Vi capsular polysaccharide production is encoded by the viaB locus, which has a limited distribution in Salmonella enterica serovars. In S. enterica serovar Typhi, viaB is encoded on a 134-kb pathogenicity island known as SPI-7 that is located between partially duplicated tRNA(pheU) sites. Functional and bioinformatic analysis suggests that SPI-7 has a mosaic structure and may have evolved as a consequence of several independent insertion events. Analysis of viaB-associated DNA in Vi-positive S. enterica serovar Paratyphi C and S. enterica serovar Dublin isolates revealed the presence of similar SPI-7 islands. In S. enterica serovars Paratyphi C and Dublin, the SopE bacteriophage and a 15-kb fragment adjacent to the intact tRNA(pheU) site were absent. In S. enterica serovar Paratyphi C only, a region encoding a type IV pilus involved in the adherence of S. enterica serovar Typhi to host cells was missing. The remainder of the SPI-7 islands investigated exhibited over 99% DNA sequence identity in the three serovars. Of 30 other Salmonella serovars examined, 24 contained no insertions at the equivalent tRNA(pheU) site, 2 had a 3.7-kb insertion, and 4 showed sequence variation at the tRNA(pheU)-phoN junction, which was not analyzed further. Sequence analysis of the SPI-7 region from S. enterica serovar Typhi strain CT18 revealed significant synteny with clusters of genes from a variety of saprophytic bacteria and phytobacteria, including Pseudomonas aeruginosa and Xanthomonas axonopodis pv. citri. This analysis suggested that SPI-7 may be a mobile element, such as a conjugative transposon or an integrated plasmid remnant.  相似文献

Electron nuclear double resonance (ENDOR) was performed on the protein-bound, stabilized, high-affinity ubisemiquinone radical, QH*-, of bo3 quinol oxidase to determine its electronic spin distribution and to probe its interaction with its surroundings. Until this present work, such ENDOR studies of protein-stabilized ubisemiquinone centers have only been done on photosynthetic reaction centers whose function is to reduce a ubiquinol pool. In contrast, QH*- serves to oxidize a ubiquinol pool in the course of electron transfer from the ubiquinol pool to the oxygen-consuming center of terminal bo3 oxidase. As documented by large hyperfine couplings (>10 MHz) to nonexchangeable protons on the QH*- ubisemiquinone ring, we provide evidence for an electronic distribution on QH*- that is different from that of the semiquinones of reaction centers. Since the ubisemiquinone itself is physically nearly identical in both QH*- and the bacterial photosynthetic reaction centers, this electronic difference is evidently a function of the local protein environment. Interaction of QH*- with this local protein environment was explicitly shown by exchangeable deuteron ENDOR that implied hydrogen bonding to the quinone and by weak proton hyperfine couplings to the local protein matrix.  相似文献

Cells capable of photosynthesis in the parasitic angiosperm Cuscuta reflexa Roxb. (dodder) are highly localized. Immunolocalization of ribulose-1,5 bisphosphate carboxylase-oxygenase (Rubisco) and autofluorescence of chlorophyll in transverse sections of stems showed that they were largely restricted to a band of cells adjacent to the vascular bundles, consequently, the concentrations of Rubisco and chlorophyll were low per unit area or fresh weight. When 14CO2 was supplied to stem segments of C. reflexa it preferentially accumulated in these cells adjacent to the vasculature. Although the conductance for CO2 movement to the cells containing chlorophyll and Rubisco was very low, both the light reactions and dark reactions of photosynthesis appeared to be functional. De-epoxidation of the xanthophyll-cycle pigments after exposure to high light, and the chlorophyll fluorescence parameters, photochemical quenching (qP), non-photochemical quenching (NPQ) and the quantum efficiency of photosystem II (φPSII) responded normally to changes in photon flux density, indicating functional light-driven electron transport. The response of CO2 exchange to photon flux density followed a typical hyperbolic curve, and positive rates of CO2 fixation occurred when external CO2 was increased to 5%. We propose that CO2 for carbon assimilation is derived from internally respired CO2 and that this layer of photosynthetic cells makes a positive contribution to the carbon budget of C. reflexa. The influence of vesicular–arbuscular mycorrhizal (M) colonization on biomass production and photosynthesis of Trifolium repens L. was investigated in two experiments in which the foliar nitrogen and phosphorus contents of non-mycorrhizal (NM) plants were manipulated to be no lower than that of M plants. Throughout both experiments there was a stimulation in the rate of CO2 assimilation of the youngest, fully expanded leaf of M compared with NM plants. In addition, M plants exhibited a higher specific leaf area compared with NM plants, a response that maximized the area available for CO2 assimilation per unit of carbon (C) invested. Despite the increased rate of photosynthesis in M plants there was no evidence that the additional C gained was converted to biomass production of M plants. It is suggested that this additional C gained by colonized plants was allocated to the mycorrhizal fungus and that it is the fungus, by acting as a sink for assimilates, that facilitated the stimulation in the rate of photosynthesis of the plant partner.  相似文献

Lake Okaro is a small, warm monomictic lake in central North Island, New Zealand, which progressed from oligotrophic to eutrophic through the 1960s. Trends in phosphorus (P) concentrations in the lake are linked to multiple restoration efforts over a 5-year period (2003–2008). The restoration procedures include a 2.3 ha constructed wetland established in February 2006 and riparian margin protection to reduce external loading, as well as an Alum application in December 2003 and sediment capping using modified zeolite in September 2008 to reduce internal loading. The annual average total phosphorus (TP) concentration in the lake decreased by 41% from 2004–2005 to 2007–2008. Two predictive models based on external P loading data generally underestimated the measured TP concentrations in the water column due to internal P loading. The relatively rapid response of TP concentrations after reduction of the internal loading using modified zeolite suggests that this technique can effect a rapid decrease in lake water TP concentrations though the trophic state of Lake Okaro showed high resilience to the reduced P loading. It is concluded that the combined effect of all restoration procedures resulted in a relatively rapid decrease in TP concentrations in Lake Okaro, which may be prolonged by continued external load reduction.  相似文献

In East African small-holder farming of maize, the cattle forage legume, Desmodium uncinatum is used as an intercrop due to its allelopathic inhibition of parasitism by Striga hermonthica, an obligate parasitic weed that can devastate the maize crop. Bioassay-guided fractionation of the root extract of D. uncinatum revealed isoschaftoside to be the main compound in the most potent fraction inhibiting growth of germinated S. hermonthica radicles. Bioassays repeated with isoschaftoside isolated from a different plant source, Passiflora incarnata, proved it to be a biologically active component. Analysis of the root exudates produced by hydroponically grown D. uncinatum showed isoschaftoside to be present in the hydroponic media at biologically active concentrations of 10–100 nM.  相似文献

In June 2008 and 2009, cover, density, and species diversity were measured on two areas of the prairie at the U. S. Department of Energy Weldon Spring Site to begin quantification of the prairie establishment and the effects of a prairie burn. Sampling began by testing for the most appropriate transect length (cover) and quadrat size (density) for quantification of vegetation. Total cover increased in the first growing season after burning. Conversely, total cover decreased in the unburned area in one year. The trend in litter cover is the opposite with litter decreasing after burning, but increasing in one year in the unburned area. Bare ground decreased in one year in the unburned area, but was unchanged after burning. Species diversity tripled after fire, but was unchanged in one year in the unburned area. The results show that litter and fire both affect plant cover. If land reclamation activities are to be an integral part of hazardous waste remediation at contaminated sites, then the success of reclamation efforts needs to be quantified along with success criteria for waste remediation of the sites. The results show that plant cover can be easily quantified, but that density measures are more biased which makes it more difficult to achieve adequate sample size for plant density.  相似文献

The limitations of Community Based Natural Resource Management (CBNRM) with respect to the difficulties of comparing local versus scientific knowledge categories within a bounded definition of ‘community’ were investigated by means of a study exploring local indigenous knowledge pertaining to harvesting technique, and the impact of soil and species type on the post-harvest coppice response of popular savanna fuelwood species, among rural inhabitants of the Bushbuckridge region of the Limpopo Province, South Africa. Soils and plants were evaluated chiefly in terms of their perceived ability to retain precipitation, making rainfall a driving force in local understanding of environmental productivity. Some indigenous knowledge showed an agreement with biological data, but overall the variability in responses, as well as the diverse scales at which indigenous and scientific knowledge is directed, were too great to allow for simplistic parallels between local ecological indices to be made. Indigenous environmental knowledge was underscored by the perceived symbolic link between environmental and social degradation. It is recommended that environmental managers incorporate indigenous knowledge as a component of a systems-level approach to natural resource management, where biological, cultural, economic, and symbolic aspects of natural resource use are nested within a broader ecosocial system. This approach to indigenous knowledge is offered as an alternative to the simple scientific evaluation that so often characterizes environmental management.  相似文献
