Cytogenetic effects of 3,4-dichloroaniline in human lymphocytes and V79 Chinese hamster cells 总被引:2,自引:0,他引:2
3,4-Dichloroaniline (3,4-DCA), an intermediate in various chemical syntheses, has been detected as an environmental contaminant in surface waters and in the effluents from dye-manufacturing plants. Tested for clastogenicity in human lymphocytes in vitro the compound was inactive in the chromosome aberration assay yet exhibited a positive sister-chromatid exchange response in the presence of a mammalian metabolic activation system. Exposure of V79 Chinese hamster cells to 3,4-DCA caused a concentration-dependent increase in the incidence of spindle disturbances, predominantly of the initial c-mitotic type. The results indicate that 3,4-DCA might induce aneuploidy in mammalian cells by interaction with the mitotic apparatus. 相似文献

Summary Cuticle/water partition coefficients (Kc/w) for d-limonene, -pinene and -pinene were determined by an extrapolation and a desorption method. The sorption experiments were carried out with isolated angiosperm and gymnosperm cuticles and with [14C]-labelled monoterpenes, which were obtained biosynthetically. Both methods were suitable for the determination of the Kc/w of volatile hydrophobic compounds. For the angiosperm cuticles the partition coefficients are of the order of 104, which indicates a high accumulation of monoterpenes in the cuticle. The values of the conifer cuticles of Picea abies (L.) Karst. and Abies alba Mill., however, are lower due to their high lignin content. This is proved by the increase of the partition coefficients after removal of polar and phenolic components. The Kc/w can be estimated with good accuracy from the octanol/water partition coefficient, which was determined experimentally. 相似文献

Collagen type conservation during metamorphic repatterning of the dermal fibers in salamanders 总被引:1,自引:0,他引:1
The orientation of the fibers in the dermis of the tiger salamander, Ambystoma tigrinum, undergoes a dramatic repatterning at metamorphosis. The pre-metamorphic, larval dermis is a tight layer composed of crossed fibers that wind helically around the trunk. This condition is retained by neotenic adults which do not undergo metamorphosis. In contrast, the neotenic adults which do not undergo metamorphosis. In contrast, the metamorphosed adult dermis consists of a superficial, loose network of fibers invested with large multicellular glands--the stratum spongiosum--and a deeper tight layer of fibers--the stratum densum. However, unlike the crossed fibers of the pre-metamorphic dermis, there is no preferred orientation to the fibers in either layer of the post-metamorphic dermis. In order to evaluate whether these two distinctly different fiber patterns are constructed from biochemically similar fibers, the collagen types present in the pre- and post-metamorphic dermis were determined using SDS-polyacrylamide gel electrophoresis. Type I collagen is the predominant collagen of the dermis and the same major collagen types are present for all individuals, whether pre- or post-metamorphic. N-Glycosylation occurs cotranslationally as soon as the growing polypeptide chain enters the endoplasmic reticulum, before the final native-like folded state is reached. We examined the role of the carbohydrate chains in the mechanism of protein folding. The in vitro folding and association of yeast invertase are used as an experimental system. External invertase contains approximately 50% carbohydrate, whereas cytoplasmic invertase is not glycosylated. The functional native state of both proteins is a homodimer. At pH greater than or equal to 6.5 and at protein concentrations below 3 micrograms/ml, the kinetics of reactivation and the final yields are similar for the two invertases. For both proteins, reactivation is a sequential reaction with a lag phase at the beginning. The nonglycosylated protein tends to aggregate during reactivation at low pH and at protein concentrations above 3 micrograms/ml. After separation of inactive material, the renatured protein is indistinguishable from the original native state by a number of physicochemical and functional criteria. The results suggest that the carbohydrate moiety does not affect the mechanism of folding and association of invertase. However, glycosylation improves the solubility of unfolded or partially folded invertase molecules and hence leads to a suppression of irreversible aggregation. Such a protective effect may also be important for the in vivo maturation of nascent glycosylated protein chains. 相似文献

Lysogenicity and phage typability of Staphylococcus aureus L-form cells are described. Spontaneously produced phages were found in thin sections of S. aureus L colonies. The dimensions of the tail and head resemble those of the morphological group BIII2 (T. Krzywy, I. Durlakowa, A. Kucharewcz-Krukowska, S. Krynski, and S. Slopek, Zentralbl. Bakteriol. Mikrobiol. Hyg. Zusammenfassung Bei einem normal intelligenten, kleinwüchsigen, 219/12 jährigen Mann mit Hypospadie, fraglicher rudimentärer Vagina und hypergonadotropem Hypogonadismus findet man bei der Biopsie des nur 7 ml großen linken Hodens eine gleichförmige tubuläre Atrophie mit Fehlen des Keimepithels und eine diffuse, kleinherdige Leydigzellwucherung. Sex-Chromatin in den Leydigzellen, im Mundabstrich und Drumsticks negativ. Karyotyp in den Lymphocyten und im Knochenmark normal weiblich (46,XX), in den Fibroblasten jedoch 45,XO.Somit weist dieser Patient Testesgewebe auf, das Y-Chromosom ist aber unauffindbar. Zur Klärung dieser Diskrepanz scheint das Vorhandensein einer dritten, zahlenmäßig stark in den Hintergrund tretenden, ein Y enthaltenden Zellpopulation die naheliegendste Hypothese zu sein.Eine Zwillingsschwester kam im Neugeborenenalter ad exitum; die Blutgruppenuntersuchungen beim Patienten und seiner Familie machen aber einen Blutchimärismus sehr unwahrscheinlich.Das klinische Bild unseres Patienten ist vollkommen verschieden von demjenigen, das beim XX/XO-Mosaik üblicherweise beobachtet wird und läßt sich auch vom klassischen Klinefelter-Syndrom deutlich abgrenzen. Es zeigt Ähnlichkeiten mit einigen Fällen von XO/XY-Mosaik.

Summary In a normally intelligent man of 219/12 years the following main physical findings were present: short stature, hypospadias, possibly a rudimentary vagina, hypergonadotropic hypogonadism, small testis (7 ml) with tubular dystrophy consisting in absence of germinal epithelium and localized proliferation of Leydig's cells. Sex chromatin in buccal mucosa cells was negative; no drumsticks were found in neutrophils. The karyotype in lymphocyte cultures and bone-marrow preparations was 46,XX; in several independent fibroblast cultures 45,XO.The most likely explanation for the presence of testicular tissue seems to be the existence of an undiscovered cell population containing a Y-chromosome.A twin sister of the patient died in early infancy; blood group investigations gave no indications for the presence of blood chimerism.The clinical picture of this patient is entirely different from that of the usual XX/XO mosaics, it also clearly differs from Klinefelter's syndrome but shows some similarities to cases of XO/XY mosaicism.

Quantum Requirement for Photosynthesis in Chlorophyll-Deficient Plants with Unusual Lamellar Structures 总被引:1,自引:0,他引:1
Neither an over-all deficiency of chlorophyll, nor an increased enzymatic capacity for maximal rates, nor an unusual lamellar structure was found to change the number of quanta required for the evolution of one molecule of oxygen in healthy aurea mutants of tobacco. The average minimal quantum number remains 10 (efficiency 0.1) as in many algae and typical higher plants. Most of the time the optimal efficiency depends on the availability of some far-red radiation, particularly in the blue region of the spectrum where blue light alone is rather inefficient. These results fit an explanation offered earlier in connection with the hydrogen or acetate photometabolism of algae in far-red light. 相似文献
