hand‐rearing in management of the population. We also tested if we could accurately predict sex of chicks using weights collected on Day 14 during the hand‐rearing process. Using this tool, we were able to correctly determine sex before fledging in more than 83% of chicks. Early sex determination is important in captive species for genetic management and husbandry purposes. While genetic sexing can be expensive, we found that using growth curves to determine sex can be a reliable and cost‐effective tool for population management of a colonial passerine.
