The emerald ash borer (Agrilus planipennis, Coleoptera: Buprestidae) is a wood-boring invasive pest devastating North American ash (Fraxinus spp.). A. planipennis overwinters primarily as a freeze-avoiding prepupa within the outer xylem or inner bark of the host tree. The range of this
species is expanding outward from its presumed introduction point in southwestern Michigan. We hypothesized that loss of cold
tolerance in response to mid-winter warm spells could limit survival and northern distribution of A. planipennis. We determined whether winter-acclimatised A. planipennis prepupae reduced their cold tolerance in response to mid-winter warm periods, and whether this plasticity was reversible
with subsequent cold exposure. Prepupae subjected to mid-winter warm spells of 10 and 15°C had increased supercooling points
(SCPs) and thus reduced cold tolerance. This increase in SCP was accompanied by a rapid loss of haemolymph cryoprotectants
and the loss of cold tolerance was not reversed when the prepupae were returned to −10°C. Exposure to temperatures fluctuating
from 0 to 4°C did not reduce cold hardiness. Only extreme warming events for several days followed by extreme cold snaps may
have lethal effects on overwintering A. planipennis populations. Thus, distribution in North America is likely to be limited by the presence of host trees rather than climatic
factors, but we conclude that range extensions of invasive species could be halted if local climatic extremes induce unidirectional

We exploit bacterial sortases to attach a variety of moieties to the capsid proteins of M13 bacteriophage. We show that pIII, pIX, and pVIII can be functionalized with entities ranging from small molecules (e.g., fluorophores, biotin) to correctly folded proteins (e.g., GFP, antibodies, streptavidin) in a site-specific manner, and with yields that surpass those of any reported using phage display technology. A case in point is modification of pVIII. While a phage vector limits the size of the insert into pVIII to a few amino acids, a phagemid system limits the number of copies actually displayed at the surface of M13. Using sortase-based reactions, a 100-fold increase in the efficiency of display of GFP onto pVIII is achieved. Taking advantage of orthogonal sortases, we can simultaneously target two distinct capsid proteins in the same phage particle and maintain excellent specificity of labeling. As demonstrated in this work, this is a simple and effective method for creating a variety of structures, thus expanding the use of M13 for materials science applications and as a biological tool. 相似文献

The primary objective of this investigation was to identify which components of endurance training (e.g., modality, duration, frequency) are detrimental to resistance training outcomes. A meta-analysis of 21 studies was performed with a total of 422 effect sizes (ESs). Criteria for the study included were (a) compare strength training alone to strength plus endurance training (concurrent) or to compare combinations of concurrent training; (b) the outcome measures include at least one measure of strength, power, or hypertrophy; and (c) the data necessary to calculate ESs must be included or available. The mean ES for hypertrophy for strength training was 1.23; for endurance training, it was 0.27; and for concurrent training, it was 0.85, with strength and concurrent training being significantly greater than endurance training only. The mean ES for strength development for strength training was 1.76; for endurance training, it was 0.78; and for concurrent training, it was 1.44. Strength and concurrent training was significantly greater than endurance training. The mean ES for power development for strength training only was 0.91; for endurance training, it was 0.11; and for concurrent training, it was 0.55. Significant differences were found between all the 3 groups. For moderator variables, resistance training concurrently with running, but not cycling, resulted in significant decrements in both hypertrophy and strength. Correlational analysis identified significant negative relationships between frequency (-0.26 to -0.35) and duration (-0.29 to -0.75) of endurance training for hypertrophy, strength, and power. Significant relationships (p < 0.05) between ES for decreased body fat and % maximal heart rate (r = -0.60) were also found. Our results indicate that interference effects of endurance training are a factor of the modality, frequency, and duration of the endurance training selected. 相似文献

Tallgrass prairie is among the most endangered ecosystems in North America. High‐diversity restorations protect remnant habitat and expand native communities. Excluding land acquisition, the most expensive step in restoration is procuring seed. Given this cost, managers want to maximize seedling establishment. Native species that flower and ripen early in the growing season are included in a diverse seeding mix but as a group they have not successfully established. For early‐maturing species, the practice of storing seeds in a cold room from harvest until sowing in the dormant season effectively eliminates exposure to the summer conditions seeds would naturally have in the wild. In this study, we compared the effect of summer sowing timing and winter sowing timing on establishment in field conditions. In August 2004 and December 2004, we broadcast a seed mix of seven early‐maturing species: Antennaria plantaginifolia (L.) Richardson. (Pussy toes), Arabis lyrata L. (Sand cress), Carex swanii (Fernald) Mack. (Downy green sedge), Hymenopappus scabiosaeus L'Hér. (Old plainsman), Lupinus perennis L. ssp. perennis var. occidentalis S. Watson. (Wild lupine), Phlox bifida Beck. (Sand phlox) and Hesperostipa spartea (Trin.) Barkworth. (Porcupine grass). We collected data on establishment and reproductive success at 12 time points from June 2005 until October 2008. Species established one growing season sooner when planted at the summer sowing time, and diversity in the summer sowing plots was higher after 4 years. Quicker establishment may have benefits such as providing early competition from weeds that may outweigh additional effort required to ensure timely planting. 相似文献

During an unusual cold‐water event in January 2010, reefs along the Florida Reef Tract suffered extensive coral mortality, especially in shallow reef habitats in close proximity to shore and with connections to coastal bays. The threatened staghorn coral, Acropora cervicornis, is the focus of propagation and restoration activities in Florida and one of the species that exhibited high susceptibility to low temperatures. Complete mortality of wild staghorn colonies was documented at 42.9% of donor sites surveyed after the cold event. Remarkably, 72.7% of sites with complete A. cervicornis mortality had fragments surviving within in situ coral nurseries. Thus, coral nurseries served as repositories for genetic material that would have otherwise been completely lost from donor sites. The location of the coral nurseries at deeper habitats and distanced from shallow nearshore habitats that experienced extreme temperature conditions buffered the impacts of the cold‐water event and preserved essential local genotypes for future Acropora restoration activities. 相似文献

Culex pipiens quinquefasciatus were fed blood meals from a live chicken (LC), chicken blood in Alsever's (AC) solution, defibrinated bovine blood (DB), or bovine blood in citrate (CB) and incubated at 28° C. The effects of different blood meal sources were evaluated with respect to rates of blood feeding and reproduction (i.e., fecundity and fertility) over two gonotrophic cycles. Mosquitoes that fed on the first blood meal were subjected to a second blood meal as follows (first blood meal / second blood meal): LC/LC, LC/DB, DB/DB, CB/CB, AC/AC. Fecundity and fertility of Cx. p. quinquefasciatus were significantly (P < 0.05) greater in mosquitoes fed LC blood; however, fecundity and fertility in different treatment groups varied by gonotrophic cycle. These results contribute to our understanding of the impact of blood meal source on feeding and reproduction in Cx. p. quinquefasciatus. The potential impacts of blood meal source on virus transmission experiments are discussed. 相似文献

Longline fishing is the most common elasmobranch capture method around the world, yet the physiological consequences of this technique are poorly understood. To quantify the sub-lethal effects of longline capture in the commonly exploited Caribbean reef shark (Carcharhinus perezi), 37 individuals were captured using standard, mid-water longlines. Hook timers provided hooking duration to the nearest minute. Once sharks were landed, blood samples were taken and used to measure a suite of physiological parameters. Control data were obtained by sampling an additional three unrestrained Caribbean reef sharks underwater at an established shark feeding site. The greatest level of physiological disruption occurred after 120-180min of hooking, whereas sharks exposed to minimal and maximal hook durations exhibited the least disturbed blood chemistry. Significant relationships were established between hooking duration and blood pH, pCO(2), lactate, glucose, plasma calcium and plasma potassium. Longline capture appears more benign than other methods assessed to date, causing a shift in the stress response from acute at the onset of capture to a sub-acute regime as the capture event progresses, apparently facilitating a degree of physiological recovery. Continued investigation into the physiological response of elasmobranchs to longline capture is vital for the effective management of such fisheries. 相似文献

Distribution ranges of plant species are related to physical variables of ecosystems that limit plant growth. Therefore, each plant species response to physical factors builds up the functional diversity of an ecosystem. The higher the species richness of an ecosystem, the larger the probability of maintaining functions and the higher the potential number of plant functional groups (FGs). Thus, the richness potentially increases the number of functions of the highly diverse Atlantic Rainforest domain in Brazil. Severe plant growth limitations caused by stress, however, decrease species richness. In the Spodosols of the Mussununga, an associated ecosystem of Atlantic Rainforest, the percentage of fine sand is directly related to water retention. Moreover, the depth of the cementation layer in the Mussununga??s sandy soil is a physical factor that can affect the plants?? stress gradients. When a shallow cementation layer depth is combined with low water retention in soils and with low fine sand percentage, the double stresses of flooding in the rainy season and water scarcity in the dry season result. This study aimed to identify FGs among Mussununga plant species responding to water stress gradients of soil and to verify the effects of the gradients on plant species richness of the Mussununga. A canonical correspondence analysis (CCA) of species abundance and soil texture variables was performed on 18 plots in six physiognomies of the Mussununga. Species richness rarefactions were calculated for each vegetation form to compare diversity. The two main axes of the CCA showed two FGs responding to soil texture and cementation layer depth: stress tolerator species and mesic species. Physical variables affect plant diversity, with species richness rising as the fine sand proportion also rises in the Mussununga. The effect of the cementation layer is not significantly related to species richness variation. 相似文献
