Ecophysiological models predict quantitative traits of one genotype in any environment, whereas quantitative trait locus (QTL) models predict the contribution of alleles to quantitative traits under a limited number of environments. We have combined both approaches by dissecting into effects of QTLs the parameters of a model of maize (Zea mays) leaf elongation rate (LER; H. Ben Haj Salah, F. Tardieu [1997] Plant Physiol 114: 893-900). Response curves of LER to meristem temperature, water vapor pressure difference, and soil water status were established in 100 recombinant inbred lines (RILs) of maize in six experiments carried out in the field or in the greenhouse. All responses were linear and common to different experiments, consistent with the model. A QTL analysis was carried out on the slopes of these responses by composite interval mapping confirmed by bootstrap analysis. Most QTLs were specific of one response only. QTLs of abscisic acid concentration in the xylem sap colocalized with QTLs of response to soil water deficit and conferred a low response. Each parameter of the ecophysiological model was computed as the sum of QTL effects, allowing calculation of parameters for 11 new RILs and two parental lines. LERs were simulated and compared with measurements in a growth chamber experiment. The combined model accounted for 74% of the variability of LER, suggesting that it has a general value for any RIL under any environment.  相似文献

Differential polyadenylation is a widespread mechanism in higher eukaryotes producing mRNAs with different 3' ends in different contexts. This involves several alternative polyadenylation sites in the 3' UTR, each with its specific strength. Ralstonia solanacearum is a plant pathogenic bacterium that undergoes a spontaneous phenotypic conversion (PC) from a wild-type pathogenic to a non-pathogenic form. PC is often associated with mutations in phcA, which is a key virulence regulatory gene. Until now, reversion to the wild-type pathogenic form has not been observed for PC variants and the biological significance of PC has been questioned. In this study, we characterized various alterations in phcA (eight IS element insertions, three tandem duplications, seven deletions and a base substitution) in 19 PC mutants from the model strain GMI1000. In five of these variants, reversion to the pathogenic form was observed in planta, while no reversion was ever noticed in vitro whatever culture media used. However, reversion was observed for a 64 bp tandem duplication in vitro in the presence of tomato root exudate. This is the first report showing a complete cycle of phenotypic conversion/reversion in a plant pathogenic bacterium.  相似文献

Stop-flow studies were used to characterize solute uptake inisolated rat lungs. These lungs were perfused at 8 or 34 ml/min for10-28 s with solutions containing125I-albumin and two or more ofthe following diffusible indicators: [3H]mannitol,[14C]urea,3HOH,201Tl+,or86Rb+.After this loading period, flow was stopped for 10-300 s and thenresumed to flush out the perfusate that remained in the pulmonary vasculature during the stop interval. Concentrations of201Tl+and86Rb+in the venous outflow decreased after the stop interval, indicating uptake from exchange vessels during the stop interval. The amount ofthese K+ analogs lost from thecirculation during the stop interval was greater when the intervalswere longer. However, losses of201Tl+at 90 s approached those at 300 s. Because extraction continued afterthe vasculature had been flushed, vascular levels had presumably fallento negligible levels during the stop interval. By 90 s of stop flow thevascular volume that was cleared of201Tl+averaged 0.657 ± 0.034 (SE) ml in the experiments perfused at 8 ml/min and 0.629 ± 0.108 ml in those perfused at 34 ml/min. Increases in perfusate K+decreased the cleared volumes of201Tl+and86Rb+.Uptake of[3H]mannitol,[14C]urea, and3HOH during the stop intervals wasobserved only when the lungs were loaded at high flow for shortintervals. Decreases in201Tl+and86Rb+concentrations in the pulmonary outflow can be used to identify thefraction of the collected samples that were within exchange vessels ofthe lung during the stop interval and may help determine thedistribution of solute and water exchange along the pulmonary vasculature.

J. Appl.Physiol. 84(1): 47-52, 1998.The stop-flow approach was used to investigate where filtration occurs in the pulmonary vasculature after elevation of left atrial pressure andaspiration of HCl. Rat lungs were perfused for 11 min at zero leftatrial pressures, and then flow was stopped for 10 min and left atrialpressures were increased to 20 cmH2O. Thereafter, 3HOH was instilled into the airspaces, and the pulmonary vasculature was flushed by perfusing it fromthe pulmonary artery to left atrium (anterograde flush) or in theopposite direction (retrograde flush). Increases in fluoresceinisothiocyanate (FITC)-dextran (molecular weight 2,000,000) indicatedfiltration, and these preceded increases in3HOH after anterograde but notretrograde flushes. This suggests that some filtration occurred throughvessels that were relatively venous compared with those through which3HOH exchange had occurred.Filtration increased fivefold after instillation of 0.1 N HCl inisotonic saline into the air spaces before perfusion. Increases inEvans blue-labeled albumin concentrations were <40% those ofFITC-dextran, indicating loss from the vasculature, but increases inunlabeled albumin and FITC-albumin were comparable.

Detailed observations of larval Drosophila chemotaxis have characterised the relationship between the odour gradient and the runs, head casts and turns made by the animal. We use a computational model to test whether hypothesised sensorimotor control mechanisms are sufficient to account for larval behaviour. The model combines three mechanisms based on simple transformations of the recent history of odour intensity at the head location. The first is an increased probability of terminating runs in response to gradually decreasing concentration, the second an increased probability of terminating head casts in response to rapidly increasing concentration, and the third a biasing of run directions up concentration gradients through modulation of small head casts. We show that this model can be tuned to produce behavioural statistics comparable to those reported for the larva, and that this tuning results in similar chemotaxis performance to the larva. We demonstrate that each mechanism can enable odour approach but the combination of mechanisms is most effective, and investigate how these low-level control mechanisms relate to behavioural measures such as the preference indices used to investigate larval learning behaviour in group assays.  相似文献

Ducks and seabirds are natural hosts for influenza A viruses (IAV). On oceanic islands, the ecology of IAV could be affected by the relative diversity, abundance and density of seabirds and ducks. Seabirds are the most abundant and widespread avifauna in the Western Indian Ocean and, in this region, oceanic islands represent major breeding sites for a large diversity of potential IAV host species. Based on serological assays, we assessed the host range of IAV and the virus subtype diversity in terns of the islands of the Western Indian Ocean. We further investigated the spatial variation in virus transmission patterns between islands and identified the origin of circulating viruses using a molecular approach. Our findings indicate that terns represent a major host for IAV on oceanic islands, not only for seabird-related virus subtypes such as H16, but also for those commonly isolated in wild and domestic ducks (H3, H6, H9, H12 subtypes). We also identified strong species-associated variation in virus exposure that may be associated to differences in the ecology and behaviour of terns. We discuss the role of tern migrations in the spread of viruses to and between oceanic islands, in particular for the H2 and H9 IAV subtypes.  相似文献

ObjectivesThe aim of this study was to investigate to what extent 9 to 11 year old children participating in a specific sport already exhibit a specific anthropometric, physical fitness and motor coordination profile, in line with the requirements of that particular sport. In addition, the profiles in children with a different training volume were compared and possible differences in training hours per week between children from a low, moderate, and high level of physical fitness and motor coordination were investigated.DiscussionThe study showed that in general, children at a young age do not exhibit sport-specific characteristics, except in children with a high training volume. It is possible that on the one hand, children have not spent enough time yet in their sport to develop sport-specific qualities. On the other hand, it could be possible that they do not take individual qualities into account when choosing a sport.  相似文献
