Larval parasitoids can substantially reduce the population density of the pollen beetle [Brassicogethes aeneus (Fabricius), syn. Meligethes aeneus (Fabricius)]. The most abundant tersilochine parasitoids of pollen beetle are Tersilochus heterocerus, Phradis interstitialis and P. morionellus. The main activity of these parasitoids was observed in the period shortly before flowering to full flowering of oilseed rape. Insecticide applications during this period may have negative effects on parasitoids. In the present study, the effects of the insecticides Biscaya (a.i. thiacloprid), Mavrik (a.i. tau-fluvalinate) and Karate Zeon (a.i. lambda-cyhalothrin) applied during the bud or flowering stage of winter oilseed rape on parasitization of pollen beetle larvae by T. heterocerus were studied in 12 field trials at different locations in Germany in 2013–2015. The effects on parasitism by Phradis spp. were assessed in 2015. Parasitism of pollen beetle larvae by T. heterocerus was found in all field trials in all experimental years, but in most trials not before full flowering. Maximum percentage of parasitized larvae at different locations ranged between 3.4 and 16.8% in 2013, 8.3 and 22.4% in 2014 and from 11.1 to 29.1% in 2015. Levels of parasitism were not significantly different between the untreated control and insecticide treatments within each location. In contrast to T. heterocerus, Phradis spp. was not detected at all locations and not before flowering declining. In field trials at Lucklum and Puch, the maximum level of parasitism by Phradis spp. was 9.4 and 18.3%, respectively. No significant effect of insecticide application on parasitism by Phradis spp. was observed between the treatments. The results of this study showed that the insecticides used in the field trials did not affect parasitization of pollen beetle larvae by T. heterocerus and Phradis spp., regardless whether applied at the bud stage, at the beginning of flowering or full flowering.  相似文献

Summary The formation and localization of the -lactamase of Acinetobacter calcoaceticus CCM 5593 is strongly affected by cultivation and induction conditions. Optimal parameters for enzyme yield are cultivation on minimal salts medium with acetate (10 g·1–1) as carbon source and addition of yeast extract (5–10 g·l–1), induction by cefotaxime (50g·ml–1) immediately after inoculation and growth for 24 h at 25° C. The strain forms a basal level of -lactamase constitutively [70 units (U)·g–1]. Nearly all of this was found to be cell-bound. However, -lactamase activity additionally produced after induction (up to 500 U·g–1 wet bacteria) was located in the culture medium (up to 96%). The proton pumping mechanism of cytochrome c oxidase on a molecular level is highly disputed. Recently theoretical calculations and real time electron transfer measurements indicated the involvement of residues in the vicinity of the ring A propionate of heme a3, including Asp399 and the CuB ligands His 325, 326. In this study we probed the interaction of Asp399 with the binuclear center and characterize the protonation state of its side chain. Redox induced FTIR difference spectra of mutations at the site in direct comparison to wild type, indicate that below pH 5 Asp 399 displays signals typical for the deprotonation of the acidic residue with reduction of the enzyme. Interestingly at a pH higher than 5, no contributions from Asp 399 are evident. In order to probe the interaction of the site with the binuclear center we followed the rebinding of CO by infrared spectroscopy for mutations on residue Asp399 to Glu, Asn and Leu. Previously different CO conformers have been identified for bacterial cytochrome c oxidases, and its pH dependent behaviour discussed to be relevant for catalysis. Interestingly we observe the lack of this pH dependency and a strong influence on the observable conformers for all mutants studied here, clearly suggesting a communication of the site with the heme-copper center and the nearby histidine residues.  相似文献

Mitotic kinesins represent potential drug targets for anticancer chemotherapy. Inhibitors of different chemical classes have been identified that target human Eg5, a kinesin responsible for the establishment of the bipolar spindle. One potent Eg5 inhibitor is S-trityl-L-cysteine (STLC), which arrests cells in mitosis and exhibits tumor growth inhibition activity. However, the underlying mechanism of STLC action on the molecular level is unknown. Here, cells treated with STLC were blocked in mitosis through activation of the spindle assembly checkpoint as shown by the phosphorylated state of BubR1 and the accumulation of mitosis specific phosphorylation on histone H3 and aurora A kinase. Using live cell imaging, we observed prolonged mitotic arrest and subsequent cell death after incubation of GFP-alpha-tubulin HeLa cells with STLC. Activated caspase-9 occurred before cleavage of caspase-8 leading to the accumulation of the activated executioner caspase-3 suggesting that STLC induces apoptosis through the intrinsic apoptotic pathway. Proteome analysis following STLC treatment revealed 33 differentially regulated proteins of various cellular processes, 31 of which can be linked to apoptotic cell death. Interestingly, four identified proteins, chromobox protein homolog, RNA-binding Src associated in mitosis 68 kDa protein, stathmin, and translationally controlled tumor protein can be linked to mitotic and apoptotic processes.  相似文献

Nitric oxide synthase in the rat carotid body and carotid sinus   总被引:5,自引:0,他引:5  
The participation of nitric oxide synthase (NOS) in the innervation of the rat carotid body and carotid sinus was investigated by means of NADPH-diaphorase histochemistry and NOS immunohistochemistry using antisera raised against purified neuronal NOS and a synthetic tridecapeptide. NOS was detected in 23% of neurons at the periphery of the carotid bodies. Some negative neurons were surrounded by NOS-positive terminals. NOS-containing varicose nerve fibres innervated the arterial vascular bed and, to a lesser extent, the islands of glomus cells. These fibres persisted after transection of the carotid sinus nerve and are probably derived from intrinsic neurons. Large NOS-positive axonal swellings in the wall of the carotid sinus were absent after transection of the sinus nerve, indicating their sensory origin. The results suggest a neuronal nitrergic control of blood flow, neuronal activity and chemoreception in the carotid body, and an intrinsic role of NO in the process of arterial baroreception.  相似文献

To prepare labeled precursors for biosynthetic studies, methods for the specific introduction of tritium and deuterium into the reducing and the terminal glucose unit of maltotriose were developed. Thus [6″-3H]- and (6″-2H)-maltotriose (17) and (18) were prepared via selective methoxytritylation, deprotection and subsequent modified Pfitzner-Moffatt oxidation, followed by reduction with sodium borotritiide or sodium borodeuteride, respectively. A simple two step procedure utilizing the Lobry de Bruyn/van Ekenstein transformation gave (2-2H)maltotriose (20).  相似文献

The group A streptococcus (GAS) Streptococcus pyogenes is known to cause self-limiting purulent infections in humans. The role of GAS pili in host cell adhesion and biofilm formation is likely fundamental in early colonization. Pilus genes are found in the FCT (fibronectin-binding protein, collagen-binding protein, and trypsin-resistant antigen) genomic region, which has been classified into nine subtypes based on the diversity of gene content and nucleotide sequence. Several epidemiological studies have indicated that FCT type 1 strains, including serotype M6, produce large amounts of monospecies biofilm in vitro. We examined the direct involvement of pili in biofilm formation by serotype M6 clinical isolates. In the majority of tested strains, deletion of the tee6 gene encoding pilus shaft protein T6 compromised the ability to form biofilm on an abiotic surface. Deletion of the fctX and srtB genes, which encode pilus ancillary protein and class C pilus-associated sortase, respectively, also decreased biofilm formation by a representative strain. Unexpectedly, these mutant strains showed increased bacterial aggregation compared with that of the wild-type strain. When the entire FCT type 1 pilus region was ectopically expressed in serotype M1 strain SF370, biofilm formation was promoted and autoaggregation was inhibited. These findings indicate that assembled FCT type 1 pili contribute to biofilm formation and also function as attenuators of bacterial aggregation. Taken together, our results show the potential role of FCT type 1 pili in the pathogenesis of GAS infections.  相似文献

The N-end rule degradation pathway states that the half-life of a protein is determined by the nature of its N-terminal residue. In Escherichia coli the adaptor protein ClpS directly interacts with destabilizing N-terminal residues and transfers them to the ClpA/ClpP proteolytic complex for degradation. The crucial role of ClpS in N-end rule degradation is currently under debate, since ClpA/ClpP was shown to process selected N-terminal degrons harbouring destabilizing residues in the absence of ClpS. Here, we investigated the contribution of ClpS to N-end rule degradation by two approaches. First, we performed a systematic mutagenesis of selected N-degron model substrates, demonstrating that ClpS but not ClpA specifically senses the nature of N-terminal residues. Second, we identified two natural N-end rule substrates of E. coli : Dps and PATase (YgjG). The in vivo degradation of both proteins strictly relied on ClpS, thereby establishing the function of ClpS as the essential discriminator of the E. coli N-end rule pathway.  相似文献
