Ostreococcus tauri, a unicellular marine green alga, is the smallest known free-living eukaryote and is ubiquitous in the surface oceans. The ecological success of this organism has been attributed to distinct low- and high-light-adapted ecotypes existing in different niches at a range of depths in the ocean. Viruses have already been characterized that infect the high-light-adapted strains. Ostreococcus tauri virus (OtV) isolate OtV-2 is a large double-stranded DNA algal virus that infects a low-light-adapted strain of O. tauri and was assigned to the algal virus family Phycodnaviridae, genus Prasinovirus. Our working hypothesis for this study was that different viruses infecting high- versus low-light-adapted O. tauri strains would provide clues to propagation strategies that would give them selective advantages within their particular light niche. Sequence analysis of the 184,409-bp linear OtV-2 genome revealed a range of core functional genes exclusive to this low-light genotype and included a variety of unexpected genes, such as those encoding an RNA polymerase sigma factor, at least four DNA methyltransferases, a cytochrome b(5), and a high-affinity phosphate transporter. It is clear that OtV-2 has acquired a range of potentially functional genes from its host, other eukaryotes, and even bacteria over evolutionary time. Such piecemeal accretion of genes is a trademark of large double-stranded DNA viruses that has allowed them to adapt their propagation strategies to keep up with host niche separation in the sunlit layers of the oceanic environment.  相似文献

Introduction

Minimally invasive radical prostatectomy (RP) (robotic and laparoscopic), have brought improvements in the outcomes of RP due to improved views and increased degrees of freedom of surgical devices. Robotic and laparoscopic surgeries do not incorporate haptic feedback, which may result in complications secondary to inadequate tissue dissection (causing positive surgical margins, rhabdosphincter damage, etc). We developed a micro-engineered device (6 mm2 sized) [E-finger]) capable of quantitative elasticity assessment, with amplitude ratio, mean ratio and phase lag representing this. The aim was to assess the utility of the device in differentiating peri-prostatic tissue types in order to guide prostate dissection.

Material and Methods

Two embalmed and 2 fresh frozen cadavers were used in the study. Baseline elasticity values were assessed in bladder, prostate and rhabdosphincter of pre-dissected embalmed cadavers using the micro-engineered device. A measurement grid was created to span from the bladder, across the prostate and onto the rhabdosphincter of fresh frozen cadavers to enable a systematic quantitative elasticity assessment of the entire area by 2 independent assessors. Tissue was sectioned along each row of elasticity measurement points, and stained with haematoxylin and eosin (H&E). Image analysis was performed with Image Pro Premier to determine the histology at each measurement point.

Results

Statistically significant differences in elasticity were identified between bladder, prostate and sphincter in both embalmed and fresh frozen cadavers (p = <0.001). Intra-class correlation (ICC) reliability tests showed good reliability (average ICC = 0.851). Sensitivity and specificity for tissue identification was 77% and 70% respectively to a resolution of 6 mm2.

Conclusions

This cadaveric study has evaluated the ability of our elasticity assessment device to differentiate bladder, prostate and rhabdosphincter to a resolution of 6 mm2. The results provide useful data for which to continue to examine the use of elasticity assessment devices for tissue quality assessment with the aim of giving haptic feedback to surgeons performing complex surgery.  相似文献

Uterine flushings, cervical mucus swabs and udder secretions collected at weekly intervals from five mixed breed beef cows (four Brucella abortus strain 19 vaccinates, and 1 non-vaccinate) were cultured for Brucella abortus . Prior to sampling, four of the five had aborted 7-to 8-month-old fetuses and one gave brith to a weak calf. The fetuses and/or udder secretions from the cows were culture positive for B. abortus at the time of parturition. Three of the cows developed persistent udder infections. Two of these cows were also shown to have brucellae in their cervical mucus for 10 and 20 days and in their uterine flushings for 17 and 41 days after parturition, respectively. One other cow had brucellae in the cervical mucus for 16 days and in the uterine flushings for up to 36 days post-abortion. All attempts to isolate the organism from this cow's udder secretions in culture were negative. In two cows with culture-positive uterine flushings, isolations of brucellae were made subsequent to normal postpabortion return to estrus.  相似文献

Volatile compounds produced by Pseudomonas perolens ATCC 10757 in sterile fish muscle (Sebastes melanops) were identified by combined gas-liquid chromatography and mass spectrometry. Compounds positively identified included methyl mercaptan, dimethyl disulfide, dimethyl trisulfide, 3-methyl-1-butanol, butanone, and 2-methoxy-3-isopropylpyrazine. Compounds tentatively identified included 1-penten-3-ol and 2-methoxy-3-sec-butylpyrazine. The substituted pyrazine derivative 2-methoxy-3-isopropylpyrazine was primarily responsible for the musty, potato-like odor produced by P. perolens.  相似文献

Much difficulty has been experienced in obtaining basic fuchsins satisfactory for the Feulgen technic. A method of purification employing sulfur dioxide is described which has been found to improve many unsatisfactory fuchsins. It is also shown that if the best grade of commercial pararosanilin base is used and proper precautions observed in converting it to the chloride or the acetate, a product can be obtained which gives excellent results in the Feulgen technic. The methods by which the base can be converted to the chloride or acetate are described in detail. Precautions are given for avoiding, in the conversion of the base to the dye salts, contamination of the product with the impurities which interfere with the staining. The use of the acetate is recommended because of its greater solubility.  相似文献

Abstract. A transition matrix model was used to explore the dynamics, rate and potential extent of changes in landscape vegetation patterns on a southern Texas Prosopis savanna. Transitions between seven vegetation classes were determined for the periods 1941–1960 and 1960–1983 on aerial photographs of three sites. During these periods, the sites were heavily grazed by cattle and were fire-free. Vegetation states assessed in grids of 20 m x 20 m cells superimposed on photographs ranged from grass-dominated to woody plant-dominated. The 1941–1960 period (denoted DRY) was characterized by prolonged drought, whereas annual rainfall during the 1960–1983 period (denoted WET) was typically normal to above-normal. The 1941 landscape consisted of herbaceous zones (6% of cells), woodland (50% of cells) and savanna parkland (44% of cells with grass/woody plant mixtures). The woodland state was the most stable, with probabilities of no change being 0.970 and 0.873 in WET and DRY periods, respectively. The herbaceous state was least stable, with corresponding values of 0.074 and 0.353. Past and future landscape structure was modelled by randomly selecting DRY or WET transitions at 20 year time steps. The model was run under a series of rainfall scenarios where the probability of selecting the WET transition matrix (P[WET]) ranged from 0 (DRY always chosen) to 1 (WET always chosen). Historical records indicate P[WET] has approximated 0.3 to 0.4 in the region. The rate of succession to states of greater woody cover increased as P[WET] increased. Forward simulations based on P[WET] > 0.2 suggest the present landscape is unstable and will develop into a closed-canopy woodland within the next 180 years, assuming the processes operating between 1941 and 1983 continue (e.g. grazing by cattle and lack of fire). Reverse simulations concur with historical observations and projections derived from woody plant growth rates in other studies and suggest that 200 to 300 yr BP these landscapes contained a substantially greater proportion of cells dominated by grassland or grassland with scattered woody plants (43 to 74%) than was present in 1983 (19%). Based upon elapsed time between predicted past and future steady states, succession from open savanna to closed-canopy woodland may occur in ca. 400 to 500 yr for P(WET) ≥ 0.33. Arresting or reversing the projected trend may require changes in climate and/or changes in livestock grazing and land management practices. The approaches employed in this study illustrate how time series maps, aerial photographs and satellite imagery can be analyzed and used to interpret, project and reconstruct local and regional changes in ecosystem structure. Difficulties and limitations associated with the use of Markov chains to model succession are identified and discussed.  相似文献
