We report the nucleotide sequence of a cloned cDNA, pMTS-3, that contains a
1-kb insert corresponding to mouse thymidylate synthase (E.C. 2.1.1.45).
The open reading frame of 921 nucleotides from the first AUG to the
termination codon specifies a protein with a molecular mass of 34,962
daltons. The predicted amino acid sequence is 90% identical with that of
the human enzyme. The mouse sequence also has an extremely high degree of
similarity (as much as 55% identity) with prokaryotic thymidylate synthase
sequences, indicating that thymidylate synthase is among the most highly
conserved proteins studied to date. The similarity is especially pronounced
(as much as 80% identity) in the 44-amino-acid region encompassing the
binding site for deoxyuridylic acid. The cDNA sequence also suggests that
mouse thymidylate synthase mRNA lacks a 3' untranslated region, since the
termination codon, UAA, is followed immediately by a poly(A) segment.

Blonder et al. ( 2014 , Global Ecology and Biogeography, 23, 595–609) introduced a new multivariate kernel density estimation (KDE) method to infer Hutchinsonian hypervolumes in the modelling of ecological niches. The authors argued that their KDE method matches or outperforms several methods for estimating hypervolume geometries and for conducting species distribution modelling. Further clarification, however, is appropriate with respect to the assumptions and limitations of KDE as a method for species distribution modelling. Using virtual species and controlled environmental scenarios, we show that KDE both under‐ and overestimates niche volumes depending on the dimensionality of the dataset and the number of occurrence records considered. We suggest that KDE may be a viable approach when dealing with large sample sizes, limited sampling bias and only a few environmental dimensions. 相似文献

Background
A nationwide survey on the microbial etiology of cases of subclinical mastitis in dairy cows was carried out on dairy farms in Sweden. The aim was to investigate the microbial panorama and the occurrence of antimicrobial resistance. Moreover, differences between newly infected cows and chronically infected cows were investigated.Methods
In total, 583 quarter milk samples were collected from 583 dairy cows at 226 dairy farms from February 2008 to February 2009. The quarter milk samples were bacteriological investigated and scored using the California Mastitis Test. Staphylococci were tested for betalactamase production and presence of resistance was evaluated in all specific udder pathogens. Differences between newly infected cows and chronically infected cows were statistically investigated using logistic regression analysis.Results
The most common isolates of 590 bacteriological diagnoses were Staphylococcus (S) aureus (19%) and coagulase-negative staphylococci (CNS; 16%) followed by Streptococcus (Str) dysgalactiae (9%), Str. uberis (8%), Escherichia (E.) coli (2.9%), and Streptococcus spp. (1.9%). Samples with no growth or contamination constituted 22% and 18% of the diagnoses, respectively. The distribution of the most commonly isolated bacteria considering only bacteriological positive samples were: S. aureus - 31%, CNS - 27%, Str. dysgalactiae - 15%, Str. uberis - 14%, E. coli - 4.8%, and Streptococcus spp. - 3.1%. There was an increased risk of finding S. aureus, Str. uberis or Str. dysgalactiae in milk samples from chronically infected cows compared to findings in milk samples from newly infected cows. Four percent of the S. aureus isolates and 35% of the CNS isolates were resistant to penicillin G. Overall, resistance to other antimicrobials than penicillin G was uncommon.Conclusions
Staphylococcus aureus and CNS were the most frequently isolated pathogens and resistance to antimicrobials was rare. 相似文献37.

This study reports on the capability of the desert plant Chilopsis linearis (Cav.) Sweet (desert willow) to uptake gold (Au) from gold-enriched media at different plant-growth stages. Plants were exposed to 20, 40, 80, 160, and 320 mg Au L(-1) in agar-based growing media for 13, 18, 23, and 35 d. The Au content and oxidation state of Au in the plants were determined using an inductively coupled plasma/optical emission spectrometer (ICP/OES) and X-ray absorption spectroscopy (XAS), respectively. Gold concentrations ranging from 20 to 80 mg Au L(-1) did not significantly affect Chilopsis linearis plant growth. The concentration of gold in the plants increased as the age of the plant increased. The Au concentrations in leaves for the 20, 40, 80, and 160 mg Au L(-1) treatments were 32, 60, 62, and 179 mg Au kg(-1) dry weight mass, respectively, demonstrating the gold uptake capability of desert willow. The XAS data indicated that desert willow produced gold nanoparticles within plant tissues. Plants exposed to 160 mg Au L(-1) formed nanoparticles that averaged approximately 8, 35, and 18 A in root, stem, and leaves, respectively. Prion diseases are associated with accumulation of the amyloid form of the prion protein, but the mechanisms of toxicity are unknown. Amyloid toxicity is also associated with fungal prions. In Podospora anserina, the simultaneous presence of [Het-s] prion and its allelic protein HET-S causes cell death in a self-/nonself-discrimination process. Here, using the prion form of a fragment of HET-s ([PrD(157)(+)]), we show that [Het-s]-HET-S toxicity can be faithfully recapitulated in yeast. Overexpression of Hsp40 chaperone, Sis1, rescues this toxicity by curing cells of [PrD(157)(+)]. We find no evidence for toxic [PrD(157)(+)] conformers in the presence of HET-S. Instead, [PrD(157)(+)] appears to seed HET-S to accumulate at the cell periphery and to form aggregates distinct from visible [PrD(157)(+)] aggregates. Furthermore, HET-S mutants that cause HET-S to be sequestered into [PrD(157)(+)] prion aggregates are not toxic. The localization of HET-S at the cell periphery and its association with cell death was also observed in the native host Podospora anserina. Thus, upon interaction with [Het-s], HET-S localizes to the cell periphery, and this relocalization, rather than the formation of mixed HET-s/HET-S aggregates, is associated with toxicity. 相似文献

Plants absorb a number of elements from soil, some of which have no known biological function and some are known to be toxic at low concentrations. As plants constitute the foundation of the food chain, some concerns have been raised about the possibility of toxic concentrations of certain elements being transported from plants to higher strata of the food chain. Special attention has been given to the uptake and biotransformation mechanisms occurring in plants and its role in bioaccumulation and impact on consumers, especially human beings. While this review draws particular attention to metal accumulation in edible plants, researched studies of certain wild plants and their consumers are included. Furthermore, this review focuses on plant uptake of the toxic elements arsenic, cadmium, chromium, mercury, and lead and their possible transfer to the food chain. These elements were selected because they are well-established as being toxic for living systems and their effects in humans have been widely documented. Arsenic is known to promote cancer of the bladder, lung, and skin and can be acquired, for example, through the consumption of As-contaminated rice. Cadmium can attack kidney, liver, bone, and it also affects the female reproduction system. Cadmium also can be found in rice. Chromium can produce cancer, and humans can be exposed through smoking and eating Cr-laden vegetables. With no further intervention, relapse rates in detoxified alcoholics are high and usually exceed 80% of all detoxified patients. It has been suggested that stress and exposure to priming doses of alcohol and to alcohol-associated stimuli (cues) contribute to the relapse risk after detoxification. This article focuses on neuronal correlates of cue responses in detoxified alcoholics. Current brain imaging studies indicate that dysfunction of dopaminergic, glutamatergic and opioidergic neurotransmission in the brain reward system (ventral striatum including the nucleus accumbens) can be associated with alcohol craving and functional brain activation in neuronal systems that process attentional relevant stimuli, reward expectancy and experience. Increased functional brain activation elicited by such alcohol-associated cues predicted an increased relapse risk, whereas high brain activity elicited by affectively positive stimuli may represent a protective factor and was correlated with a decreased prospective relapse risk. These findings are discussed with respect to psychotherapeutic and pharmacological treatment options. 相似文献
