Previous studies have shown that sera from HIV-1-infected individuals contain antibodies able to mediate antibody-dependent cellular cytotoxicity (ADCC). These antibodies preferentially recognize envelope glycoprotein (Env) epitopes induced upon CD4 binding. Here, we show that a highly conserved tryptophan at position 69 of the gp120 inner domain is important for ADCC mediated by anti-cluster A antibodies and sera from HIV-1-infected individuals.  相似文献

Onion guard cells, in contrast to those of Vicia and Pisum, do not require an alkaline treatment in order to fluoresce. Fluorescing compounds of Allium cepa L. were characterized using in-vivo microspectrophotometry; furthermore, invitro chemical analysis for epidermal tissue, intact guard and epidermal cells, and isolated guard-cell protoplasts was performed. The emission intensity (max 520 nm) decreased when intact onion guard cells were excited with 436 nm light, but increased (max 470 nm) when excited at 365 nm. This photodecomposition at 436 nm is typical of flavins or flavoproteins whereas an increase in fluorescence intensity with excitation at 365 nm may be explained by the presence of other substances. The presence of flavins could not be unambiguously confirmed from these results. Indeed, the absorption spectra of the vacuolar area of guard cells did not show the peak at 445 nm which is characteristic for flavins. Furthermore, there was no decrease of absorption at the excitation wavelengths of 440 and 330 nm. Since spectral data indicate the presence at high amounts of flavonoids in guard and epidermal cells, this may reduce the sensitivity for the detection of flavins in guard cells. Using thin-layer chromatography and high-performance liquid chromatography together with hydrolytic procedures, flavonol glycosides with kaempferol and quercetin as aglycones substituted with sulphate and glucuronate were identified. The PS120 variant of Chinese hamster lung fibroblasts which lacks Na+/H+ exchange activity was used to investigate bicarbonate transport systems and their role in intracellular pH (pHi) regulation. When pHi was decreased by acid load, bicarbonate caused pHi increase and stimulated 36Cl- efflux from the cells, both in a Na+-dependent manner. These results together with previous findings that bicarbonate stimulates 22Na+ uptake in PS120 cells (L'Allemain, G., Paris, S., and Pouyssegur, J. (1985) J. Biol. Chem. 260, 4877-4883) demonstrate the presence of a Na+-linked Cl-/HCO3- exchange system. In cells with normal initial pHi, bicarbonate caused Na+-independent pHi increase in Cl(-)-free solutions and stimulated Na+-independent 36Cl- efflux, indicating that a Na+-independent Cl-/HCO3- exchanger is also present in the cell. Na+-linked and Na+-independent Cl-/HCO3- exchange is apparently mediated by two distinct systems, since a [(tetrahydrofluorene-7-yl)oxy]acetic acid derivative selectively inhibits the Na+-independent exchanger. An additional distinctive feature is a 10-fold lower affinity for chloride of the Na+-linked exchanger. The Na+-linked and Na+-independent Cl-/HCO3- exchange systems are likely to protect the cell from acid and alkaline load, respectively.  相似文献

The Protein Data Bank currently contains about 600 data sets of three-dimensional protein coordinates determined by X-ray crystallography or NMR. There is considerable redundancy in the data base, as many protein pairs are identical or very similar in sequence. However, statistical analyses of protein sequence-structure relations require nonredundant data. We have developed two algorithms to extract from the data base representative sets of protein chains with maximum coverage and minimum redundancy. The first algorithm focuses on optimizing a particular property of the selected proteins and works by successive selection of proteins from an ordered list and exclusion of all neighbors of each selected protein. The other algorithm aims at maximizing the size of the selected set and works by successive thinning out of clusters of similar proteins. Both algorithms are generally applicable to other data bases in which criteria of similarity can be defined and relate to problems in graph theory. The largest nonredundant set extracted from the current release of the Protein Data Bank has 155 protein chains. In this set, no two proteins have sequence similarity higher than a certain cutoff (30% identical residues for aligned subsequences longer than 80 residues), yet all structurally unique protein families are represented. Periodically updated lists of representative data sets are available by electronic mail from the file server "netserv@embl-heidelberg.de." The selection may be useful in statistical approaches to protein folding as well as in the analysis and documentation of the known spectrum of three-dimensional protein structures.  相似文献

Some analogies were demonstrated between the spontaneous acquisition of phonemes, especially gutturals and clicks, and their also spontaneous extinction during the early babble phase of infants on the one hand and the appearance and involution of some blastemata throughout the human ontogenesis on the other hand. Also analogues seem to be the producing of the so-called instinctives as mam(m)a and papa by somewhat older babies which are able to pass over from the babbling into permanent words of the adults' speech in which they persist if used without shifting of sounds since they are produced de novo generation by generation, but they are subordinate to shifting and possible extinction if used in the form of derivatives in the standard language, and some phenomena of the phylogenesis as the survival of less differentiated species contrary to the relatively quick extinction of the highly specialized ones. Thus the glottogenesis is a dialectic process consisting of interaction by formation, differentiation, and involution.  相似文献

A probable carbon flow from the Calvin cycle to branched chain amino acids and lipids via phosphoenolpyruvate (PEP) and pyruvate was examined in spinach (Spinacia oleracea) chloroplasts. The interpendence of metabolic pathways in and outside chloroplasts as well as product and feedback inhibition were studied. It was shown that alanine, aromatic, and small amounts of branched chain amino acids were formed from bicarbonate in purified intact chloroplasts. Addition of PEP only favored formation of aromatic amino acids. Mechanisms of regulation remained unclear. Concentrations of PEP and pyruvate within the chloroplast impermeable space during photosynthetic carbon fixation were 15 times higher than in the reaction medium. A direct carbon flow to pyruvate was identified (0.1 micromoles per milligram chlorophyll per hour). Pyruvate was taken up by intact chloroplasts slowly, leading to the formation of lysine, alanine, valine, and leucine plus isoleucine (approximate ratios, 100-500:60-100:40-100:2-10). The Km for the formation of valine and leucine plus isoleucine was estimated to be 0.1 millimolar. Ten micromolar glutamate optimized the transamination reaction regardless of whether bicarbonate or pyruvate was being applied. Alanine and valine formation was enhanced by the addition of acetate to the reaction mixture. The enhancement probably resulted from an inhibition of pyruvate dehydrogenase by acetyl-S-coenzyme A formed from acetate, and resulting accumulation of hydroxyethylthiamine diphosphate and pyruvate. High concentrations of valine and isoleucine inhibited their own and each others synthesis and enhanced alanine formation. When pyruvate was applied, only amino acids were formed; when complemented with bicarbonate, fatty acids were formed as well. This is probably the result of a requirement of acetyl-S-coenzyme A-carboxylase for bicarbonate.  相似文献

1. Predation on vectors of pathogens can indirectly influence infection spread. In addition to the consumptive aspect of predation, non-consumptive, predator-induced changes in various vector traits can lead to trait-mediated indirect effects on pathogen spread, potentially operating in various directions and magnitudes. 2. A widespread non-consumptive effect of predation is the alteration of individual prey development rates. Yet, the implications of this phenomenon for the spread of vector-borne plant pathogens have not been studied. It is hypothesized that the epidemiological effects of predator-induced changes in vector development rate depend on the pattern in which the transmission biology of the vector changes along its ontogeny. 3. A general epidemiological model was developed that considers the role of predation in the infection dynamics of a plant pathogen, while incorporating vector stage structure to allow for variation in its development rate. 4. By contrasting scenarios that represent typical plant disease systems, this study confirms that the magnitude of the effect of altered development rate on infection prevalence depends on the disparity between juvenile and adult vectors in their pathogen transmission potential. 5. The model also reveals that the effect of predator-induced change in development rate can impact pathogen spread counterintuitively. Specifically, slowing down vector development can result in increased pathogen prevalence due to apparent competition between infected and uninfected vector populations. 6. More detailed, stage-specific studies of non-consumptive predator effects on vectors are likely to advance our understanding of plant disease ecology, and the development of more effective biological control practices in agriculture.  相似文献

Soybean cultivars (Glycine max(L.) Merr.) infected with soybean cyst nematode (SCN; Heterodera glycinesIchinohe) often show symptoms similar to K deficiency. The objectives of this experiment were to determine if SCN infection affected macronutrient concentrations in soybean seedling vegetative tissues, determine whether increased K fertility can overcome these possible effects, and to determine if these possible effects are localized at the site of infection or expressed systemically throughout the root system. Soybean plants were grown with root systems split into two halves. This allowed differential K (0.2, 2.4 and 6.0 mM K nutrient solutions) and SCN (0 and 15 000 eggs/plant) treatments to be applied to opposite root-halves of the same plant. Thirty days after plants were inoculated with SCN, macronutrient concentrations of shoot and root tissues were determined. Potassium concentration in leaf blades was not affected; but K concentrations in leaf-petiole and stem tissues were increased with SCN infection. Roots infected with SCN contained lower K concentrations than uninfected roots, but only for the 2.4 mM K treatment. Thus, at the medium level of K fertility, SCN reduced K concentration in soybean roots, and increasing K fertility to the high level overcame the effect. Because K concentrations in the shoot tissues were not reduced by SCN infection, above ground portions of the plant may be able to overcome limitations that occur in roots during the first 30 days of infection. Increasing K fertility level in soybean fields may not benefit vegetative growth of soybean infected with SCN.  相似文献
