Flowering phenology, dynamics of seed development and reproductive success of Saxifraga moschata Wulfen, occurring from the alpine to the nival zone, was investigated in three climatically varying years on early- and late-thawing sites at 2340 m a.s.l. in the Austrian Alps. The growing season was particularly short in 2001, about normal in 2002 and exceptionally long and warm in 2003. S. moschata is a mid-season flowering species, which started to bloom about 1 month after snowmelt. From onset of anthesis until seed maturity an individual flower required 6–7 weeks, and all individuals of a site 9–11 weeks. In 2001, late flowering individuals lost about 40% of the seed crop because of too early snowfalls in autumn. No clear relationship could be found between thermal time and the length of different reproductive phases, which indicates that temperature was not a limiting factor for reproductive development in the growing seasons 2001–2003.

Plants from the earliest-thawing site produced about double the reproductive structures (reproductive shoots per individual, number of flowers per inflorescence, number of ovules per ovary), and had a significantly higher reproductive output (number of fruits and mature seeds per inflorescence) compared with individuals from the late-thawing site. Reproductive success was additionally affected by the seasonal climate and was lowest in the warm and dry summer of 2003. Fruit/flower ratio ranged between 0.6 and 0.8, seed/ovule ratio between 0.4 and 0.5 and relative reproductive success (RRS) between 0.3 and 0.4, depending on the site and year.

Different from our expectations, it took S. moschata relatively long to undergo reproductive development; however, its phenological plasticity and ability to continue seed maturation even under snow are most advantageous for a life at higher altitudes.  相似文献

Top predators are known to play an important role in the assembly of communities via two mechanisms: (1) by altering the colonization (or emigration) patterns of prey through behavioral habitat selection, and (2) by altering vital rates (e.g. mortality, birth) of prey after colonization. While both these mechanisms act to determine assembly, research has focused on either their combined overall effects (confounding them), or examined them singly. As a result, it remains unclear how these mechanisms act to sequentially shape community structure. In this study, we experimentally disentangle habitat selection and post‐colonization effects of predaceous fish to test their independent and combined influence on the assembly of insect and larval amphibian communities in experimental freshwater habitats. Specifically, we ask, ‘do the behavioral choices of colonists continue to structure aquatic communities even after post‐colonization processes have occurred?’ Like previous studies, we found that colonization was strongly reduced by the presence of fish cues. More importantly, these effects of fish on prey colonization behavior combined independently with post‐colonization processes to determine the overall effect of predators on community assembly. Although habitat selection and predation both reduced abundance and biomass of most taxa in the post‐colonization communities, these factors had qualitatively different effects on aspects of trophic structure. Habitat selection altered the ratio of secondary to primary consumer abundance and biomass, while post‐colonization predation drove strong trophic cascades not observed in response to habitat selection. Our results suggest that behavioral choices regarding habitat selection can have lasting and unique effects on the structure of aquatic communities.  相似文献

Due to the presence of moisture and nutrients, brewery filling line surfaces are susceptible to unwanted microbial attachment. Knowledge of the attaching microbes will aid in designing hygienic control of the process. In this study the bacterial diversity present on brewery filling line surfaces was revealed by next generation sequencing. The two filling lines studied maintained their characteristic bacterial community throughout three sampling times (13–163 days). On the glass bottle line, γ-proteobacteria dominated (35–82% of all OTUs), whereas on the canning line α-, β- and γ-proteobacteria and actinobacteria were most common. The most frequently detected genera were Acinetobacter, Propinobacterium and Pseudomonas. The halophilic genus Halomonas was commonly detected, which might be due to its tolerance to alkaline foam cleaners. This study has revealed a detailed overall picture of the bacterial groups present on filling line surfaces. Further effort should be given to determine the efficacy of washing procedures on different bacterial groups.  相似文献

Enteric neural crest-derived cells (ENCCs) migrate along the intestine to form a highly organized network of ganglia that comprises the enteric nervous system (ENS). The signals driving the migration and patterning of these cells are largely unknown. Examining the spatiotemporal development of the intestinal neurovasculature in avian embryos, we find endothelial cells (ECs) present in the gut prior to the arrival of migrating ENCCs. These ECs are patterned in concentric rings that are predictive of the positioning of later arriving crest-derived cells, leading us to hypothesize that blood vessels may serve as a substrate to guide ENCC migration. Immunohistochemistry at multiple stages during ENS development reveals that ENCCs are positioned adjacent to vessels as they colonize the gut. A similar close anatomic relationship between vessels and enteric neurons was observed in zebrafish larvae. When EC development is inhibited in cultured avian intestine, ENCC migration is arrested and distal aganglionosis results, suggesting that ENCCs require the presence of vessels to colonize the gut. Neural tube and avian midgut were explanted onto a variety of substrates, including components of the extracellular matrix and various cell types, such as fibroblasts, smooth muscle cells, and endothelial cells. We find that crest-derived cells from both the neural tube and the midgut migrate avidly onto cultured endothelial cells. This EC-induced migration is inhibited by the presence of CSAT antibody, which blocks binding to β1 integrins expressed on the surface of crest-derived cells. These results demonstrate that ECs provide a substrate for the migration of ENCCs via an interaction between β1 integrins on the ENCC surface and extracellular matrix proteins expressed by the intestinal vasculature. These interactions may play an important role in guiding migration and patterning in the developing ENS.  相似文献

A micromodification for protein determination in tissue material using Amido black 10 B is described. Compared with the method of LOWRY et al. it requires a comparable time expenditure, but has three principal advantages: 1) it is 5-10fold more sensitive; 2) the calibration curve is linear over a virtually unlimited range; 3) it is feasible in the presence of a number of substances frequently used in protein analyses and making difficult or impossible measurement according to LOWRY et al.  相似文献

High blood levels of homocysteine (Hcy) are found in patients affected by homocystinuria, a genetic disorder caused by deficiency of cystathionine β-synthase (CBS) activity, as well as in nutritional deficiencies (vitamin B12 or folate) and in abnormal renal function. We previously demonstrated that lipid and protein oxidative damage is increased and the antioxidant defenses diminished in plasma of CBS-deficient patients, indicating that oxidative stress is involved in the pathophysiology of this disease. In the present work, we extended these investigations by evaluating DNA damage through the comet assay in peripheral leukocytes from CBS-deficient patients, as well as by analyzing of the in vitro effect of Hcy on DNA damage in white blood cells. We verified that DNA damage was significantly higher in the CBS-deficient patients under treatment based on a protein-restricted diet and pyridoxine, folic acid, betaine and vitamin B12 supplementation, when compared to controls. Furthermore, the in vitro study showed a concentration-dependent effect of Hcy inducing DNA damage. Taken together, the present data indicate that DNA damage occurs in treated CBS-deficient patients, possibly due to high Hcy levels.  相似文献
