Capsaicin, reported to elevate hormone sensitive lipase (HSL), is also found to inhibit the Ca++ and calmodulin-dependent cAMP phosphodiesterase (PDE) activity in adipose tissue of rats, fed high fat diet. The dependence of the enzyme activity on Ca++ and calmodulin in vitro, in control rats, is shown by its substantial lowering in the presence of EGTA and inhibition by trifluoperazine (TFP) (IC50 between 10-20 microM). This enzyme activity is also inhibited by both red pepper extract (80% inhibition with 50 microliter) and capsaicin (IC50 between 0.3-1 microM) in a dose dependent manner. Capsaicin has been found to inhibit Ca++-dependent PDE activity by 60% in the test rats. Enzyme inhibition in vivo, due to capsaicin, was overcome by addition of calmodulin to the assay system. Inclusion of fluphenazine or capsaicin in assay inhibited not only the calmodulin-restored enzyme activity from test rats but also that of control rats. These results suggest a possible mechanism for the stimulation of lipolytic activity by capsaicin in vivo. 相似文献

ABSTRACTPhytases hydrolyze phytates to liberate soluble and thus readily utilizable inorganic phosphate. Although phytases are produced by various groups of microbes, yeasts being simple eukaryotes and mostly non-pathogenic with proven probiotic benefits can serve as ideal candidates for phytase research. The full potential of yeast phytases has not, however, been exploited. This review focuses attention on the present status of knowledge on the production, characterization, molecular characteristics, and cloning and over-expression of yeast phytases. Pternopetalum arunachalense M. Bhaumik & P. Satyanar. is a new species from Arunachal Pradesh in India and is described and illustrated here. This new species is similar to P. trichomanifolium (Franch.) A procedure for the purification of a stable acetyl-coenzyme A synthetase (ACS) from aerobic cells of Saccharomyces cerevisiae is presented. The steps include differential centrifugation, solubilization of the bound enzyme from the crude mitochondrial fraction, ammonium sulfate fractionation, crystallization to constant specific activity from ammonium sulfate solutions followed by Bio-Gel A-1.5 m column chromatography. The resulting enzyme preparation is homogeneous as judged by chromatography on Bio-Gel columns, QAE-Sephadex A-50 anion exchange columns, analytical ultracentrifugal studies, and polyacrylamide gel electrophoresis.Sedimentation velocity runs revealed a single symmetric peak with an s20,w value of 10.6. The molecular weight of the native enzyme, as determined by gel filtration and analytical ultracentrifugation, is 250,000 ± 500. In polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulfate, the molecular weight of the single polypeptide chain is 83,000 ± 500. The purified enzyme is inhibited by palmityl-coenzyme A with a Hill interaction coefficient, n, of 2.88. These studies indicate that the ACS of aerobic S. cerevisiae is composed of three subunits of identical or nearly identical size. 相似文献
