The reaction of Cu(II) or Cd(II) salts with 2,4,6-iPr3C6H2PO3H2, 2,4,6-iPr3C6H2CH2PO3H2 or 2,6-iPr2C6H3OPO3H2 in the presence of strong chelating nitrogen ligands such as 2,2′-bipyridine (bpy), 1,10-phenanthroline (phen), 2-pyridylpyrazole (pypz) or 3,5-dimethyl pyrazole (dmpz) as the ancillary ligands afforded dinuclear copper or cadmium complexes [Cu2(2,4,6-iPr3C6H2PO3H)4(bpy)2] (4), [Cu2(2,6-iPr2C6H3OPO3H)2(bpy)2(OAc)2(CH3OH)2]·(CH3OH) (5), [Cd2(2,6-iPr2C6H3OPO3H)4 (bpy)2(CH3OH)2]·2(CH3OH) (6), [Cd2(2,6-iPr2C6H3OPO3H)4(phen)2] (7), [Cu2(2,6-iPr2C6H3OPO3H)2(PyPz)2(CH3OH)2] (8) and [Cu2(2,4,6-iPr3C6H2CH2PO3H)2(DMPz)2Cl2]·(CH3OH) (9) The molecular structures of 4-7 are grossly similar. The common structural features in these complexes are that the two metal centers are bridged by two bidentate [RPO2(OH)] ligands generating a central eight-membered ring. Each of the metal centers also contains a chelating nitrogen ligand and a monodentate phosphonate or a phosphate ligand. In 5 and 6 other terminal ancillary ligands are also present. In compound 8, each of the two copper centers contains a monodentate [RPO2(OH)] ligand along with a molecule of methanol. The two coppers are bridged by two monoanionic pyridylpyrazole ligands. The molecular structure of 9 is similar to that of 4-7. However, in 9 each of the two copper centers contain only terminal monodentate ligands in the form of two chlorides and a pyrazole. Magnetic studies on all of these copper complexes reveal an anti-ferromagnetic behavior at low temperatures. In addition, these complexes were found to be artificial nucleases and can convert supercoiled pBR322 DNA form I into nick form II in 1 min in the presence of an external oxidant through a hydrolytic and/or an oxidative pathway.  相似文献

Peptides were synthesized with very high purity and yield on a highly solvating copolymer of 1,4-butanediol dimethacrylate cross-linked polystyrene support (PS-BDODMA). The polymer was synthesized by radical aqueous suspension polymerization using toluene as the diluent and 1% polyvinyl alcohol as the suspension stabilizer. The supports were highly resistant to various chemical reagents and solvents that are frequently used in polypeptide synthesis. The peptides synthesized on this support were designed from the core (C), envelope (E1 and E2) and nonstructural protein (NS1-NS5) regions of the prototype hepatitis C viral (HCV) polyprotein, and were used to develop a peptide-based immunoassay (PBEIA) for the detection of HCV antibodies. The purity of these peptides was tested by HPLC. Peptide identity was confirmed by amino acid analysis and MALDI-TOF-MS. A single peptide chosen from a conserved area (E2/NS1) at the C-terminus of the hypervariable region (HVRI) was found to be sufficient for effective and reliable diagnosis of HCV infection in infected individuals, as well as also apparently healthy individuals. The CD spectrum of the peptide shows no preference for any ordered secondary structure. When used, peptide mixtures from various protein regions of HCV reduced the sensitivity and reliability of the diagnosis partly because of epitope masking.  相似文献

A novel actinobacterium, strain SK68, was isolated from the rhizosphere of peanut plant and its salinity stress alleviation ability was studied using tomato (Solanum lycopersicum cv. Micro-Tom) plants. Based on 16S rDNA based phylogenetic analysis, strain SK68 has been identified as a Streptomyces sp. Strain SK68 had branched substrate mycelium bearing smooth surfaced spores and the spore colour is brownish grey on ISP4 medium. It exhibited enzyme activities such as xylanase, cellulase, amylase, and pectinase and degraded hypoxanthine, casein, and L-tyrosine. The strain SK68 differed in its banding pattern in BOX-PCR and RAPD fingerprinting compared to the closely matching type strains Streptomyces erythrochromogenes NBRC 3304T (AB184746), S. flavotricini NBRC 12770T (AB184132), S. racemochromogenes NBRC 12906T (AB184235), and S. polychromogenes NBRC 13072T (NR041109). Strain SK68 was evaluated for its salinity stress-alleviating activity in tomato plants with 180 mmol/L NaCl under gnotobiotic condition. A significant increase in plant biomass was observed in strain SK68-inoculated tomato plants under salt stress compared to control and salt-stressed non-inoculated plants.  相似文献

Retro-ACP (74-65) fragment was synthesized on a novel 4% tri-(propylene glycol) glycerolate diacrylate cross-linked polystyrene (PS-TRPGGDA) support. The peptide is grown from the functional site present in the cross-linker, which makes it unique and cost-effective among other styrene based polymer supports. A comparative study with Merrifield resin indicates high yield and purity of the peptide synthesized on the novel support.  相似文献

Bivalve Condition Index (CI) is widely employed in environmental monitoring programmes as it integrates physiological responses to stress with changes in somatic growth. Besides indicating, the commercial quality of a bivalve population it may be also be used to compare the relative health of animals in nearby populations. In this study, CI of green mussels, Perna viridis was evaluated as the intrinsic response to the variations in the environment in two potential mussel mariculture sites in the tropical monsoon region with diverse remoteness to riverine outflow. Condition index of mussels from site with relatively higher riverine influence (RI) was compared with mussels from area of reduced riverine (RR) influence along Karnataka coast (Eastern Arabian Sea). The dominant patterns of spatial trends in 12 environmental variables (temperature, salinity, pH, dissolved oxygen, rainfall, chlorophyll-a (chl-a), suspended particulate matter (SPM) particulate organic matter (POM), particulate inorganic matter (PIM), POM/SPM, chla/POM and PIM%) of the sites were resolved by Principal Component Analysis (PCA). The stepwise multiple regression analysis related the spatial variability in CI to variations in water temperature and organic content of the seston (chlorophyll-a and particulate organic matter). The discriminant analysis performed with monthly mean water temperature, chl-a levels and CI ratio [high CI ratio (CIhigh) and low CI ratio (CIlow)] indicated that 83.9% of the CIhigh was associated with high chl-a and high water temperature group whereas, 72.4% of CIlow were associated with low chl-a and low water temperature regime. The RR site presented less variation in environmental parameters offering a more conducive environment for the growth of mussels, characterised by better CI with low seasonal variations. Whereas, in the site proximate to riverine discharge, reserves were alternatively channelled into energy-consuming processes and hence unfavourable environmental conditions showed poor tissue condition due to utilization of energy reserves which were potentially destined for growth. Thus, average CI, besides its representation of the general health condition of the bivalves can potentially be used in selecting suitable sites for bivalve mariculture in comparable hydrological environment.  相似文献

Significant recovery after treatment with the whole plant slurry of A.longifolia Nees. was observed in plasma AST, ALT and cholesterol levels in CCl4 induced hepatotoxic rats. This was amply supported by electron micrographs, which indicated normalization of cytoarchitecture of mitochondria and endoplasmic reticulum. The results suggest that the slurry of the plant is useful as a liver tonic.  相似文献

Mutations in the adenomatous polyposis coli (APC) gene are found in most colorectal cancers. They cause constitutive activation of proliferative pathways when both alleles of the gene are mutated. However studies on individuals with familial adenomatous polyposis (FAP) have shown that a single mutated APC allele can also create changes in the precancerous colon crypt, like increased number of stem cells, increased crypt fission, greater variability of DNA methylation patterns, and higher somatic mutation rates. In this paper, using a computational model of colon crypt dynamics, we evolve and investigate a hypothesis on the effect of heterozygous APC mutation that explains these different observations. Based on previous reports and the results from the computational model we propose the hypothesis that heterozygous APC mutation has the effect of increasing the chances for a stem cell to divide symmetrically, producing two stem cell daughters. We incorporate this hypothesis into the model and perform simulation experiments to investigate the consequences of the hypothesis. Simulations show that this hypothesis links together the changes in FAP crypts observed in previous studies. The simulations also show that an APC(+/-) stem cell gets selective advantages for dominating the crypt and progressing to cancer. This explains why most colon cancers are initiated by APC mutation. The results could have implications for preventing or retarding the onset of colon cancer in people with inherited or acquired mutation of one APC allele. Experimental validation of the hypothesis as well as investigation into the molecular mechanisms of this effect may therefore be worth undertaking.  相似文献

Pouteria campechiana (Kunth) Baehni. is used as a remedy for coronary trouble, liver disorders, epilepsy, skin disease, and ulcer. Therefore, the present study aims to investigate the antioxidant and hepatoprotective effect of polyphenolic-rich P. campechiana fruit extract against acetaminophen-intoxicated rats. Total phenolic and flavonoid contents of egg fruit were estimated followed by the determination of antioxidant activities. Treatment with P. campechiana fruit extract effectively scavenged the free radicals in a concentration-dependent manner within the range of the given concentrations in all antioxidant models. The presence of polyphenolic compounds were confirmed by high-performance thin-layer chromatography (HPTLC). The animals were treated with acetaminophen (250 mg/kg body weight; p.o.) thrice at the interval of every 5 days after the administration of P. campechiana aqueous extract and silymarin (50 mg/kg). Acetaminophen treatment was found to trigger an oxidative stress in liver, leading to an increase of serum marker enzymes. However, treatment with P. campechiana fruit extract significantly reduced the elevated liver marker enzymes (aspartate transaminase, alanine transaminase, and alkaline phosphatase) and increased the antioxidant enzymes (viz., superoxide dismutase and catalase) and glutathione indicating the effect of the extract in restoring the normal functional ability of hepatocytes. These results strongly suggest that P. campechiana fruit extract has strong antioxidant and significant hepatoprotective effect against acetaminophen-induced hepatotoxicity.  相似文献

The aim of this study was to enhance the viability of probiotic strains Pediococcus pentosaceus KID7, Lactobacillus plantarum KII2, Lactobacillus fermentum KLAB6 and Lactobacillus helveticus KII13 in gastrointestinal transit, freeze-drying condition and during storage time by microencapsulation using a combination of alginate, fenugreek gum and locust bean gum. The microcapsules were prepared using various ratio of alginate to fenugreek gum to locust bean gum and tested for its dissolution in colonic fluid. The combination that efficiently dissolved in colonic fluid was selected for co-encapsulation of the probiotic strains and prebiotics to produce synbiotic microcapsules. Further, we observed that the bacteria encapsulated with alginate-fenugreek gum-locust bean gum (AFL) matrix tolerated gastrointestinal condition efficiently compared to non-encapsulated bacteria. The encapsulated bacterial cells retained higher viability than non-encapsulated cells during freeze-drying condition and subsequent storage for 3 months at 4°C. These results show the utility of AFL matrix in microencapsulation of probiotics for use in food industry.  相似文献

The big-fin reef squid, Sepioteuthis cf. lessoniana (Lesson 1930), is an important commodity species within artisanal and near-shore fisheries in the Indian and Indo-Pacific regions. While there has been some genetic and physical evidence that supports the existence of a species complex within S. cf. lessoniana, these studies have been extremely limited in scope geographically. To clarify the extent of cryptic diversity within S. cf. lessoniana, this study examines phylogenetic relationships using mitochondrial genes (cytochrome oxidase c, 16s ribosomal RNA) and nuclear genes (rhodopsin, octopine dehydrogenase) from nearly 400 individuals sampled from throughout the Indian, Indo-Pacific, and Pacific Ocean portions of the range of this species. Phylogenetic analyses using maximum likelihood methods and Bayesian inference identified three distinct lineages with no clear geographic delineations or morphological discriminations. Phylogeographic structure analysis showed high levels of genetic connectivity in the most widespread lineage, lineage C and low levels of connectivity in lineage B. This study provides significant phylogenetic evidence for cryptic lineages within this complex and confirms that cryptic lineages of S. cf. lessoniana occur in sympatry at both small and large spatial scales. Furthermore, it suggests that two closely related co-occurring cryptic lineages have pronounced differences in population structure, implying that underlying differences in ecology and/or life history may facilitate co-occurrence. Further studies are needed to assess the range and extent of cryptic speciation throughout the distribution of this complex. This information is extremely useful as a starting point for future studies exploring the evolution of diversity within Sepioteuthis and can be used to guide fisheries management efforts.  相似文献
