Mechanism of durable resistance: a new approach   总被引:2,自引:0,他引:2  
Summary Wheat genotypes, including backcross derivatives of Thatcher carrying Lr10 and Lr23, substitution lines for Lr10 and Lr23 in Chinese Spring background and Chinese Spring and Thatcher were analysed against 21 pathotypes of leaf rust in seedling tests. Adult plant responses in all these stocks were observed in the field nurseries under exposure to the inoculum of the Indian virulent races of leaf rust. The seedling data demonstrated that both the substitution lines and the backcross derivatives for each gene carry identical pattern of infection for resistance. The high level of adult plant resistance in the substitution lines, in contrast to the backcross derivatives in Thatcher, has been postulated to be due to the combination of resistance contributed by Lr10 and adult plant Chinese Spring resistance or to Lr23 and Chinese Spring adult plant resistance. It has been suggested that genes Lr10 and Lr23 added to the Chinese Spring background provide sources for durable resistance, since Chinese Spring has continued to provide a moderate level of adult plant resistance to leaf rust for a very long time.  相似文献

Summary The effects of destruxin B, a host-specific toxin of Alternaria brassicae that causes black spot disease in oilseed brassicas, were studied on in vitro pollen germination and pollen-tube growth of Brassica campestris var brown sarson, B. juncea, B. napus cvs Westar and Cresor, B. nigra and Sinapis alba. Pollen grains of B. nigra, B. juncea and B. campestris were the most sensitive and those of S. alba the least sensitive to the toxin. Effects of the toxin were also studied on the leaves of these species, and the degree of sensitivity of leaves of different species was comparable to that of their pollen grains. The results on the responses of pollen grains as well as leaves to the toxin are in agreement with the degree of susceptibility/resistance of these species to A. brassicae reported in the literature, indicating that the genes imparting susceptibility/restistance are expressed in the pollen, a prerequisite for pollen selection. Results are also presented which show that the toxin fed to the cut end of isolated inflorescence axis is readily taken up by the developing pollen and results in the inhibition of germination of susceptible pollen. This technique offers a simple and effective method for application of selection pressure to eliminate pollen grains susceptible to the toxin from effecting fertilization.  相似文献

Summary In the stamenless-2 (sl-2) mutant of tomato (Lycopersicon esculentum Mill.), the breakdown in microsporogenesis corresponded with various abnormalities in the ultrastructure of the tapetal cell wall. In some mutant anthers, the inner tangential wall was excessively loosened allowing the passage of tapetal cell wall material and cytoplasmic contents into the anther locule. This presumably altered the osmoticum of the locule and resulted in plasmolysis of the microspores. Membranous fragments commonly observed in the normal tapetal cell wall, and presumed to have a role in transfer of materials from the tapetum to microspores, were absent from thesl-2 mutant. This was associated with reduced transfer of materials, such as lipids, to the developing pollen grains. In addition, a lining of sporopollenin-like deposits that coated the inner tangential wall of the normal tapetum, was discontinuous in the mutant. In mutant anthers where the tapetal cell wall did not loosen, the transfer of all materials was restricted and this resulted in the collapse of sporogenous material.  相似文献

Arabidopsis thaliana (L.) Heynh. has been used as a model system to investigate the regulatory genes that control and coordinate the determination, differentiation and morphogenesis of the floral meristem and floral organs. We show here that benzylaminopurine (BAP), a cytokinin, influences flower development inArabidopsis and induces partial phenocopies of known floral homeotic mutants. Application of BAP to wild-type inflorescences at three developmental stages results in: (i) increase in floral organ number; (ii) formation of abnormal floral organs and (iii) induction of secondary floral buds in the axils of sepals. These abnormalities resemble the phenotypes of mutants,clv1 (increase in organ number),ap1,ap2,ap3 (abnormal floral organs) andap1 (secondary floral buds in the axils of first-whorl organs). In addition, BAP induces secondary floral buds in the axils of perianth members ofapt2-6, ap3-1 andag mutants, and accentuates the phenotype of theapt2-1 mutant to resemble theapt2-6 mutant. These observations suggest that exogenous BAP suppresses the normal functioning of the genes for floral meristem identity and thereby affects flower development and the later stages of floral organ differentiation.Abbreviations BAP N6-benzylaminopurine - CK cytokinin  相似文献

Growth substances, α-naphthaleneacetic (NAA) and kinetin, had an important role in the regulation of lateral root (LR) formation in lettuce ( Lactuca sativa L. cv. Grand Rapids) seedling roots. NAA (10-5 M ) was a potent stimulator of LR initiation and caused a 600% increase in the number of lateral root primordia (LRP) compared to untreated roots. NAA was required for only the first 20 h of the 72 h treatment period for maximum stimulation of LRP initiation. Kinetin (2 × 10-5 M ) effectively prevented the spontaneous formation of LRP and inhibited the NAA-stimulated production of LRP. Kinetin inhibition was maximal during the first 20 h of NAA treatment and this effect was not overcome by subsequent supply of NAA. Also, lettuce roots were most sensitive to kinetin at 20 h of NAA treatment, when the first signs of cell division were observed in the pericycle.  相似文献

Summary Near isogenic/substitution lines of stem rust resistance genes in different backgrounds of Marquis, Chinese Spring and W 2691 and certain varieties with known genes for stem rust resistance were tested against each of 19 Indian cultures of stem rust races/biotypes (14, 15, 17, 21, 21A-1, 24, 34, 40, 40A, 42, 42B, 117, 117A, 117A-1, 122, 184, 194, 222 and 295). Sr 24 (Sear's 3D/Ag), Sr 24 (TR 380-27 4/3 Ag 14-White seeded recombinant with Agent type resistance), Sr 25 (Sear's 7D/Ag), Sr 26 (Eagle), Sr 26 (Knott's 6A/Ag translocation), Sr 27 (WRT 238-5), Combination line (Sr Tt1 + Sr 9b) were observed to be completely effective against all the 19 cultures tested. In addition, a number of lines, such as TAF2d (Sr Agi), Line W(Sr Tt2) and Combination III (Sr Tt1 + Sr 9e), were found to be effective against at least three of the most prevalent races (21, 40A and 117A-1) and a virulent race 122 in Indian natural population. Lines carrying genes other than Sr 2, Sr 9a, Sr 9f (Chinese Spring) and Sr 15 (Norka), and Line E were found to be resistant to one or more cultures of stem rust.The background effect upon the expression of a gene was observed by comparing the range of infection on single gene host lines in either different backgrounds and/or in cultivars with known genes for stem rust resistance against the 12 cultures of stem rust races found in India.  相似文献

Summary Two bread wheat accessions, E5008 and E6160, have been genetically analysed for resistance genes effective against Indian cultures of stem rust races, 15C and 122. The inheritance of resistance to each race has been determined from the F1 and F2 of the crosses (resistant parents with the susceptible variety, Agra Local) and F2 progenies from the backcross to Agra Local. Tests have been performed to see if the two varieties carry common genes/s for resistance. The identity of the genes for resistance has been established from relevant crosses with single gene lines carrying known genes for resistance.A single dominant gene effective to race 15C in E5008 has been demonstrated to be Sr9b. Of the two recessive genes, each producing distinct infection types (0; and 1–3) against race 122, one gene has been inferred to be Sr12 and the second to be a hitherto undesignated gene.The resistance of E6160 against race 15C is controlled by two genes, one dominant and one recessive. The dominant gene has been identified as Sr9b. The recessive gene has been inferred to be a new gene. Similarly, a dominant gene effective against race 122 in E6160 has been observed to be different from those so far designated. In addition, the presence of modifier gene/s in the variety, E6160 has been suggested.  相似文献

Leaf development in the normal (lobed margin) and the solanifolia (sf/sf) mutant (entire margin) of tomato (Lycopersicon esculentum) was compared at the light and scanning electron microscope levels. The shoot apices of the mutant plants contained microbodies near the axil of the youngest leaf, which were absent in the normal plants. The structural and morphological events in the initiation of leaf primordia were similar in the two genotypes. The pattern of leaflet emergence was also similar in the two types of plants, but the timing of leaflet production was different. The first pair of leaflet primordia in the normal plants was produced on P3, whereas in the mutant it was not produced until P5. The adult leaves of sf/sf plants were larger than those of normal, and the greater leaf area in the mutant was associated with a greater adaxial epidermal cell and areole area. A continuous marginal fimbriate vein (MFV) was present along the margin of each of the normal leaflets. However, a continuous MFV was absent in the mutant leaflets. It is suggested that the absence of a continuous MFV in the mutant might alter the nutritional and hormonal supply to the leaf margin, which ultimately leads to a modified leaf, i.e., with an entire margin.  相似文献
