The transpiration rate of individual 6-day-old oat plants was forced to oscillate by cyclic sine-shaped changes in the leaf irradiance (frequency 2 cycles h?1, amplitude and average value 1.4 mW cm?2, red light 620–800 nm). By means of a specially designed cuvette with three chambers the transpiration rate from three different segments of the leaf could be measured simultaneously. The leaf segments were illuminated individually and the illumination on each leaf segment could be modulated independently. The experiments showed that there was a strong correlation between the transpiration rates from the different leaf segments, dependent on a coupling mechanism in the plant. The coupling phenomenon disappeared when the root system was eliminated or when the water potential of the root medium was lowered. It was experimentally shown that CO2 diffusion in the leaf could not be the primary cause for the coupling. Therefore the stomatal dependence on the leaf water potential was considered the most probable reason for the coupling. The frequency of the forcing light cycles could be linearly changed during an experiment and this swept-frequency technique was used to obtain a frequency response of one single oat plant. The technique made it also possible to study the strength of the coupling between different leaf segments.  相似文献

Effects of one week of benzodiazepine drug administration on central benzodiazepine receptor binding characteristics were evaluated in a series of experiments in male Sprague-Dawley rats. Administration of short- and intermediate-acting benzodiazepines was observed to increase the number of available receptor binding sites (Bmax) without changing affinity of drug for receptor. Furthermore, these changes did not occur after administration and withdrawal of long-acting benzodiazepines. In addition, there appeared to be a relationship between the affinity of the different benzodiazepines for the receptor and the degree of increase in the number of receptor binding sites. The results may help to explain the relationship between withdrawal of certain benzodiazepine drugs and the occurrence of rebound phenomena in clinical situations.  相似文献

1. The contribution of non‐genetic maternal effects to offspring performance is well established and the evidence for paternal effects has also been increasing recently. Studies determining the relative contributions of the two parents to offspring success are, however, still rare. 2. In this study, two stressors were applied to adult red flour beetles (Tribolium castaneum) – starvation and cold stress – and a full‐factorial design was used to distinguish between maternal and paternal reproductive decisions and their effects on the offspring. 3. Starvation had a stronger negative effect than cold stress on both males and females, and the likelihood of starved females producing offspring was very low. Furthermore, starved fathers led to lower offspring mass at the larval stage, probably leading to impaired starvation tolerance of the offspring. 4. Cold‐stressed fathers were less likely than unstressed fathers to reproduce, whereas cold‐stressed mothers demonstrated a similar effect by producing fewer offspring. 5. Applying stress probably led to energy saving that came at the expense of reproduction intensity. It is suggested that the smaller offspring mass is a negative consequence of the parental exposure to stress. 6. The differences between the consequences of the two stressors applied and between the relative contribution of each parent could perhaps be explained by the distinct physiological responses of each sex to each of the stressors.  相似文献

A sediment core encompassing the last 2000 years was extracted from Lake Laja, Chile, (36°54′S, 71°05′W) using an Uwitec drilling platform. The sediment was subsampled for loss on ignition, nutrients, biogenic silica, and biological proxies (diatoms, chironomids, pollen). The sedimentary profile was characterized by several coarse volcanic layers. Loss on ignition, nutrients, and biogenic silica showed an increasing trend that suggests a recent shift to a higher trophic status. Diatom assemblages also suggested higher nutrient content with increased abundances of Aulacoseira granulata, A. distans, and Asterionella formosa. At the same time, a marked change in the benthic and facultative planktonic taxa may be associated with cooling. This period of change coincides with the European Little Ice Age (LIA). The chironomid profile showed four key zones distinguished largely by changes in the abundance of Tanytarsini, Parachironomus, and Macropelopia. Like diatoms, chironomids also seemed to reflect a shift to higher trophic conditions in the upper part reflected by increasing abundance of taxa such as Tribelos/Phaenopsectra, Cricotopus/Orthocladius, and Ablabesmyia. The most striking feature in the chironomid assemblage is the abundance of Podonominae, Parapsectrocladius, and Limnophyes/Compterosmittia, which could be associated with a cold-dry period between 1500 and 1900 AD in Lake Laja (the period of the European LIA). Pollen assemblages indicated fluctuations in humidity through changes in Nothofagus dombeyi-type, Poaceae, and Ephedra, and we inferred a strong human impact over the last 100 years from the appearance of Plantago and increased levels of Poaceae and Asteraceae subf. Cichorioidae. Finally, the three proxies showed the occurrence of a cold-dry event in Lake Laja (~1550–1900 AD), which roughly coincides with the European LIA. However the data from this research, does not prove neither rejects the existence of the occurrence of the MWP in the central Andes.  相似文献

Four esterase genes and general esterase activity were investigated in the gut of the termite Reticulitermes flavipes. Two genes (RfEst1 and RfEst2) share significant translated identity with a number of insect JH esterases. The two remaining genes (RfEst3 and RfEst4) apparently code for much shorter proteins with similarity to fungal phenolic acid esterases involved in hemicellulose solubilization. All four genes showed consistently high midgut expression. This result was further supported by colorimetric activity assays and Native polyacrylamide gel electrophoresis, which showed significant esterase activity and a number of isoforms in the midgut. The greatest esterase activity and isoform composition were detected when α‐naphthyl propionate was used as a substrate. Moreover, esterase activity and diverse isoforms were present in gut mitochondrial, microsomal, and cytosolic sub‐cellular protein fractions, as well as in the hindgut lumen. These findings reveal an agreement between gut esterase gene expression and activity distributions, and support the idea that R. flavipes gut esterase activity is host (not symbiont)‐derived. In addition, these findings support the hypotheses that termite gut esterases may play important roles in lignocellulose digestion and caste differentiation. This study provides important baseline data that will assist ongoing functional‐genomic efforts to identify novel genes with roles in semiochemical, hormone, and lignocellulose processing in the termite gut. © 2009 Wiley Periodicals, Inc.  相似文献

Tumor-specific memory T cells are detectable in the bone marrow (BM) of a majority of breast cancer patients. In vitro they can be reactivated to IFN-γ producing, cytotoxic effector cells and reject autologous, xenotransplanted tumors in NOD/SCID mice after specific restimulation with autologous dendritic cells (DC). In this study, we demonstrate the presence of specific tumor-reactive BM memory T cells in altogether 56 out of 129 primarily operated breast cancer patients by short-term IFN-γ EliSpot assays with unstimulated T cells and tumor antigen presenting, autologous DCs. We observed tumor-reactive BM memory T cells predominantly in patients with primarily metastatic disease (P = 0.011) or with increased concentrations of tumor marker CA 15-3 in the peripheral blood (P = 0.004), respectively. Memory T cell reactivity against HLA-A*0201-restricted peptides from the tumor-associated antigens MUC1, Hpa16–24 and Hpa183–191 was also detected particularly in patients with elevated peripheral CA 15-3 concentrations (P < 0.05). Altogether these data indicate that the systemic presence of tumor-derived antigens promotes an induction of tumor-specific cellular immune responses in the human BM.  相似文献
