Investigators have reported improved endurance performance and attenuated post-exercise muscle damage with carbohydrate-protein beverages (CHO+P) versus carbohydrate-only beverages (CHO). However, these benefits have been demonstrated only when CHO+P was administered in beverage-form, and exclusively in male subjects. Thus, the purposes of this study were to determine if an oral CHO+P gel improved endurance performance and post-exercise muscle damage compared to a CHO gel, and determine if responses were similar between genders. Thirteen cyclists (8 men, 5 women; VO(2)peak = 57.9 +/- 7.0 ml x kg(-1) x min(-1)) completed two timed cycle-trials to volitional exhaustion at 75% of VO(2)peak. At 15-minute intervals throughout these rides, subjects received CHO or CHO+P gels, which were matched for carbohydrate content (CHO = 0.15 g CHO x kg BW(-1); CHO+P = 0.15 g CHO + 0.038 g protein x kg BW(-1)). Trials were performed using a randomly counterbalanced, double-blind design. Subjects rode 13% longer (p < 0.05) when utilizing the CHO+P gel (116.6 +/- 28.5 minutes) versus the CHO gel (102.8 +/- 25.0 minutes). In addition, men (101.8 +/- 24.6; 114.8 +/- 26.2) and women (104.4 +/- 28.6; 119.6 +/- 34.9) responded similarly to the CHO and CHO+P trials, with no significant treatment-by-gender effect. Postexercise creatine kinease (CK) was not significantly different between treatments. However, CK increased significantly following exercise in the CHO trial (183 +/- 116; 267 +/- 214 U x L(-1)), but not the CHO+P trial (180 +/- 133; 222 +/- 141 U x L(-1)). Therefore, to prolong endurance performance and prevent increases in muscle damage, it is recommended that male and female cyclists consume CHO+P gels rather than CHO gels during and immediately following exercise. 相似文献

2004年在海拔1916m昆明两种冷害(水温19.5±0.7℃,低田温冷泉水温17.8±1.1℃)、阿子营冷害(海拔2150m,水温18.2±0.22℃)条件下对5个亲本及其25个近等基因系进行耐冷性鉴定,用Statistica对17个农艺性状进行形态聚类和SSR分子标记聚类分析。结果表明:(1)已培育的穗期耐冷性NILs与轮回亲本十和田的形态极为相似,但与耐冷性相关的性状(穗颈长、实粒数、结实率、花药长和花药体积)有明显的差异;(2)从78个SSR标记筛选出了7个标记在十和田和NILs间存在多态性, 其中RM7452标记与耐冷基因连锁,各个近等基因系间遗传背景相似,但与十和田耐冷性差异大,证明了这些NILs是水稻穗期耐冷基因精细定位和克隆的理想材料。 相似文献

We develop an approximate maximum likelihood method to estimate flanking nucleotide context-dependent mutation rates and amino acid exchange-dependent selection in orthologous protein-coding sequences and use it to analyze genome-wide coding sequence alignments from mammals and yeast. Allowing context-dependent mutation provides a better fit to coding sequence data than simpler (context-independent or CpG "hotspot") models and significantly affects selection parameter estimates. Allowing asymmetric (nonreciprocal) selection on amino acid exchanges gives a better fit than simple dN/dS or symmetric selection models. Relative selection strength estimates from our models show good agreement with independent estimates derived from human disease-causing and engineered mutations. Selection strengths depend on local protein structure, showing expected biophysical trends in helical versus nonhelical regions and increased asymmetry on polar-hydrophobic exchanges with increased burial. The more stringent selection that has previously been observed for highly expressed proteins is primarily concentrated in buried regions, supporting the notion that such proteins are under stronger than average selection for stability. Our analyses indicate that a highly parameterized model of mutation and selection is computationally tractable and is a useful tool for exploring a variety of biological questions concerning protein and coding sequence evolution. 相似文献

Research on genetically engineered mice provides insights into the etiology, therapy, and genetic basis of human diseases. An important variable that affects the results of mouse studies is the health status of the animals. Pathogen burdens may confound observations and obscure underlying mechanisms. Mouse resource centers frequently rederive infected mouse strains. We review our experience on the use of a well-established technique, embryo transfer to rederive infected mouse strains. The following mouse pathogens were eliminated by embryo transfer: Mouse Parvovirus, Mouse Hepatitis Virus, Mouse Rotavirus, Mouse Encephalomyelitis Virus, Mouse Adenovirus, Helicobacter species, endoparasites, and ectoparasites. We rederived transgenic mouse lines, gene-targeted mouse lines, and lines with spontaneous mutations. In the majority of strains, fertilized eggs for embryo transfer were obtained by mating superovulated egg donors with males of the desired genotype. A total of 309 embryo transfers were performed to rederive 96 mouse strains. The pregnancy rate was 76%; 1996 pups were born, of which 43% carried the desired genotype. We performed 44 additional embryo transfers to rederive 15 other strains. The pregnancy rate was lower (45%) and none of the 135 pups carried the desired genotype. Although we successfully eliminated the pathogens in all transfers, we were unable to obtain pups with the desired genotype in 15 of 111 mouse lines. Multiple factors affect the efficiency of rederivation by embryo transfer. They include the response to superovulation by embryo donors, the number and age of stud males, the yield of fertilized eggs, the number of embryo transfers, and genotyping. 相似文献

Electroporation of germinating pollen grains of Nicotiana gossei (L.) Domin under a variety of conditions showed that DNA was taken up by the pollen without detrimental effects on the viability of the pollen. By optimizing both the field strength of the electroporation pulse and the DNA concentration in the electroporation medium up to 6% of the donor DNA can be taken up by the germinating pollen while maintaining a pollen viability of 90%. Field strengths as high as 9 kV/cm could be applied to germinating pollen grains without detrimental effects on viability. Southern hybridizations demonstrated that DNA encoding the marker enzyme β-glucuronidase (GUS) was incorporated into electroporated pollen. Germinating pollen, treated in this manner, is capable of producing 300–400 seeds per capsule of viable seed when applied to the stigmas of compatible flowers of N. gossei which has been emasculated 4 days earlier. 相似文献
