The fluid dynamic design and characterization of a low mechanical stress agitator and bioreactor vessel for large-scale bioprocessing using anchorage dependent mammalian cells is considered. The complex and fragile nature of mammalian cells puts stringent constraints on the design of agitators for stirred tank bioreactors. Traditional agitators have difficulty meeting the competing requirements of fluidization and mixing while maintaining low enough mechanical stress levels to avoid cell damage. A rotating disc agitator design is proposed. Flow visualization and laser Doppler velocimeter measurements reveal fluidization of microcarriers with a gentle (low turbulence level), highly three-dimensional flow characterized by good mixing at low hydrodynamic shear stress levels. 相似文献

Summary The marked binding of antibodies specific for 5-methylcytidine to the short arm of chromosome 15 distinguishes this chromosome from the other human acrocentrics. This method has been used to study over 60 individuals including 12 who did not have Down's syndrome, but who did have an extra G-group sized acrocentric chromosome. In six cases the extra chromosome did not show intensive binding of anti-5-methylcytidine. In the other six cases, the extra chromosome contained a 5-methylcytidine rich band at each end indicating that both ends were derived from chromosome 15 and contained centromeric heterochromatin normally present on the short arm of chromosome 15. The duplication of short arm material in the abnormal chromosomes was confirmed in all cases by quinacrine staining, nucleolar organizer (Ag-AS) staining or C-banding. In three cases, the abnormal chromosome appeared to arise from two different chromosomes 15. Several possible mechanisms for the production of the abnormal chromosome are discussed. A 10-Hz frequency sound caused flight or avoidance responses in juvenile spring chinook salmon Oncorhynchus tshawytscha and rainbow trout O. mykiss . Groups of fish were placed in 3-m diameter circular tanks with a water depth of 1 m. The sound source was a 25-cm diameter aluminium tube with a piston in one end. The piston was driven back and forth by an eccentric coupling to an electric motor at a frequency of 10 Hz and with peak to peak amplitude of 4 cm. The sound source was turned on for 5 s when the fish was within 1 m. Initial tests always resulted in a strong flight response, but after three to four tests the fish more typically simply swam away as far as possible from the source. A highly sensitive method for the detection of in vivo induction of sister-chromatid exchange (SCE) has been developed in mice subjected to partial hepatectomy. SCE induction by either acetylaminofluorene (AAF) or cyclophosphamide, drugs requiring metabolic activation, is significantly greater in both regenerating liver and bone-marrow cells of partial hepatectomized animals than in marrow cells of unhepatectomized mice. These experiments have confirmed the ability of AAF, a well known mutagen-carcinogen, to induce SCE formation, even though the cytogenic effects of this drug on non-hepatectomized mice is very small. The in vivo system described has demonstrated the influence of the liver on drug-induced damage to extra-hepatic tissues. The procedures developed should facilitate the detection of drug-induced cytogenic damage and permit the comparison of inter-tissue differences in SCE induction with tissue-specific differences in drug-activation pathways. 相似文献

Levels of oestrogens in plasma of mature fall-spawning rainbow trout Salmo gairdneri were determined using a radio-immunoassay. No significant difference was found in oestrogen concentration between the sexes (male =2.5 ng/ml; female =4.4 ng/ml); between individual variability was great. Four blood samplings over a 24-h period via cardiac puncture of males revealed no diel variation or change in estrogen levels due to the stress of bleedings. Although no gonado-somatic index-estrogen relationship could be demonstrated for either sex, there was correlation between oestrogen and androgen levels in the female. 相似文献

A fundamental requirement for cellular vitality is the maintenance of plasma ion concentration within strict ranges. It is the function of the kidney to match urinary excretion of ions with daily ion intake and nonrenal losses to maintain a stable ionic milieu. NADPH oxidase is a source of reactive oxygen species (ROS) within many cell types, including the transporting renal epithelia. The focus of this review is to describe the role of NADPH oxidase-derived ROS toward local renal tubular ion transport in each nephron segment and to discuss how NADPH oxidase-derived ROS signaling within the nephron may mediate ion homeostasis. In each case, we will attempt to identify the various subunits of NADPH oxidase and reactive oxygen species involved and the ion transporters, which these affect. We will first review the role of NADPH oxidase on renal Na(+) and K(+) transport. Finally, we will review the relationship between tubular H(+) efflux and NADPH oxidase activity. 相似文献

We describe the migration distances and timing of the adult Pacific lamprey, Entosphenus tridentatus, in the Willamette River Basin (Oregon, U.S.A.). We conducted aerial surveys to track radio-tagged fish upstream of a major
waterfall and hydropower complex en route to spawning areas. We detected 24 out of the 43 fish that passed the waterfall-hydropower
complex. Of the detected fish, 17 were detected multiple times. Their maximum migration distance upstream in the mainstem
Willamette approximated a normal distribution. The maximum distance migrated upstream did not significantly correlate with
total body length (r = −0.186, P = 0.385) or date that the fish passed Willamette Falls (r = −0.118, P = 0.582). Fish migrated primarily during the spring to early summer period before stopping during the summer, when peak river
temperatures (≥20°C). However, at least three fish continued to migrate upstream after September. Behavior ranged from relatively
slow migration, followed by holding; to rapid migration, followed by slow migration further up in the basin. This study provides
