Neomycin and related aminoglycosidic antibiotics displace calcium from synaptosomes of guinea pig cerebral cortex and from preparations of phosphatidylinositol diphosphate. At low drug concentrations, inhibition of synaptosomal calcium binding is competitive (Ki = 3·10−5M), at high concentrations it is non-competitive (Ki = 4·10−4M). Monomolecular films of phosphatidylinositol diphosphate are contracted by low concentrations of neomycin in the subphase, and are expanded at high concentrations. This expansion persists even at the collapse pressure indicating a strong interaction between the drug and the lipid.  相似文献

There is a need to develop new scaffold materials with controlled surface properties for tissue engineering applications. For that purpose novel biodegradable poly[(organo)phosphazenes] were synthesized. A cell-binding molecule, galactose, was introduced via a spacer, either 6-aminohexanol (AH) or poly(ethylene glycol) (PEG; M(w) = 3400). Some polymers were substituted with an additional PEG chain of different molecular weights (M(w) = 750 or 5000). The polyphosphazene derivatives were characterized by 1H NMR. T(g) and T(m) were determined using differential scanning calorimetry. A detailed surface analysis of the polymers using X-ray photoelectron spectroscopy (XPS), secondary ion mass spectroscopy (SIMS), and dynamic contact angle (DCA) measurements was performed. Typical backbone and side chain fragments were detected by SIMS and confirmed the polymer composition. Compared to that of the reference polymer (having only amino acid ester side groups), an increased value of the specific ether carbon groups from PEG confirmed the enrichment of PEG at the surface of PEG-Gal polymers. However, the values were lower than expected. DCA studies showed that the galactose moieties were present at the surface after exposure to an aqueous environment. XPS results confirmed the similarity between experimental and theoretical values for the AH-Gal polymers. This indicated the presence of galactose moieties at the surface, which was confirmed by the DCA data because the contact angles were low compared to those of the other polymers.  相似文献

We report on the feasibility of applying porous gelatin hydrogels, prepared by a novel and controlled cryogenic treatment, as cell-interactive scaffolds for tissue engineering applications. Despite the large number of publications on gelatin as a biomaterial, a detailed study of screening a limited number of gelatin scaffolds for their interaction with a panel of human cells has, to the best of our knowledge, not yet been published. In the present work, we have evaluated two types of porous gelatin scaffolds that differ in their pore geometry and pore size. Type I hydrogels contained top-to-bottom transverse channels (i.e. cones) with a decreasing diameter from the top (330 microm) to the bottom (20-30 microm). Type II hydrogels contained spherical pores with a diameter of 135 microm. Both types of scaffolds were evaluated by confocal laser scanning microscopy in terms of adhesion, spreading, and proliferation of human cells (endothelial, epithelial, fibroblast, glial, and osteoblast) by visualizing cells using calcein-acetoxy methyl ester as a vital stain. The results indicated that cells attached, spread, and proliferated on both types of hydrogels. In addition, the scaffolds developed can be used for the long-term culturing of human cells.  相似文献

Switchgrass (Panicum virgatum L.) is being developed into a perennial, herbaceous, cellulosic feedstock crop for use in temperate regions of the USA. Information on spatial and temporal variation for stands and biomass yield among and within fields in large agroecoregions is not available. Spatial and temporal variation information is needed to model feedstock availability for biorefineries. In this 5-yr study, the spatial and temporal variation for biomass yield and stands was determined among and within 10 fields located in North Dakota, South Dakota, and Nebraska. Switchgrass fields were managed for bioenergy from 2000 to 2004 for the Nebraska locations and 2001 to 2005 for the South Dakota and North Dakota locations. A global positioning system (GPS) receiver was used to repeatedly measure within field quadrat sites for switchgrass stands using frequency grid (2.25 m2) measurements in June for five growing seasons. Sixteen quadrat (≥1 m2) yield samples were taken post-killing frost in the establishment year and in August in subsequent years at each location. Topographic within field effects on switchgrass stand frequency and biomass yields were largely insignificant. Stands tended to increase from establishment year to year 3 and then begin to plateau. Weather factors, which were the principal source of temporal variation, were more important in switchgrass yield variation than on switchgrass stand frequencies. Temporal standard deviations for yield were higher on quadrat sites with higher than average field means while temporal standard deviations were smaller in quadrat sites that had lower than average field means at six locations. In the Northern Great Plains agroecoregion, there is greater temporal and spatial variation for switchgrass biomass yields among fields than within fields. Results indicate that modeling feedstock availability for a biorefinery can be based on field scale yields.  相似文献

The binding of polyphosphoinositides (phosphatidylinositol phosphate and phosphatidylinositol bisphosphate) to the antibiotic neomycin is utilized for the purification of these lipids. Neomycin is reductively coupled to reactive glass beads (Glycophase-CPG) and serves as the stationary phase in column chromatography. A total lipid extract is prepared from tissues with chloroform-methanol-KC1 or chloroform-methanol-HC1 and washed once with acidified methanol-water. After the addition of an equal volume of methanolic 200 mM ammonium acetate, the extract is directly applied to the column. All lipids but the polyphosphoinositides are removed from the column by rinsing with 150 mM ammonium acetate in chloroform-methanol-water. Increasing the salt concentration to 600 mM elutes phosphatidylinositol phosphate. While further increases in ionic strength are not sufficient for a quantitative removal of phosphatidylinositol bisphosphate, the lipid is completely eluted by the addition of either ammonia or HC1 to the solvent. The column can be recycled and used repeatedly.  相似文献

There is currently renewed interest in assessing the feasibility of the sterile insect technique (SIT) to control African malaria vectors in designated areas. The SIT relies on the sterilization of males before mass release, with sterilization currently being achieved through the use of ionizing radiation. This paper reviews previous work on radiation sterilization of Anopheles mosquitoes. In general, the pupal stage was irradiated due to ease of handling compared to the adult stage. The dose-response curve between the induced sterility and log (dose) was shown to be sigmoid, and there was a marked species difference in radiation sensitivity. Mating competitiveness studies have generally been performed under laboratory conditions. The competitiveness of males irradiated at high doses was relatively poor, but with increasing ratios of sterile males, egg hatch could be lowered effectively. Males irradiated as pupae had a lower competitiveness compared to males irradiated as adults, but the use of partially-sterilizing doses has not been studied extensively. Methods to reduce somatic damage during the irradiation process as well as the use of other agents or techniques to induce sterility are discussed. It is concluded that the optimal radiation dose chosen for insects that are to be released during an SIT programme should ensure a balance between induced sterility of males and their field competitiveness, with competitiveness being determined under (semi-) field conditions. Self-contained 60Co research irradiators remain the most practical irradiators but these are likely to be replaced in the future by a new generation of high output X ray irradiators.  相似文献

There is a lack of methods suitable for generation of data about the dynamics of effects on cell membranes with a high sensitivity. Such methods are urgently needed to support the optimisation of interaction of substances, particles or materials with cell. The goal of this article is to use an improved microhole chip system to monitor the alterations of cells due to the interactions of polymer-DNA complexes. This should demonstrate exemplarily that subtoxic effect of biological relevant particles or substances at relevant concentrations can be monitored for several hours. By using a microhole cell chip and a microfluidic unit single cells can be electrically interfaced via microholes and the use of small electrodes with high impedances is not necessary. For separation and positioning of the cells onto the hole negative pressure is applied on the reverse side of the chip. Under cell culture conditions the cell starts to spread on the biocompatible insulating chip membrane resulting in a stable interface to an adherent growing cell. After the spreading process is finished, the polymer/polyplex solution is added and the impedance is measured with respect to time. To illustrate the cellular parameter which can affect the measured impedance a simple simulation based on the finite element method (FEM) is performed. It was shown for the first time that the impedance-based method predicated on the microhole chip can be used for biological relevant substances at relevant concentrations and that it is more sensitive than the well-established biological marker.  相似文献

Due to their biocompatibility, biodegradability, and low immunogenicity, recombinant spider silk proteins have a high potential for a variety of applications when processed into morphologies such as films, capsules, beads, or hydrogels. Here, hydrogels made of the engineered and recombinantly produced spider silk protein eADF4(C16) were analyzed in detail. It has previously been shown that eADF4(C16) nanofibrils self-assemble by a mechanism of nucleation-aggregation, providing the basis of silk hydrogels. We focused on establishing a reproducible gelation process by employing different protein concentrations, chemical crosslinking, and functionalization of eADF4(C16) with fluorescein. Fluorescein strongly influenced assembly as well as the properties of the hydrogels, such as pore sizes and mechanical behavior, possibly due to its interference with packing of silk nanofibrils during hydrogel formation.  相似文献

Hydrogels are physically or chemically cross-linked polymer networks that are able to absorb large amounts of water. They can be classified into different categories depending on various parameters including the preparation method, the charge, and the mechanical and structural characteristics. The present review aims to give an overview of hydrogels based on natural polymers and their various applications in the field of tissue engineering. In a first part, relevant parameters describing different hydrogel properties and the strategies applied to finetune these characteristics will be described. In a second part, an important class of biopolymers that possess thermosensitive properties (UCST or LCST behavior) will be discussed. Another part of the review will be devoted to the application of cryogels. Finally, the most relevant biopolymer-based hydrogel systems, the different methods of preparation, as well as an in depth overview of the applications in the field of tissue engineering will be given.  相似文献

This article describes the synthesis of biodegradable polyphosphazenes. The rate of degradation can be varied in a controllable manner by the introduction of hydrolysis-sensitive amino acid ester side groups or by blending of polymers. Biodegradable polyphosphazenes can be used for the preparation of drug-containing implants and this is illustrated for devices containing the cytostatic agent mitomycin C. This article reviews data about the degradation characteristics of poly[(amino acid ester)phosphazene] derivatives that have been discussed previously.
