The removal of seeded coliphage f2 and indigenous enteroviruses from primary and secondary wastewaters applied by spray irrigation to sandy loam and silt loam soils in field test cells was examined. The amount of f2 recovered from 170-cm-deep soil percolate samples taken over a 53-day period never exceeded 0.1% of applied virus levels and was usually below detection limits. Indigenous enterovirus levels in percolate waters also constituted only a small portion of those found in the wastewaters. At 10 days after seeding, f2 virus was present throughout the soil column but tended to accumulate around the soil core middepths. Coliphage f2 disappeared from the soil surface regions at a high rate, and by 53 days very little virus could be detected within the length of the soil columns. Sterilized soil core segments from different depths were studied to determine their virus adsorption capabilities when suspended in either wastewater, test cell percolate water, or distilled water containing divalent cations. The adsorptive capacity of Windsor and Charlton soils for poliovirus 1 and coliphage f2 increased greatly with the soil sample depth until leveling off at the midcore depths. Studies on the effects of decapitation, head transplantation, azadirachtin, and ecdysone therapy on the ultrastructural organization of the midgut of Rhodnius prolixus, a vector of the protozoan Trypanosoma cruzi, show a distinct effect on the organization of the epithelial cells. When insects are decapitated or treated with azadirachtin, the ultrastructural organiza tion of these compartments changed significantly and drastically blocked the development of T. cruzi infection. In converse experiments, head transplantation or oral therapy with ecdysone significantly re versed the T. cruzi infectivity and reestablished the organization of the stomach and intestine in decapitated or azadirachtin-treated insects. These results indicat that a brain factor, possibly the prothoracicotropic hormone which stimulates ecdysteroid production on the prothoracic glands, may act directly or indirectly on both the midgut cell organiza tion and the intestinal microenvironment, interfering in the trypanosome survival and infection of the vector R. prolixus. 相似文献

Most trees form symbioses with ectomycorrhizal fungi (EMF) which influence access to growth-limiting soil resources. Mesocosm experiments repeatedly show that EMF species differentially affect plant development, yet whether these effects ripple up to influence the growth of entire forests remains unknown. Here we tested the effects of EMF composition and functional genes relative to variation in well-known drivers of tree growth by combining paired molecular EMF surveys with high-resolution forest inventory data across 15 European countries. We show that EMF composition was linked to a three-fold difference in tree growth rate even when controlling for the primary abiotic drivers of tree growth. Fast tree growth was associated with EMF communities harboring high inorganic but low organic nitrogen acquisition gene proportions and EMF which form contact versus medium-distance fringe exploration types. These findings suggest that EMF composition is a strong bio-indicator of underlying drivers of tree growth and/or that variation of forest EMF communities causes differences in tree growth. The effects of lignoids on feeding, ecdysis and diuresis in fourth-instar larvae of Rhodnius prolixus (Hemiptera) were investigated. Up to 100 microg/ml burchellin, podophyllotoxin, pinoresinol, sesamin, licarin A, or nordihydroguaiaretic acid (NDGA) in the diet did not induce antifeedant effects. Pinoresinol and NDGA significantly inhibited ecdysis. In experiments in vivo, burchellin and podophyllotoxin reduced the production of urine after feeding. 5-Hydroxytryptamine (5-HT), a diuretic hormone, partially counteracted this effect of burchellin. In experiments in vitro, using isolated Malpighian tubules, (i) burchellin reduced diuretic hormone levels in the hemolymph but not the amount of diuretic hormone stored in the thoracic ganglionic masses (including axons), (ii) burchellin decreased the volume of urine secreted by isolated Malpighian tubules, and (iii) 5-HT could not overcome the effect of burchellin upon the Malpighian tubules. We conclude that burchellin interfered with the release, but not with the production of diuretic hormone by the thoracic ganglionic mass or induced an antidiuretic hormone and directly affected the Malpighian tubules. 相似文献

Several species of kinetoplastid protozoa cause major human infectious diseases. Trypanosoma cruzi is responsible for the fatal Chagas disease in large parts of South America, the various species of Leishmania cause a number of different human diseases with millions of patients world-wide, and the African trypanosome Trypanosoma brucei is the agent of human sleeping sickness, a disastrously re-emerging epidemic of fatal infections in Sub-Saharan Africa. Chemotherapy of all of these infections is in a very unsatisfactory state. cAMP signalling pathways in humans have provided interesting drug targets for a number of clinical conditions, from asthma to impotency. Similarly, cAMP signalling in kinetoplastids might offer useful targets for the development of novel antiparasitic drugs, which makes their exploration an urgent need. Current knowledge suggests that cAMP signalling proceeds along very similar pathways in all kinetoplastid pathogens (T. cruzi, the Leishmanias and T. brucei). Their adenylyl cyclases are structurally very different from the human enzymes and appear to function as enzyme-linked cell surface receptors. They might represent the major sensory apparatus of the kinetoplastids, guiding much of their environmental sensing and host/parasite interaction. The cAMP-specific phosphodiesterases of the kinetoplastids are rather similar to those of human cells and might function in similar ways. Essentially nothing is known on downstream effectors of cAMP in the kinetoplastids. Homologues of protein kinase A and its regulatory subunits have been identified, but their biochemical properties seem to be disctinct from that of mammalian protein kinase A. 相似文献

The hyperinsulinism-hyperammonemia syndrome (HHS) has been shown to result from 'gain-of-function' mutations of the glutamate dehydrogenase (GlDH) gene, GLUD1. In the original report, all mutations were found in a narrow range of 27 base pairs within exons 11 and 12 which predicted an effect on the presumed allosteric domain of the enzyme and all these mutations were associated by a diminished inhibitory effect of guanosine triphosphate (GTP) on GlDH activity. We have investigated 14 patients from seven European families with mild hyperinsulinism. In four families, more than one member was affected. In eight cases hyperammonemia was documented, and eight cases had signs of significant leucine sensitivity. In one of the families, a novel heterozygous missense mutation in exon 6 [c.833C>T (R221C)] was detected, and in all other cases from six unrelated families the novel heterozygous missense mutation c.978G>A (R269H) was found in exon 7. When GIDH activity was measured in lymphocytes isolated from affected patients, both mutations were shown to result in a normal basal activity but a diminished sensitivity to GTP. It is the first time that this effect is reported for mutations located in the presumed catalytic site and outside the GTP allosteric domain of the enzyme. The observation of the high prevalence of the exon 7 mutation both in familial and sporadic cases of HHS suggests a mutation hot spot and justifies a mutation screening for this novel mutation by mismatch PCR-based restriction enzyme digestion in patients with hyperinsulinism. 相似文献

The rate at which migrant birds replenish their energy stores at intermittent stopovers largely determines overall migration
speed, the manner in which migration proceeds and success of migration. In this study, data on the fuel deposition rate (FDR)
of three long-distance migrants from 17 ringing sites along their autumn migration route were used to examine: (1) effects
of endogenous factors on FDR, and (2) how relationships between exogenous factors and FDR affect the organisation of migration.
We developed a model to estimate FDR from retrapped birds which takes into account time of day and various other factors which
might influence FDR. The two endogenous factors, moult and current energy stores, generally reduced FDR. This may result in
lower departure energy loads and more stopovers than expected from optimal migration theory. Differences between species with
respect to seasonal, year-to-year and geographical patterns of FDR could be related to differences in availability and predictability
of food resources, and help to explain differences in the organisation of migration. A low FDR in northern and central Europe
could be related to low, but predictable, food resources and an early departure during moult of the reed warbler (Acrocephalus scirpaceus); FDRs varying between years were related to large spatial and year-to-year variation in the density of the main prey of
the sedge warbler (Acrocephalus schoenobaenus); and a high FDR in the garden warbler (Sylvia borin) was related to abundant food resources, due to a switch from a purely invertebrate diet to a mixed diet including fruits
which are abundant over large areas of Europe and north Africa. This study demonstrated that the organisation of migration
is the outcome of a complex interplay of the seasonal timing of moult, food availability and predictability and a seasonal
switch in diet, and can be modified by individual birds in response to a limited amount of time in which to migrate.
