Aging-Associated Dysfunction of Akt/Protein Kinase B: S-Nitrosylation and Acetaminophen Intervention

Background
Aged skeletal muscle is characterized by an increased incidence of metabolic and functional disorders, which if allowed to proceed unchecked can lead to increased morbidity and mortality. The mechanism(s) underlying the development of these disorders in aging skeletal muscle are not well understood. Protein kinase B (Akt/PKB) is an important regulator of cellular metabolism and survival, but it is unclear if aged muscle exhibits alterations in Akt function. Here we report a novel dysfunction of Akt in aging muscle, which may relate to S-nitrosylation and can be prevented by acetaminophen intervention.Principal Findings
Compared to 6- and 27-month rats, the phosphorylation of Akt (Ser473 and Thr308) was higher in soleus muscles of very aged rats (33-months). Paradoxically, these increases in Akt phosphorylation were associated with diminished mammalian target of rapamycin (mTOR) phosphorylation, along with decreased levels of insulin receptor beta (IR-β), phosphoinositide 3-kinase (PI3K), phosphatase and tensin homolog deleted on chromosome 10 (PTEN) and phosphorylation of phosphoinositide-dependent kinase-1 (PDK1) (Ser241). In vitro Akt kinase measurements and ex vivo muscle incubation experiments demonstrated age-related impairments of Akt kinase activity, which were associated with increases in Akt S-nitrosylation and inducible nitric oxide synthase (iNOS). Impairments in Akt function occurred parallel to increases in myocyte apoptosis and decreases in myocyte size and the expression of myosin and actin. These age-related disorders were attenuated by treating aged (27-month) animals with acetaminophen (30 mg/kg body weight/day) for 6-months.Conclusions
These data demonstrate that Akt dysfunction and increased S-nitrosylation of Akt may contribute to age-associated disorders in skeletal muscle and that acetaminophen may be efficacious for the treatment of age-related muscle dysfunction. 相似文献272.

Although phytases are produced by various groups of microbes, yeasts being simple eukaryotes and mostly non-pathogenic with proven probiotic benefits can serve as ideal candidates for phytase research. This review focuses attention on the present status of knowledge on the production, characterization, molecular characteristics, and cloning and over-expression of yeast phytases. Vitamin C, Vitamin E, scopoletin and damnacanthal are the major constituents of Noni (Morinda citrifolia). These compounds areknown to have good medicinal properties and they are known to act as antioxidants. Loss of vision in elderly is due to opaquenessof the lens proteins such as gamma-D-crystallin during oxidative stress conditions. Therefore, it is of importance to find thepotential interaction of Vitamin C, Vitamin E, Scopoletin and Damnacanthal with the lens protein gamma-D-crystallin. Hence, theirphysical binding to gamma-D crystallin (PDB ID: 2G98) was evaluated using molecular and structural docking procedures. Resultsshow the potential binding of all the above anti-oxidants to gamma-D-crystalline with equal affinity. The total lipid content of Acremonium alabamensis and Thermomucor indicae-seudaticae ranged 2.6–7.3 and 8.5–13.0% of dry mycelium, respectively during development. Neutral lipid fraction increased during growth while polar and phospholipids declined. Both moulds contained palmitic, oleic, linoleic and palmitoleic acids as major fatty acid components in lipids. Degree of unsaturation of lipids of A. alabamensis was greater than that of T. indicae-seudaticae. Neutral lipids were more unsaturated than the polar lipids. The ratio of unsaturation index of polar lipids to neutral lipids was either one or less than one. The principal phospholipids of these moulds were phosphatidyl choline, phosphatidyl ethanolamine and phosphatidic acid. The changes in the pattern of production and detoxification of ammonia have been studied in the skeletal muscles and blood of rats of different age groups (1, 3, 6, 12 and 24 months), subjected to exhaustive exercise. The protein profiles at exhaustion showed a sharp drop in all muscles and the decrement was more in the senile rats. In general, the muscle and blood ammonia content increased with age with a corresponding increase in AMP deaminase activity implicating the possibility of elevated purine nucleotide deamination during senescence. However, glutamate oxidation was decreased and urea and glutamine formation was increased consequent to ammonia production during senescence under intensive physical stress. The possible alterations in protein levels and ammonia production and its disposal in different skeletal muscle types of senile exhausted rats have been discussed in relation to detoxication capacity of the fibre types. 相似文献

A new metabolite of vitamin D3 has been isolated in pure form from incubations of rat kidney homogenates with 25-hydroxyvitamin D3 [25-OH-D3]. It was identified as 23,25-dihydroxy-24-oxo-vitamin D3 [23,25(OH)2-24-oxo-D3] by means of ultraviolet absorption spectrophotometry and mass spectrometry. Also, 25-OH-D3-26,23-lactone and 24R,25-dihydroxyvitamin D3 were obtained from the same incubation mixtures. The three enzymatic steps in the conversion of alpha-ketoisovalerate to alpha-ketoisocaproate were examined in wild-type and in leucine auxotrophic stocks of yeast. Procedures for the reliable assay of each of the enzymatic steps in crude extracts were devised. Crude extracts of the prototrophic haploid stock catalyzed all three enzymatic steps. Examination of a series of leucine auxotrophs permitted a correlation between the three enzymatic steps and the genetic lesions affecting 10 different loci. This examination revealed that a single locus (le-6) affected primarily alpha-isopropylmalate synthetase, the first step in the pathway. Lesions in six loci (le-1, le-4, le-5, le-7, le-8, and le-10) lead primarily to a deficiency in the activity of the second enzyme in the pathway, alpha-isopropylmalate isomerase. Stocks with lesions in three loci (le-2, le-3, and le-9) were primarily blocked in the third step of the pathway, catalyzed by beta-isopropylmalate dehydrogenase. The results with the mutants provide strong evidence that the pathway for leucine biosynthesis proposed by Strassman and his colleagues is the sole significant pathway in yeast. 相似文献

Summary Cell-bound phytase of Pichia anomala was produced in glucose–beef extract medium in shake flasks and in a laboratory fermenter at 25 °C for 24 h at 250 rev/min.
In the fermenter the biomass production increased and the fermentation time was reduced from 24 to 16 h. Two-week-old broiler
chicks were fed with the biomass-supplemented feed [at 100 g/7.5 kg; 50-phytase units/bird/day]. The overall weight gain in
the biomass-fed chicks was higher (90.2%) than that of the control group (77.7%). The biomass incorporation in the feed of
broiler chicks also resulted in a better phosphorus retention (29% in the control, and 73.68% in the biomass-fed) in the body,
consequently an improved growth. There was a decrease in the excretion of phosphorus in the faeces of the chicks fed with
phytase-supplemented diet (188.9 mg/g dry matter) as compared to the chicks fed on unsupplemented broiler finisher ration
(509.4 mg/g dry matter). This eliminated the need to supplement phosphorus in their diet and also reduced phosphorus pollution.
The feed conversion ratio was also lowered for chicks, which were biomass-fed as compared to the control. 相似文献

Two new bastadin alkaloids, bastadins-22 (1) and 23 (2), together with six known bastadins, bastadins-6 (3), -12 (4) (formerly basatadin-9), -14 (5), -15 (6), -16 (7), -19 (8), and a common steroid cholesterol were isolated from the sponge Dendrilla cactos. The structures of the isolates were established by the study of extensive spectroscopic analysis (1D and 2D NMR and MS data).
