Spirometry, roentgenography and electrocardiography were performed during community health surveys in 1976-78 in 176 Inuit and other long-time residents of the northeastern (Arctic Bay) and western (Inuvik) Canadian Arctic, and the results were related to age, ethnic origin, occupation and history of climatic exposure, smoking and hospitalization for tuberculosis. In Arctic Bay the young men showed excellent respiratory function, normal-sized pulmonary arteries and normal electrocardiograms, but abnormalities of all three types were increasingly frequent and severe after age 25. The forced mid-expiratory flow (FMF) fell to less than 50% of the norm by age 40, and dilatation of the pulmonary artery, hypertrophy of the right ventricle, right bundle branch block and a pseudoinfarction pattern on the ECG were frequently associated. In contrast, the men in Inuvik, an urbanized centre, maintained above normal respiratory function until age 40, and the FMF and pulmonary artery diameter remained normal in the older men except for Inuit and white trappers over 60 years old who had run fox trap lines along the Arctic coast in the 1920s and 30s. These data suggest that inhalation of extremely cold air at maximum ventilation may be a prime factor in the chronic obstructive lung disease of Inuit hunters, whereas smoking has only a minor role and hospitalization for tuberculosis appears to protect from rather than contribute to this disorder.  相似文献

An improved and simplified enzyme-linked immunosorbent assay (ELISA) was developed for the detection and quantification of parasporal crystalline toxins from Bacillus thuringiensis subsp. kurstaki. The improved procedure involved pretreatment of the polystyrene cuvettes with glutaraldehyde before antibody coating. A direct comparison of treated and untreated cuvettes is provided. ELISAs were then used for the analysis of the entomocidal crystalline proteins in commercial and experimental formulations of B. thuringiensis subspp. kurstaki and israelensis.  相似文献

An lagerndem Winterknoblauch ist unter außenluft‐(x + 6, 5 bzw. 8, 3 °C) und maschinengekühlten Bedingungen (x‐1 . . .‐2 °C) in zwei Lagerperioden die Entwicklung der einzelnen Fäuleerreger verfolgt und in Abhängigkeit von der Lagerdauer statistisch quantifiziert worden. An Hand des Masseanteiles befallener Zwiebeln und der Befallsintensität wird eine zunehmende Ausbreitung von Penicillium spp. und einer Gruppe mit Befall durch mehrere Fäuleerreger unter beiden Lagerbedingungen und von einer Botrytis‐Species (vermutlich B. porri Buchw.) im maschinengekühlten Lager belegt. Fäuleverluste durch Helminthosporium allii Campanile und Bakterien zeigen dagegen mit fortschreitender Lagerdauer einen abnehmenden Verlauf. Mit Eintreten von lagerungsbedingter Seneszenz steigen die Verluste progressiv an. Kaltlagerbedingungen verzögern dagegen ihre Ausbreitung. Die Verluste durch alle Fäulerreger (Fäule gesamt) zeigen eine gleichmäßige Zunahme während der Lagerdauer.  相似文献

The ambr bioreactors are single‐use microbioreactors for cell line development and process optimization. With operating conditions for large‐scale biopharmaceutical production properly scaled down, microbioreactors such as the ambr15? can potentially be used to predict the effect of process changes such as modified media or different cell lines. While there have been some recent studies evaluating the ambr15? technology as a scale‐down model for fed‐batch operations, little has been reported for semi‐continuous or continuous operation. Gassing rates and dilution rates in the ambr15? were varied in this study to attempt to replicate performance of a perfusion process at the 5 L scale. At both scales, changes to metabolite production and consumption, and cell growth rate and therapeutic protein production were measured. Conditions were identified in the ambr15? bioreactor that produced metabolic shifts and specific metabolic and protein production rates that are characteristic of the corresponding 5 L perfusion process. A dynamic flux balance (DFB) model was employed to understand and predict the metabolic changes observed. The DFB model predicted trends observed experimentally, including lower specific glucose consumption and a switch from lactate production to consumption when dissolved CO2 was maintained at higher levels in the broth. © 2017 American Institute of Chemical Engineers Biotechnol. Epstein-Barr virus (EBV) is capable of adopting three distinct forms of latency: the type III latency program, in which six EBV-encoded nuclear antigens (EBNAs) are expressed, and the type I and type II latency programs, in which only a single viral nuclear protein, EBNA1, is produced. Several groups have reported heavy CpG methylation of the EBV genome in Burkitt's lymphoma cell lines which maintain type I latency, and loss of viral genome methylation in tumor cell lines has been correlated with a switch to type III latency. Here, evidence that the type III latency program must be inactivated by methylation to allow EBV to enter the type I or type II restricted latency program is provided. The data demonstrates that the EBNA1 gene promoter, Qp, active in types I and II latency, is encompassed by a CpG island which is protected from methylation. CpG methylation inactivates the type III latency program and consequently allows the type I or II latency program to operate by alleviating EBNA1-mediated repression of Qp. Methylation of the type III latency EBNA gene promoter, Cp, appears to be essential to prevent type III latency, since EBNA1 is expressed in all latently infected cells and, as shown here, is the only viral antigen required for activation of Cp. EBV is thus a pathogen which subverts host-cell-determined methylation to regulate distinct genetic programs.  相似文献

The introduced parasitoid,Aphytis melinusDeBach (Hymenoptera: Aphelinidae), is used for augmentative biological control of California red scale,Aonidiella aurantii(Maskell) (Homoptera: Diaspididae). Commercially reared wasps are reared on oleander scale,Aspidiotus neriiBouché (Homoptera: Diaspididae). Oleander scale covers lack the chemical,O-caffeoyltyrosine, a kairomone mediating host selection byA. melinus.Wasps reared on oleander scale but individually exposed, or primed, toO-caffeoyltyrosine more readily accepted California red scale for probing in laboratory bioassays and parasitized a greater proportion of available California red scale in the field than wasps reared similarly but not exposed toO-caffeoyltyrosine. Thus, it may be possible to improve host recognition of commercial, insectary-rearedA. melinusby exposing them toO-caffeoyltyrosine prior to release. The goal of this study was to develop an inexpensive but effective means of priming thousands of wasps simultaneously toO-caffeoyltyrosine. The most effective method, but potentially the most expensive, was simply to spray parasitized oleander scale on their host plant with diluteO-caffeoyltyrosine prior to wasp emergence. In additional experiments, using controlled doses of syntheticO-caffeoyltyrosine applied to scale covers, we showed that primed wasps require both a lower minimum dose ofO-caffeoyltyrosine for recognition and also respond to measuredO-caffeoyltyrosine doses more consistently than unprimed wasps. The ability to mass-prime thousands of wasps prior to release is a crucial step toward realizing the concept of behavioral improvement of host selection of parasitoids on a commercial scale.  相似文献

Abstract. Despite international recognition that alvar habitats are important reservoirs of biodiversity, they remain little studied in North America. In this paper, the results are reported on an investigation of alvars in the central portion of their known distribution on this continent. 210 plots were distributed among seven sites and were quantitatively sampled for vascular plants, lichens, bryophytes and a suite of environmental variables. Detrended and Canonical Correspondence Analyses and other methods were used to investigate differences among alvars, within alvars and between alvars and adjacent forested habitats. The plant communities and environmental conditions were highly similar among sites in the study region, yet very different from surrounding habitats. There were abrupt changes in vegetation and environmental conditions from alvar to forest, without the presence of transition zones in the vegetation or environmental gradients as the forest was approached. The environmental factors associated with the change from alvar to forest and with variation within alvar habitat were examined. Some alvars in the study were found to contain stunted, slow-growing trees reaching ages of 524 yr. These same sites appear to have remained unburned for several centuries, while other sites likely burned 90 yr ago. The plant communities were very similar between the alvars that lacked a major, biomass-removing disturbance in centuries and alvars that had experienced catastrophic fire relatively recently. Maintenance of the plant communities and open nature of alvars appears site-specific rather than habitat specific.  相似文献
