Correct 3' processing of chloroplast precursor mRNAs (pre-mRNAs) requires a stem-loop structure within the 3' untranslated region. In spinach, a stable 3' stem-loop-protein complex has been shown to form in vitro between petD pre-mRNA, encoding subunit IV of the cytochrome b6/f complex, and chloroplast proteins. This complex contains three chloroplast stem-loop binding proteins (CSPs), namely, CSP29, CSP41, and CSP55. Here, we report the purification of CSP41 and cloning of the csp41 gene and show that CSP41 is encoded by a single nuclear gene. Characterization of bacterially expressed CSP41 demonstrates that this protein binds specifically to the 3' stem-loop structure and a downstream AU-rich element of petD pre-mRNA and that its binding affinity is enhanced by associating with CSP55. Our data also show that CSP41 has substantial nonspecific endoribonuclease activity. These data suggest that CSP41 could be involved in 3' processing of petD pre-mRNA and/or in RNA degradation. The fact that different reaction conditions favor RNA binding over ribonuclease activities suggests a possible mode of in vivo regulation. 相似文献

Ormia depleta(Wiedemann) (Diptera: Tachinidae), a South American fly that homes on the calling songs of its hosts, was brought from Piracicaba, Brazil; colonized; and released in Florida for the biological control ofScapteriscusspp. mole crickets. It became established at some release sites and rapidly spread through most of peninsular Florida. The seasonal distributions of phonotactic females in Florida differed from the single summer peak recorded at Rio Claro and Piracicaba, Brazil (23° S). Near Bradenton (27° N), spring and fall peaks with a summer hiatus were quickly established and remain evident. At two sites near Gainesville (30° N), a strong fall peak and a modest-to-none spring peak developed. Peaks of phonotactic females seem to follow times of peak host availability and sometimes coincide with minima of host availability. Limits to geographical expansion ofO. depletaare poorly understood. At Gainesville, the site of earliest introduction, establishment was rapid and soundtrap catches increased for the first 3 years, exceeding 1000 per year at one site. Catches then declined for 3 years, almost reaching zero in 1994. On the other hand, annual sound-trap catches ofO. depletaat Bradenton exceeded 1000 in <2 years and have remained above that level for 5 years. Winters in Gainesville are more severe than those in Bradenton, but minimum winter temperatures in Gainesville were substantially lower duringO. depleta's increase than during its decline. Perhaps mild winters cause the flies to become active early—only to starve for lack of adequate winter nectar sources.O. depletamay yet spread to the northern limits of itsScapteriscushosts as present populations adapt to new environments. Bovine pancreatic ribonuclease A (RNase A) catalyzes the cleavage of the P-O(5') bond in RNA. Although this enzyme has been the object of much landmark work in bioorganic chemistry, the nature of its rate-limiting transition state and its catalytic rate enhancement had been unknown. Here, the value of k(cat)/K(m) for the cleavage of UpA by wild-type RNase A was found to be inversely related to the concentration of added glycerol. In contrast, the values of k(cat)/K(m) for the cleavage of UpA by a sluggish mutant of RNase A and the cleavage of the poor substrate UpOC(6)H(4)-p-NO(2) by wild-type RNase A were found to be independent of glycerol concentration. Yet, UpA cleavage by the wild-type and mutant enzymes was found to have the same dependence on sucrose concentration, indicating that catalysis of UpA cleavage by RNase A is limited by desolvation. The rate of UpA cleavage by RNase A is maximal at pH 6.0, where k(cat) = 1.4 × 10(3) s(-1) and k(cat)/K(m) = 2.3 × 10(6) M(-1)s(-1) at 25°C. At pH 6.0 and 25°C, the uncatalyzed rate of [5,6-(3)H]Up[3,5,8-(3)H]A cleavage was found to be k(uncat) = 5 × 10(-9) s(-1) (t(1/2) = 4 years). Thus, RNase A enhances the rate of UpA cleavage by 3 × 10(11)-fold by binding to the transition state for P-O(5') bond cleavage with a dissociation constant of <2 × 10(-15) M. 相似文献

We have characterized a mitochondrial gene in Oenothera, designated orf454, capable of encoding a component of the cytochrome c biogenesis system. This open reading frame is interrupted by an intron of 941 nucleotides showing high similarity to a group II intron residing in the rpl2 gene. RNA editing, which is observed at 18 cytidine positions within the orf454 reading frame, improves the similarity to protein-coding sequences in bacteria and higher plants and removes the last 16 amino acids. orf454 also shows high sequence similarity to two overlapping reading frames (orf169 and orf322) of Marchantia mitochondria. These ORFs belong to an operon-like cluster of genes in the liverwort that is not conserved in Oenothera mitochondria. However, in bacteria these reading frames are organized like the Marchantia gene cluster. It has been shown by genetical analysis in Rhodobacter capsulatus that these genes are essential for cytochrome c biogenesis. Genes of bacterial operons — ccl1 in Rhodobacter and yejR and nrfE in Escherichia coli — show high sequence similarity to the mitochondrial reading frames orf577 and orf454 of Oenothera. orf454, which we describe here, is homologous to the C-terminal region of these bacterial genes, while the previously described orf577 is homologous to the N-terminal region. 相似文献

对水稻卷叶品种流岗卷叶粳与4个平展叶品种及1个卷叶标志基因(rl3)系的杂交或回交后代进行了考察。结果表明,流岗卷叶粳的卷叶特性以单基因不完全显性方式遗传; 该基因与rl3基因不等位,当rl3位点处于隐性纯合时两者以累加方式发生互作。这一等位基因可作标志基因使用,暂定名为Rl(t)。
Abstract:Liugangjuanyejing is a rolled leaf mutant of rice.A genetic study on the rolled-leaf character was carried out by crossing it with four flat-leaf cultivars and a genetic marker line (with a rolled-leaf allele rl3).The results showed that this character was controlled by an incomplete dominant gene which was non-allelic to rl3 locus and that there existed additive effect between the two loci when the rl3 locus was homozygous recessiveness.This new rolled-leaf allele was provisionally named as Rl(t) and could be used as a genetic marker of rice. 相似文献
